    Risk assessment in non-standard forms of civil engineering consulting services

    Although a large body of research exists on risk assessment in civil engineering projects and of owners, contractors, concessionaires and financiers of such projects, there is a lacuna in such research on engineering consultants, particularly those associated with non-standard forms of consulting services. This paper seeks to explore the genesis of the underlying risks in non-standard forms of engineering consulting services, systematically classify the risks, and develop a Risk Breakdown Structure and a generic Framework for efficient assessment of these risks, which is a prerequisite for sound risk management in the engineering consulting industry. The research adopts a mixed method approach, synthesising exploratory type multiple-case studies and questionnaire surveys, carried out in 14 engineering consulting firms having extensive experience in the delivery of non-standard consulting services. This paper provides empirical insights of the genetic makeup of risks associated with non-standard forms of consulting services. Such risks are found to be predominantly linked to design office based activities that underline the importance of design function in engineering consulting practice. Loss of reputation and/or goodwill is rated as the most severe potential impact on consultants. Proposed Risk Assessment Framework provides the engineering consulting industry with a functional tool for efficient risk management

    A simple household level natural water purification system to purify water to meat small family requirement. It disused with six important components including Storage, Sedimentation, Biological treatment, and Filtering, Disinfection and Drinking water storage. During the treatment process Electrical conductivity and Total Dissolved Solids values is not changed but pH is slightly increased in the bio treatment compartment due to excretion of ammonia form by the fish. A Significant change of Coli form bacterial count between UV treatment together with sand and Charcoal filter with comparing drinking water tank It is 90.95% reduction comparing with raw water E coli count is completely removed from this treatment process

    Urban vertical luxury living is becoming particularly widespread, and the abundance of luxury apartment buildings in Colombo continues to transform the city. The main objective of the study is to identify the factors that significantly influence the purchasing decision of customers in respect of luxury apartments with reference to Colombo and suburbs and further investigation of the level of importance of the attributes of identified factors on the customers’ point of view. Through a comprehensive literature review, 08 main factors and 53 attributes were identified to examine the customers’ purchasing behaviour in luxury apartments. The study has revealed that the most influencing factor is Financial feasibility of the consumer while the Physical factors and Locational importance are equally significance. Basic Amenities, Environment, Goodwill, Emotional and Recreation have ranked accordingly. The 53 attribites were deeply exposed the contrast of the customers’ consideration and preference. In addition to the financial attributes, dwelling type, dedicated parking units, and floor area of the apartment, electricity backup, security, greenery spaces, and developer have shown the more than 80% of customers’ attention and predilection as the most important attributes which influenced to customers’ purchasing dececision on luxury apartments

    Characterization of sex-related differences in allergen house dust mite-challenged airway inflammation, in two different strains of mice

    Abstract Biological sex impacts disease prevalence, severity and response to therapy in asthma, however preclinical studies often use only one sex in murine models. Here, we detail sex-related differences in immune responses using a house dust mite (HDM)-challenge model of acute airway inflammation, in adult mice of two different strains (BALB/c and C57BL/6NJ). Female and male mice were challenged (intranasally) with HDM extract (~ 25 μg) for 2 weeks (N = 10 per group). Increase in serum HDM-specific IgE showed a female bias, which was statistically significant in BALB/c mice. We compared naïve and HDM-challenged mice to define immune responses in the lungs by assessing leukocyte accumulation in the bronchoalveolar lavage fluid (BALF), and profiling the abundance of 29 different cytokines in BALF and lung tissue lysates. Our results demonstrate specific sex-related and strain-dependent differences in airway inflammation. For example, HDM-driven accumulation of neutrophils, eosinophils and macrophages were significantly higher in females compared to males, in BALB/c mice. In contrast, HDM-mediated eosinophil accumulation was higher in males compared to females, in C57BL/6NJ mice. Differences in lung cytokine profiles indicated that HDM drives a T-helper (Th)17-biased response with higher IL-17 levels in female BALB/c mice compared to males, whereas female C57BL/6NJ mice elicit a mixed Th1/Th2-skewed response. Male mice of both strains showed higher levels of specific Th2-skewed cytokines, such as IL-21, IL-25 and IL-9, in response to HDM. Overall, this study details sex dimorphism in HDM-mediated airway inflammation in mice, which will be a valuable resource for preclinical studies in allergic airway inflammation and asthma

    A Pilot Study Examining the Effects of Music Training on Attention in Children with Fetal Alcohol Spectrum Disorders (FASD)

    Prior studies indicate differences in brain volume and neurophysiological responses of musicians relative to non-musicians. These differences are observed in the sensory, motor, parietal, and frontal cortex. Children with a fetal alcohol spectrum disorder (FASD) experience deficits in auditory, motor, and executive function domains. Therefore, we hypothesized that short-term music training in children with an FASD due to prenatal alcohol exposure may improve brain function. Children (N = 20) with an FASD were randomized to participate in either five weeks of piano training or to a control group. Selective attention was evaluated approximately seven weeks apart (pre-/post-music training or control intervention), examining longitudinal effects using the Attention Networks Test (ANT), a well-established paradigm designed to evaluate attention and inhibitory control, while recording EEG. There was a significant group by pre-/post-intervention interaction for the P250 ms peak of the event-related potential and for theta (4–7 Hz) power in the 100–300 ms time window in response to the congruent condition when the flanking stimuli were oriented congruently with the central target stimulus in fronto-central midline channels from Cz to Fz. A trend for improved reaction time at the second assessment was observed for the music trained group only. These results support the hypothesis that music training changes the neural indices of attention as assessed by the ANT in children with an FASD. This study should be extended to evaluate the effects of music training relative to a more closely matched active control and determine whether additional improvements emerge with longer term music training

    Combination of IL-17A/F and TNF-alpha uniquely alters the bronchial epithelial cell proteome to enhance proteins that augment neutrophil migration

    Background: The heterodimer interleukin (IL)-17A/F is elevated in the lungs in chronic respiratory disease such as severe asthma, along with the pro-inflammatory cytokine tumor necrosis factor-alpha (TNF-alpha). Although IL-17A/F and TNF-alpha are known to functionally cooperate to exacerbate airway inflammation, proteins altered by their interaction in the lungs are not fully elucidated.Results: We used Slow Off-rate Modified Aptamer-based proteomic array to identify proteins that are uniquely and/or synergistically enhanced by concurrent stimulation with IL-17A/F and TNF-alpha in human bronchial epithelial cells (HBEC). The abundance of 38 proteins was significantly enhanced by the combination of IL-17A/F and TNF-alpha, compared to either cytokine alone. Four out of seven proteins that were increased > 2-fold were those that promote neutrophil migration; host defence peptides (HDP; Lipocalin-2 (LCN-2) and Elafin) and chemokines (IL-8, GRO alpha). We independently confirmed the synergistic increase of these four proteins by western blots and ELISA. We also functionally confirmed that factors secreted by HBEC stimulated with the combination of IL-17A/F and TNF-alpha uniquely enhances neutrophil migration. We further showed that PI3K and PKC pathways selectively control IL-17A/F + TNF-alpha-mediated synergistic production of HDPs LCN-2 and Elafin, but not chemokines IL-8 and GRO alpha. Using a murine model of airway inflammation, we demonstrated enhancement of IL-17A/F, TNF-alpha, LCN-2 and neutrophil chemokine KC in the lungs, thus corroborating our findings in-vivo.Conclusion: This study identifies proteins and signaling mediated by concurrent IL-17A/F and TNF-alpha exposure in the lungs, relevant to respiratory diseases characterized by chronic inflammation, especially neutrophilic airway inflammation such as severe asthma
