    A panel of human lung carcinoma lines representing the four main histological types (squamous, small-cell, large-cell and adenocarcinoma), and derived from both primary and metastatic sites, has been established in xenograft and in tissue culture. The highest take rates were achieved when biopsy specimens were obtained from large tumour masses and cultured lines were most readily established after preliminary passages as xenografts. The established lines exhibited an overlapping spectrum of biochemical and morphological characteristics, and showed a tendency to change from one cell type to another, in keeping with the concept of a common endodermal cell of origin. Radiation resistance appeared to be related to the large-cell phenotype

    An analysis of the most distant catalogued open clusters -- Re-assessing fundamental parameters with Gaia EDR3 and ASteCA\texttt{ASteCA}

    Several studies have been presented in the last few years applying some kind of automatic processing of data to estimate the fundamental parameters of open clusters. These parameters are later on employed in larger scale analyses, for example the structure of the Galaxy's spiral arms. The distance is one of the more straightforward parameters to estimate, yet enormous differences can still be found among published data. This is particularly true for open clusters located more than a few kpc away. We cross-matched several published catalogues and selected the twenty-five most distant open clusters (>>9000 pc). We then performed a detailed analysis of their fundamental parameters, with emphasis on their distances, to determine the agreement between catalogues and our estimates.} Photometric and astrometric data from the Gaia EDR3 survey was employed. The data was processed with our own membership analysis code (pyUPMASK), and our package for automatic fundamental cluster's parameters estimation (ASteCA\texttt{ASteCA}). We find differences in the estimated distances of up to several kpc between our results and those catalogued, even for the catalogues that show the best matches with ASteCA\texttt{ASteCA} values. Large differences are also found for the age estimates. As a by-product of the analysis we find that vd Bergh-Hagen 176 could be the open cluster with the largest heliocentric distance catalogued to date. Caution is thus strongly recommended when using catalogued parameters of open clusters to infer large-scale properties of the Galaxy, particularly for those located more than a few kpc away.Comment: Accepted for publication in A&

    We show that the coherent oscillations of the e.m. field induced by the collective quantum fluctuations of the nuclear matter field associated with the giant resonances, with frequencies Ο‰A≃78Aβˆ’1/3\omega_{A}\simeq 78A^{-{1/3}} MeV, give rise to a significant (e+eβˆ’)(e^+e^-) pair production in high energy Heavy Ion collisions.     Pumilio-2 Function in the Mouse Nervous System

    Coordinated mRNA translation at the synapse is increasingly recognized as a critical mechanism for neuronal regulation. Pumilio, a translational regulator, is known to be involved in neuronal homeostasis and memory formation in Drosophila. Most recently, the mammalian Pumilio homolog Pumilio-2 (Pum2) has been found to play a role in the mammalian nervous system, in particular in regulating morphology, arborization and excitability of neuronal dendrites, in vitro. However, the role of Pum2 in vivo remains unclear. Here, we report our investigation of the functional and molecular consequences of Pum2 disruption in vivo using an array of neurophysiology, behavioral and gene expression profiling techniques. We used Pum2-deficient mice to monitor in vivo brain activity using EEG and to study behavior traits, including memory, locomotor activity and nesting capacities. Because of the suspected role of Pum2 in neuronal excitability, we also examined the susceptibility to seizure induction. Finally, we used a quantitative gene expression profiling assay to identify key molecular partners of Pum2. We found that Pum2-deficient mice have abnormal behavioral strategies in spatial and object memory test. Additionally, Pum2 deficiency is associated with increased locomotor activity and decreased body weight. We also observed environmentally-induced impairment in nesting behavior. Most importantly, Pum2-deficient mice showed spontaneous EEG abnormalities and had lower seizure thresholds using a convulsing dosage of pentylenetetrazole. Finally, some genes, including neuronal ion channels, were differentially expressed in the hippocampus of Pum2-deficient mice. These findings demonstrate that Pum2 serves key functions in the adult mammalian central nervous system encompassing neuronal excitability and behavioral response to environmental challenges

    Depletion oil recovery for systems with widely varying initial composition

    Abstract The principal depletion drive mechanism is the expansion of oil and gas initially in the reservoir-neglecting water influx. The main factors in depletion drive reservoir performance are total cumulative compressibility, determined mostly by initial composition (gas-oil ratio), saturation pressure, PVT properties, and relative permeability. In this paper, we systematically study the effect of initial composition on oil recovery, all other parameters held constant. We also evaluate other aspects of reservoir performance, but the main emphasis is surface oil recovery including condensate. To analyze the effect of initial composition, a series of fluid systems was selected by a recombination of separator samples at varying gas-oil ratios. The systems ranged from low-GOR oils to high-GOR gas condensates, with a continuous transition from gas to oil through a critical mixture. Black oil and compositional material balance calculations, 2D fine-grid, and 3D coarse-grid models have been used to investigate the effect of initial fluid composition on reservoir depletion performance. Systematic variation of relative permeabilities was also used to map the range of fluid systems, which were most sensitive to relative permeability. For reservoir oils, the depletion recovery of surface oil initially increases with increasing initial gas-oil ratio. Oil recovery reaches a maximum for moderate-GOR oil reservoirs, followed by decreasing oil recoveries with increasing initial solution GOR. A minimum oil recovery is reached at a near-critical oil. For gas reservoirs, depletion drive condensate recovery increases monotonically from a near-critical gas towards near 100% condensate recovery for veryhigh GOR systems. STO recovery from oil reservoirs depends increasingly on gas-oil relative permeabilities as initial solution GOR increases, up to a point. At higher initial solution GORs, oil recovery becomes less dependent on relative permeability and, as the fluid system transitions to a gas at the critical point, relative permeability dependence rapidly diminishes. Condensate recovery from www.elsevier.com/locate/petrol gas condensate systems is more or less independent of gas-oil relative permeabilities, with only slight dependence for nearcritical gases.

    Defining Human Embryo Phenotypes by Cohort-Specific Prognostic Factors

    Hundreds of thousands of human embryos are cultured yearly at in vitro fertilization (IVF) centers worldwide, yet the vast majority fail to develop in culture or following transfer to the uterus. However, human embryo phenotypes have not been formally defined, and current criteria for embryo transfer largely focus on characteristics of individual embryos. We hypothesized that embryo cohort-specific variables describing sibling embryos as a group may predict developmental competence as measured by IVF cycle outcomes and serve to define human embryo phenotypes.We retrieved data for all 1117 IVF cycles performed in 2005 at Stanford University Medical Center, and further analyzed clinical data from the 665 fresh IVF, non-donor cycles and their associated 4144 embryos. Thirty variables representing patient characteristics, clinical diagnoses, treatment protocol, and embryo parameters were analyzed in an unbiased manner by regression tree models, based on dichotomous pregnancy outcomes defined by positive serum beta-human chorionic gonadotropin (beta-hCG). IVF cycle outcomes were most accurately predicted at approximately 70% by four non-redundant, embryo cohort-specific variables that, remarkably, were more informative than any measures of individual, transferred embryos: Total number of embryos, number of 8-cell embryos, rate (percentage) of cleavage arrest in the cohort and day 3 follicle stimulating hormone (FSH) level. While three of these variables captured the effects of other significant variables, only the rate of cleavage arrest was independent of any known variables.Our findings support defining human embryo phenotypes by non-redundant, prognostic variables that are specific to sibling embryos in a cohort

    Viral Single-Strand DNA Induces p53-Dependent Apoptosis in Human Embryonic Stem Cells

    Human embryonic stem cells (hESCs) are primed for rapid apoptosis following mild forms of genotoxic stress. A natural form of such cellular stress occurs in response to recombinant adeno-associated virus (rAAV) single-strand DNA genomes, which exploit the host DNA damage response for replication and genome persistence. Herein, we discovered a unique DNA damage response induced by rAAV transduction specific to pluripotent hESCs. Within hours following rAAV transduction, host DNA damage signaling was elicited as measured by increased gamma-H2AX, ser15-p53 phosphorylation, and subsequent p53-dependent transcriptional activation. Nucleotide incorporation assays demonstrated that rAAV transduced cells accumulated in early S-phase followed by the induction of apoptosis. This lethal signaling sequalae required p53 in a manner independent of transcriptional induction of Puma, Bax and Bcl-2 and was not evident in cells differentiated towards a neural lineage. Consistent with a lethal DNA damage response induced upon rAAV transduction of hESCs, empty AAV protein capsids demonstrated no toxicity. In contrast, DNA microinjections demonstrated that the minimal AAV origin of replication and, in particular, a 40 nucleotide G-rich tetrad repeat sequence, was sufficient for hESC apoptosis. Our data support a model in which rAAV transduction of hESCs induces a p53-dependent lethal response that is elicited by a telomeric sequence within the AAV origin of replication

    Efficient Culturing and Genetic Manipulation of Human Pluripotent Stem Cells

    Human pluripotent stem cells (hPSC) hold great promise as models for understanding disease and as a source of cells for transplantation therapies. However, the lack of simple, robust and efficient culture methods remains a significant obstacle for realizing the utility of hPSCs. Here we describe a platform for the culture of hPSCs that 1) allows for dissociation and replating of single cells, 2) significantly increases viability and replating efficiency, 3) improves freeze/thaw viability 4) improves cloning efficiency and 5) colony size variation. When combined with standard methodologies for genetic manipulation, we found that the enhanced culture platform allowed for lentiviral transduction rates of up to 95% and electroporation efficiencies of up to 25%, with a significant increase in the total number of antibiotic-selected colonies for screening for homologous recombination. We further demonstrated the utility of the enhanced culture platform by successfully targeting the ISL1 locus. We conclude that many of the difficulties associated with culturing and genetic manipulation of hPSCs can be addressed with optimized culture conditions, and we suggest that the use of the enhanced culture platform could greatly improve the ease of handling and general utility of hPSCs
