    New U-Th Dates from Vindija, Velika pećina (Kličevica) and Mujina pećina and Their Implications for Chronology of the Middle Paleolithic in Croatia

    This paper reports new results obtained by Uranium-Thorium (U-Th) dating of animal bones, tooth and flowstone samples from three Croatian Middle Paleolithic sites. Dates were obtained on bones and teeth from Vindija (Hrvatsko zagorje) and flowstones from Velika pećina in Kličevica and Mujina pećina (both in Dalmatia). Obtained results support the previously established chronology of the Middle Paleolithic of Croatia by confirming that the oldest layers of Vindija belong to MIS 6 and that Velika pećina in Kličevica was visited by Neandertals after 40 ka BP

    Vindija Cave and The Modern Human Peopling of Europe

    Vindija cave in Croatia has yielded the youngest securely dated Neandertal skeletal remains in Central/Eastern Europe. In addition, these remains have been found in association with archaeological material exhibiting Upper Paleolithic elements. Due to its geographic location and date, the Vindija remains are particularly crucial for the understanding of initial modern human peopling of Europe and the nature of the Neandertal demise. The significance of archaeological and paleontological finds and hominin fossils from this site is discussed in the light of new finds at Vindija and recent developments in the fields of paleoanthropology and prehistoric archaeology. Furthermore, the impact of revised chronology for several crucial specimens and sites throughout Europe, including Vindija, is discussed

    Rescue of Ī±B Crystallin (HSPB5) Mutants Associated Protein Aggregation by Co-Expression of HSPB5 Partners

    HSPB5 (also called Ī±B-crystallin) is a ubiquitously expressed small heat shock protein. Mutations in HSPB5 have been found to cause cataract, but are also associated with a subgroup of myofibrillar myopathies. Cells expressing each of these HSPB5 mutants are characterized by the appearance of protein aggregates of primarily the mutant HSPB5. Like several members of the HSPB family, HSPB5 can form both homo-oligomeric and hetero-oligomeric complexes. Previous studies showed that co-expression of HSPB1 and HSPB8 can prevent the aggregation associated with the HSPB5 (R120G) mutant in cardiomyocytes and in transgenic mice. In this study, we systematically compared the effect of co-expression of each of the members of the human HSPB family (HSPB1-10) on the aggregation of three different HSPB5 mutants (R120G, 450 Ī” A, 464 Ī” CT). Of all members, co-expression of HSPB1, HSPB4 and HSPB5 itself, most effectively prevent the aggregation of these 3 HSPB5 mutants. HSPB6 and HSPB8 were also active but less, whilst the other 5 HSPB members were ineffective. Co-expression of Hsp70 did not reduce the aggregation of the HSPB5 mutants, suggesting that aggregate formation is most likely not related to a toxic gain of function of the mutants per se, but rather related to a loss of chaperone function of the oligomeric complexes containing the HSPB5 mutants (dominant negative effects). Our data suggest that the rescue of aggregation associated with the HSPB5 mutants is due to competitive incorporation of its partners into hetero-oligomers hereby negating the dominant negative effects of the mutant on the functioning of the hetero-oligomer

    Plastic collapse of general frames

    A structural theory is presented for the large static plastic deformation of space frames composed of thin walled members. Displacements comparable to the overall structural dimensions are contemplated. The frame is considered to consist of an arbitrary number of beam elements connected at node points. The analysis assumes that plastic deformation is confined to idealized hinges located at the node points. As a basis for a general frame computer program, the equations for a beam element are derived as a relationship between appropriate generalized force and deformation rates. The structural constitutive theory employed for the plastic hinge includes biaxial bending, torsion, and axial extension. It accounts for reduction in the load carrying capacity of the hinge due to local deformation.     Personalised hip therapy : development of a non-operative protocol to treat femoroacetabular impingement syndrome in the FASHIoN randomised controlled trial

    Introduction Femoroacetabular impingement (FAI) syndrome is increasingly recognised as a cause of hip pain. As part of the design of a randomised controlled trial (RCT) of arthroscopic surgery for FAI syndrome, we developed a protocol for non-operative care and evaluated its feasibility. Methods In phase one, we developed a protocol for non-operative care for FAI in the UK National Health Service (NHS), through a process of systematic review and consensus gathering. In phase two, the protocol was tested in an internal pilot RCT for protocol adherence and adverse events. Results The final protocol, called Personalised Hip Therapy (PHT), consists of four core components led by physiotherapists: detailed patient assessment, education and advice, help with pain relief and an exercise-based programme that is individualised, supervised and progressed over time. PHT is delivered over 12ā€“26ā€…weeks in 6ā€“10 physiotherapist-patient contacts, supplemented by a home exercise programme. In the pilot RCT, 42 patients were recruited and 21 randomised to PHT. Review of treatment case report forms, completed by physiotherapists, showed that 13 patients (62%) received treatment that had closely followed the PHT protocol. 13 patients reported some muscle soreness at 6ā€…weeks, but there were no serious adverse events. Conclusion PHT provides a structure for the non-operative care of FAI and offers guidance to clinicians and researchers in an evolving area with limited evidence. PHT was deliverable within the National Health Service, is safe, and now forms the comparator to arthroscopic surgery in the UK FASHIoN trial (ISRCTN64081839)
