The representation of morphologically complex words
In dieser Dissertation wurde die Repräsentation morphologisch komplexer Wörter im mentalen Lexikon untersucht. Morphologie beschäftigt sich mit der inneren Struktur von Wörtern. Die Frage, ob morphologisch komplexe Wörter als komplette gebrauchsfertige Einheiten oder zerlegt in ihre Morpheme im mentalen Lexikon gespeichert sind, wird kontrovers diskutiert (z.B. Butterworth, 1983, Caramazza, Laudanna & Romani, 1988; Schreuder & Baayen, 1995; Taft & Ardasinski, 2006). In der vorliegenden Arbeit wurden Sprachproduktions- und Sprachwahrnehmungsexperimente zu folgenden Fragen durchgeführt:
1) Ist die modifizierte Variante einer Ăśbersetzungsaufgabe geeignet, um
morphologische Prozesse bei der Sprachproduktion zu untersuchen?
2) Treten morphologische Effekte unabhängig von semantischer und phonologischer Verwandtschaft auf?
3) Greifen das Sprachproduktionssystem und das Spracherkennungssystem auf
dieselben morphologisch dekomponierten Wortform-Repräsentationen zu
Effects of Place of Articulation Changes on Auditory Neural Activity: A Magnetoencephalography Study
In casual speech, phonemic segments often assimilate such that they adopt features from adjacent segments, a typical feature being their place of articulation within the vocal tract (e.g., labial, coronal, velar). Place assimilation (e.g., from coronal /n/ to labial /m/: rainbow→*raimbow) alters the surface form of words. Listeners' ability to perceptually compensate for such changes seems to depend on the phonemic context, on whether the adjacent segment (e.g., the /b/ in “rainbow”) invites the particular change. Also, some assimilations occur frequently (e.g., /n/→/m/), others are rare (e.g., /m/→/n/). We investigated the effects of place assimilation, its contextual dependency, and its frequency on the strength of auditory evoked mismatch negativity (MMN) responses, using pseudowords. Results from magnetoencephalography (MEG) revealed that the MMN was modulated both by the frequency and contextual appropriateness of assimilations
Betriebsfeste Auslegung hochbelasteter Kunststoffbauteile
Bauteile aus kurzfaserverstärkten Kunststoffen werden aus Gründen des Leichtbaus aber auch der Kostenersparnis immer mehr im Fahrzeugbau eingesetzt, z.B. als Komponenten von Saugsystemen. Diese Bauteile müssen ihre Funktion über die Einsatzdauer ohne Ausfall bestehen. Dies erfordert eine Bauteilauslegung gegen die zyklischen Betriebsbelastungen unter Berücksichtigung nicht nur des Werkstoffs sondern auch der konstruktiven Formgebung (Spannungskonzentration) und Umgebung (Temperatur). Im Folgenden wird ein kurzer Einblick über eine Untersuchung gegeben, die in Zusammenarbeit mit der Robert Bosch GmbH Zentralabteikung Kunststofftechnik, Forschung und Vorausentwicklung im Fraunhofer-Institut für Betriebsfestigkeit LBF durchgeführt wurde. Anhand einiger ausgewählter Ergebnisse wird auf das Untersuchungsspektrum des Instituts eingegangen
Targeted siRNA nanocarrier: a platform technology for cancer treatment
The small arginine-rich protein protamine condenses complete genomic DNA into the sperm head. Here, we applied its high RNA binding capacity for spontaneous electrostatic assembly of therapeutic nanoparticles decorated with tumour-cell-specific antibodies for efficiently targeting siRNA. Fluorescence microscopy and DLS measurements of these nanocarriers revealed the formation of a vesicular architecture that requires presence of antibody-protamine, defined excess of free SMCC-protamine, and anionic siRNA to form. Only these complex nanoparticles were efficient in the treatment of non-small-cell lung cancer (NSCLC) xenograft models, when the oncogene KRAS was targeted via EGFR-mediated delivery. To show general applicability, we used the modular platform for IGF1R-positive Ewing sarcomas. Anti-IGR1R-antibodies were integrated into an antibody-protamine nanoparticle with an siRNA specifically against the oncogenic translocation product EWS/FLI1. Using these nanoparticles, EWS/FLI1 knockdown blocked in vitro and in vivo growth of Ewing sarcoma cells. We conclude that these antibody-protamine-siRNA nanocarriers provide a novel platform technology to specifically target different cell types and yet undruggable targets in cancer therapy by RNAi.ISSN:0950-9232ISSN:1476-559
