    Terminal differentiation is not a major determinant for the success of stem cell therapy - cross-talk between muscle-derived stem cells and host cells

    We have found that when muscle-derived stem cells (MDSCs) are implanted into a variety of tissues only a small fraction of the donor cells can be found within the regenerated tissues and the vast majority of cells are host derived. This observation has also been documented by other investigators using a variety of different stem cell types. It is speculated that the transplanted stem cells release factors that modulate repair indirectly by mobilizing the host's cells and attracting them to the injury site in a paracrine manner. This process is loosely called a 'paracrine mechanism', but its effects are not necessarily restricted to the injury site. In support of this speculation, it has been reported that increasing angiogenesis leads to an improvement of cardiac function, while inhibiting angiogenesis reduces the regeneration capacity of the stem cells in the injured vascularized tissues. This observation supports the finding that most of the cells that contribute to the repair process are indeed chemo-attracted to the injury site, potentially through host neo-angiogenesis. Since it has recently been observed that cells residing within the walls of blood vessels (endothelial cells and pericytes) appear to represent an origin for post-natal stem cells, it is tempting to hypothesize that the promotion of tissue repair, via neo-angiogenesis, involves these blood vessel-derived stem cells. For non-vascularized tissues, such as articular cartilage, the regenerative property of the injected stem cells still promotes a paracrine, or bystander, effect, which involves the resident cells found within the injured microenvironment, albeit not through the promotion of angiogenesis. In this paper, we review the current knowledge of post-natal stem cell therapy and demonstrate the influence that implanted stem cells have on the tissue regeneration and repair process. We argue that the terminal differentiation capacity of implanted stem cells is not the major determinant of the cells regenerative potential and that the paracrine effect imparted by the transplanted cells plays a greater role in the regeneration process

    Obesity and Related Factors among Jerash University Population

    This study aims to measure the prevalence of obesity, to assess the food habits and physical activity among university population in Jerash University, Northern Jordan. A cross-sectional survey of (203) university students and employee (147 males and 56 females) aged 20 to 60 years were selected randomly during the spring 2016 semester. Participants filled out a self-reported questionnaire (included questions on eating and physical activity habits). Weights, heights body mass index (BMI) measured. Study results showed that (39.4%) of sample were overweight and (24.1%) were obese. Prevalence of overweight and obesity was more common among males compared to females (40.8% and 25.2% vs. 35.7% and 21.45 respectively). In contrast 12.5% females were under weight compared to 5.4% males. Majority of the sample (68%) reported taking meals irregularly. A total of (64.3%) of the subjects reported the consumption of colored vegetable while (30%) reported eating fruits daily. More than half of both males and females did not eat fast foods. The prevalence of overweight and obesity were higher in males within the age of 20-25 years while in contrast it is higher in females within the age of more than 30 years (36% and 22.5% in males vs. 7.1% and 26% in females). More than one third of the participants did not engage in any sport exercises for at least 30 minutes during the week. Keywords: Overweight, Obesity, Food consumption, physical activity, Jordan

    Crystalline Field Effects on Magnetic and Thermodynamic properties of a Ferrimagnetic Centered Rectangular Structure

    The magnetic properties and phase diagrams of the mixed spin Ising model, with spins S=1 and {\sigma}=1/2 on a centered rectangular structure, have been investigated using Monte Carlo simulations based on the Metropolis algorithm. Every spin at one lattice site has four nearest-neighbor spins of the same type and four of the other type. We have assumed ferromagnetic interaction between the same spins type, antiferromagnetic for different spin types. An additional single-site crystal field term on the S=1 site was considered. We have shown that the crystal field enhances the existence of the compensation behavior of the system. In addition, the effects of the crystal field and exchange coupling on the magnetic properties and phase diagrams of the system have been studied.     Influence of Storage Temperature and Duration of Tomato Leaf Samples on Proline Content

    In arid and semi-arid countries such as Jordan, shortage in water sources might affect agricultural development and reduces the effectiveness of economic benefits of most crops planted in such areas. Tomato is an important agricultural crop and faces severe drought stress due to climate changes, therefore, measurement of proline accumulation in plant tissues is used as an indicator for drought stress tolerance. This research was conducted at Jarash University Campus in northern Jordan. A field experiment was carried out to investigate the impact of different storage temperature (+4ºC, - 20ºC and -80ºC) and different storage durations (0, 3, 6 and 11 weeks) on proline content in five different Jordanian tomato landraces. Results indicated that the average free proline content for samples tested directly after leaves collection was 7.1 µmol/g. Proline content in leaves stored at +4 ºC for 3, 6, and 11 weeks was 4.8, 1.8, and 1.1µmol/g, respectively, while for -20ºC was 11.8, 7.9, and 9.5 µmol/g for samples stored for 3, 6, 11 weeks respectively. In contrast the highest values for these parameters were obtained from samples stored at -80ºC, the average measured values of free proline content were 9.5, 7.8, and 12.9 µmol/g at 3, 6, and 11 weeks of storage, respectively. Based on the results obtained by this research, it is recommended to measure proline content directly after leaves collection. However, for large number of samples, keeping the samples at -20ºC not longer than six weeks could be a solution. Finally, we highly recommend the development of in-field method for measurement of free proline content

    Pathological and molecular study of kidneys in apparently healthy cattle and sheep with special reference to Leptospira species in central and northern Jordan

    Background and Aim: Renal patVhological conditions can cause significant economic losses for livestock owners. This study investigated and described the gross pathology and histopathology of lesions found in the kidneys of sheep and cattle, with particular attention to the presence of Leptospira spp. Materials and Methods: Three hundred and sixty kidneys from apparently healthy sheep and cattle (208 and 152, respectively) were collected from slaughterhouses in Jordan for gross and histopathological examinations, multiplex polymerase chain reaction (PCR) analysis, and gene sequencing of Leptospira spp. Results: Histopathological analysis of the samples revealed the following conditions: interstitial nephritis (4.44%), glomerulonephritis (4.16%), acute tubular necrosis (17.22%), and renal congestion (4.72%). In addition, in 26.9% of the samples, hyaline material was observed in the interstitium of the cortex and medulla. The PCR results revealed that 83 (23.1%) of the 360 samples were positive for Leptospira spp. 16S rRNA, 42/152 (28%) of cattle, and 41/208 (20%) of sheep. Four samples (two sheep and two cattle) were sequenced and deposited in GenBank with accession numbers from OL701310 to OL701313. Basic Local Alignment Search Tool search and nucleotide similarities between OL701311 and OL701313 resulted in the highest similarities with different Leptospira borgpetersenii strains, whereas OL701310 showed the highest nucleotide similarity (99.2%) with the Leptospira interrogans strain. Similarly, phylogenetic analysis revealed that OL701311 to OL701313 clustered together with different serovars of L. borgpetersenii, whereas OL701310 clustered with the L. interrogans clade. Conclusion: This is the first study to reveal a close association between pathogenic Leptospira spp. and kidney disorders in Jordanian cattle and sheep. These findings may help expand the current understanding of the causes and mechanisms of renal disease in cattle and sheep and contribute to developing more effective prevention and treatment programs

    Accurate Estimates of Microarray Target Concentration from a Simple Sequence-Independent Langmuir Model

    Background: Microarray technology is a commonly used tool for assessing global gene expression. Many models for estimation of target concentration based on observed microarray signal have been proposed, but, in general, these models have been complex and platform-dependent. Principal Findings: We introduce a universal Langmuir model for estimation of absolute target concentration from microarray experiments. We find that this sequence-independent model, characterized by only three free parameters, yields excellent predictions for four microarray platforms, including Affymetrix, Agilent, Illumina and a custom-printed microarray. The model also accurately predicts concentration for the MAQC data sets. This approach significantly reduces the computational complexity of quantitative target concentration estimates. Conclusions: Using a simple form of the Langmuir isotherm model, with a minimum of parameters and assumptions, and without explicit modeling of individual probe properties, we were able to recover absolute transcript concentrations with high R 2 on four different array platforms.     K-shell photoionization of ground-state Li-like carbon ions [C3+^{3+}]: experiment, theory and comparison with time-reversed photorecombination

    Absolute cross sections for the K-shell photoionization of ground-state Li-like carbon [C3+^{3+}(1s2^22s 2^2S)] ions were measured by employing the ion-photon merged-beams technique at the Advanced Light Source. The energy ranges 299.8--300.15 eV, 303.29--303.58 eV and 335.61--337.57 eV of the [1s(2s2p)3^3P]2^2P, [1s(2s2p)1^1P]2^2P and [(1s2s)3^3S 3p]2^2P resonances, respectively, were investigated using resolving powers of up to 6000. The autoionization linewidth of the [1s(2s2p)1^1P]2^2P resonance was measured to be 27±527 \pm 5 meV and compares favourably with a theoretical result of 26 meV obtained from the intermediate coupling R-Matrix method. The present photoionization cross section results are compared with the outcome from photorecombination measurements by employing the principle of detailed balance.Comment: 3 figures and 2 table

    Application of Equilibrium Models of Solution Hybridization to Microarray Design and Analysis

    Background: The probe percent bound value, calculated using multi-state equilibrium models of solution hybridization, is shown to be useful in understanding the hybridization behavior of microarray probes having 50 nucleotides, with and without mismatches. These longer oligonucleotides are in widespread use on microarrays, but there are few controlled studies of their interactions with mismatched targets compared to 25-mer based platforms. Principal Findings: 50-mer oligonucleotides with centrally placed single, double and triple mismatches were spotted on an array. Over a range of target concentrations it was possible to discriminate binding to perfect matches and mismatches, and the type of mismatch could be predicted accurately in the concentration midrange (100 pM to 200 pM) using solution hybridization modeling methods. These results have implications for microarray design, optimization and analysis methods. Conclusions: Our results highlight the importance of incorporating biophysical factors in both the design and the analysis of microarrays.     Prevalence, types and demographic features of child labour among school children in Nigeria

    BACKGROUND: To determine the prevalence, types and demographic features of child labour among school children in Nigeria. METHODS: A cross-sectional interview study of 1675 randomly selected public primary and secondary school pupils aged 5 to less than 18 years was conducted in the Sagamu Local Government Area of Ogun State, Nigeria from October 1998 to September 1999. RESULTS: The overall prevalence of child labour was 64.5%: 68.6% among primary and 50.3% among secondary school pupils. Major economic activities included street trading (43.6%), selling in kiosks and shops (25.4%) and farming (23.6%). No child was involved in bonded labour or prostitution. Girls were more often involved in labour activities than boys (66.8% versus 62.1%, p = 0.048): this difference was most obvious with street trading (p = 0.0004). Most of the children (82.2%) involved in labour activities did so on the instruction of one or both parents in order to contribute to family income. Children of parents with low socio-economic status or of poorly educated parents were significantly involved in labour activities (p = 0.01 and p = 0.001 respectively). Child labour was also significantly associated with increasing number of children in the family size (p = 0.002). A higher prevalence rate of child labour was observed among children living with parents and relations than among those living with unrelated guardians. CONCLUSION: It is concluded that smaller family size, parental education and family economic enhancement would reduce the pressure on parents to engage their children in labour activities
