    A perspective on the control of FES-supported standing

    This special section is about the control of electrical stimulators to restore standing functions to paraplegics. It addresses several important topics regarding the interactions of the intact central nervous systems (CNS) with the artificial control system. The topics are as follows: how paraplegics use their arms to help themselves stand up with functional electrical stimulation (FES); the user-driven artificial control of FESsupported standing up; a controller which is promising for the control of sitting down; the application of reinforcement machine learning for the controllers of standing up; arms-free\ud standing with voluntary upper body balancing and artificially controlled ankle stiffness; and cognitive feedback in balancing. This Commentary introduces the papers in this section and relates them to earlier research

    Feedback control of unsupported standing

    This paper presents the results of continuing work on feedback control of unsupported standing in paraplegia. Our experimental setup considers a situation in which all joints above the ankle are braced, and stabilising torque at the ankle is generated by stimulation of the plantarflexors. A previous study showed that short periods of unsupported standing with paraplegic subjects could be achieved. In order to improve consistency and reliability of unsupported standing we are currently investigating several modifications to the control strategy. The paper reports progress towards this goal

    Design of feedback controllers for paraplegic standing

    The development, implementation and experimental evaluation of feedback systems for the control of the upright posture of paraplegic persons in standing is described. While the subject stands in a special apparatus, stabilising torque at the ankle joint is generated by electrical stimulation of the paralysed calf muscles of both legs using surface electrodes. This allows the subject to stand without the need to hold on to external supports for stability- this is termed 'unsupported standing'. Sensors in the apparatus allow independent measurement of left and right ankle moments together with measurement of the inclination angle. A nested loop structure for control of standing is implemented, where a high-bandwidth inner loop provides control of the ankle moments, while the angle controller in the outer loop regulates the inclination angle. A number of important modifications to a control strategy which was previously tested with both neurologically intact and paraplegic subjects are presented. The new strategy is described, and an experimental evaluation with intact subjects is reported. The experimental results show that the control system for unsupported standing performs reliably, and according to the design formulation. There aa-e a number of design choices, appropriate to different situations, and the practical effect of each is clear. This allows easy 'tuning' during an experimental session. This is important since the complete design procedure, from muscle dynamics identification to control design, has to be carried out as quickly as possible while the subject is standing in the apparatus. A number of recommendations are made regarding the preferred design choices for control of unsupported standing

    © 2015 Mitchell et al. Abstract Background: Mass spectrometry imaging (MSI) is a powerful tool for the study of intact tissue sections. Here, its application to the study of the distribution of lipids in sections of reconstructed living skin equivalents during their development and maturation is described. Methods: Living skin equivalent (LSE) samples were obtained at 14 days development, re-suspended in maintenance medium and incubated for 24 h after delivery. The medium was then changed, the LSE re-incubated and samples taken at 4, 6 and 24 h time points. Mass spectra and mass spectral images were recorded from 12 ÎŒm sections of the LSE taken at each time point for comparison using matrix assisted laser desorption ionisation mass spectrometry. Results: A large number of lipid species were identified in the LSE via accurate mass-measurement MS and MSMS experiments carried out directly on the tissue sections. MS images acquired at a spatial resolution of 50 ÎŒm × 50 ÎŒm showed the distribution of identified lipids within the developing LSE and changes in their distribution with time. In particular development of an epidermal layer was observable as a compaction of the distribution of phosphatidylcholine species. Conclusions: MSI can be used to study changes in lipid composition in LSE. Determination of the changes in lipid distribution during the maturation of the LSE will assist in the identification of treatment responses in future investigations

    Variability of Objectively Measured Sedentary Behavior

    The primary purpose of this study was to evaluate variability of sedentary behavior (SB) throughout a 7-d measurement period and to determine if G7 d of SB measurement would be comparable with the typical 7-d measurement period. Methods: Retrospective data from Ball State University_s Clinical Exercise Physiology Laboratory on 293 participants (99 men, 55 T 14 yr, body mass index = 29 T 5 kgImj2; 194 women, 51 T 12 yr, body mass index = 27 T 7 kgImj2) with seven consecutive days of data collected with ActiGraph accelerometers were analyzed (ActiGraph, Fort Walton Beach, FL). Time spent in SB (either G100 counts per minute or G150 counts per minute) and breaks in SB were compared between days and by sex using a two-way repeated-measures ANOVA. Stepwise regression was performed to determine if G7 d of SB measurement were comparable with the 7-d method, using an adjusted R2 of Q0.9 as a criterion for equivalence. Results: There were no differences in daily time spent in SB between the 7 d for all participants. However, there was a significant interaction between sex and days, with women spending less time in SB on both Saturdays and Sundays than men when using the 100 counts per minute cut-point. Stepwise regression showed using any 4 d would be comparable with a 7-d measurement (R2 9 0.90). Conclusions: When assessed over a 7-d measurement period, SB appears to be very stable from day to day, although there may be some small differences in time spent in SB and breaks in SB between men and women, particularly on weekend days. The stepwise regression analysis suggests that a measurement period as short as 4 d could provide comparable data (91% of variance) with a 1-wk assessment. Shorter assessment periods would reduce both researcher and subject burden in data collection
