    Law, Deviation and Paradigmatic Change: Copyright and its Metaphors

    Drawing on debates in Sweden about Internet freedom, particularly those connected to copyright and file sharing, and on the European legislative trend of amending copyright, this chapter analyses metaphors and conceptions in terms of a societal paradigmatic shift and the collision of mentalities. Kuhnian paradigms are wedded with the mentalities of the French Annales School of historic research. The chapter argues that the “building blocks” of these mentalities and paradigms can be studied in metaphors, in public debates or in legislation, which may reveal the conceptions they emanate from. This chapter touches upon ethical, moral and legal issues related to the digitalization of society. The relevancy of this chapter in relation to the theme of the book is found in the conceptualization of “deviancy”. One has to ask from what perspective or paradigm the judgment of the behavior takes place, and in what historical context it is made

    HĂ„kan Larsson: Sport and gender This article concerns bodily materialisation as it occurs in youth sport. It is based on interviews with teenagers 16 to 19 years of age doing track and field athletics. The purpose of the article is to elucidate how the notion of a “natural body“ can be seen as a cultural effect of sports practice and sports discourse. On the one hand, the body is materialised as a performing body, and on the other as a beautiful body. The “performing body“ is a single-sexed biological entity. The “beautiful body“ is a double-sexed and distinctly heterosexually appealing body. As these bodies collide in teenager track and field, the female body materialises as a problematic body, a body that is at the same time the subject of the girl’s personality. The male body materialises as an unproblematic body, a body that is the object of the boy’s personality. However, the body as “(a problematic) subject“ or as “(an unproblematic) object“ is not in itself a gendered body. Rather, these are positions on a cultural grid of power-knowledge relations. A girl might position herself in a male discourse, and a boy might position himself in a female discourse, but in doing so, they seem to have to pay a certain price in order not to be seen as queer

    Rapid effects of progesterone on ciliary beat frequency in the mouse fallopian tube

    <p>Abstract</p> <p>Background</p> <p>The physiological regulation of ciliary beat frequency (CBF) within the fallopian tube is important for controlling the transport of gametes and the fertilized ovum. Progesterone influences gamete transport in the fallopian tube of several mammalian species. In fallopian tubes isolated from cows, treatment with 20 micromolar progesterone caused a rapid reduction of the tubal CBF. The aims of this study were to establish methodology for studying fallopian tube CBF in the mouse, as it is an important model species, and to investigate if progesterone rapidly affects the CBF of mice at nM concentrations.</p> <p>Methods</p> <p>A method to assess tubal CBF of mice was developed. Fallopian tubes were dissected and the tissue was cut in small pieces. Tissue samples with moving cilia were located under an inverted bright field microscope and held still against the bottom of a petri dish by a motorized needle system. Images were acquired over 90 minutes at 35 degrees C with a high-speed camera and used for assessing changes in the CBF in response to the addition of hormone.</p> <p>Results</p> <p>The baseline CBF of the mouse fallopian tube was 23.3 +/- 3.8 Hz. The CBF was stable over at least 90 minutes allowing establishment of a baseline frequency, addition of hormone and subsequent recordings. Progesterone at concentrations of 20 micromolar and 100 nM significantly reduced the CBF by 10% and 15% respectively after 30 minutes compared with controls.</p> <p>Conclusions</p> <p>The present study demonstrates that the mouse, despite its small size, is a useful model for studying the fallopian tube CBF ex vivo. The rapid reduction in CBF by 100 nM progesterone suggests that gamete transport in the fallopian tube could be mediated by progesterone via a non-genomic receptor mechanism.</p

    Gendered relations? Associations between Swedish parents, siblings, and adolescents' time spent sedentary and physically active

    IntroductionThe family is assumed to be fundamental in youth socialization processes and development, connected to social and cultural practices such as healthy lifestyles and physical activity. However, gender patterns in physical activity among adolescents and the structural drivers of gender inequality (e.g., parentage and siblingship) are poorly understood. The aim of this study was to explore further how gender structures relate to adolescents' time spent being sedentary and physically active, using contemporary gender theory.MethodsThis cross-sectional study involved 1,139 adolescents aged 13-14 and their parents, including 815 mothers and 572 fathers. Physical activity and time spent sedentary were assessed through accelerometry among adolescents and through a self-report questionnaire for parents validated against accelerometry.ResultsThe results showed significant relationships between mothers' moderate-to-vigorous physical activity (MVPA) and girls' MVPA on weekdays and weekends, and fathers' MVPA was significantly related to girls' MVPA on weekdays. Our results imply that the relationship between Swedish parents' and adolescent girls' physical activity in higher intensities are to some extent gendered practices. However, time spent sedentary does not seem to show any patterns of being performed according to binary ideas of gender. Further, our exploratory analyses suggest that these results somewhat intersect with parents' educational level and relate to intra-categorical aspects of doing gender. The results also indicate slight gendered patterns in the “doing” of brotherhood for time spent sedentary, however, for boys only on weekends.DiscussionThe study contributes to the understanding of gender norms as constraints and enablers for adolescents' participation in physical activity. The results can spur public health and physical activity research to apply a contemporary gender theory approach, and to expand the research agenda connected to what relates to gender inequalities in physical activity practices

    Distribution and hormonal regulation of membrane progesterone receptors ÎČ and Îł in ciliated epithelial cells of mouse and human fallopian tubes

    <p>Abstract</p> <p>Background</p> <p>The controlled beating of cilia of the fallopian tube plays an important role in facilitating the meeting of gametes and subsequently transporting the fertilized egg to its implantation site. Rapid effects of progesterone on ciliary beat frequency have been reported in the fallopian tubes of cows, but the identity of the receptors mediating this non-genomic action of progesterone is not known. We recently identified a member of the non-genomic membrane progesterone receptor family, mPR gamma, as a candidate for mediating these actions of progesterone. Here, we investigated the possible presence of a related receptor, mPR beta, in the fallopian tubes of mice and women as well as the possible hormonal regulation of mPR beta and gamma.</p> <p>Methods</p> <p>Western blot and immunohistochemistry with specific antibodies were used to characterize the expression and cellular localization of the mPRs in mouse and human tissues. Taqman (Quantitative Polymerase Chain Reaction) assays were used to quantify mRNA levels in the fallopian tubes of two different mouse models after injections with different hormones and specific antagonists.</p> <p>Results</p> <p>In the fallopian tubes of both mouse and human, the expression of mPR beta and mPR gamma proteins was exclusively found in the ciliated cells. Whereas mPR beta was found on the cilia, mPR gamma was localized at the base of the same ciliated cells, as previously reported. In gonadotropin-primed mice, both mPRs genes were down-regulated after an injection with progesterone. Treatment with estradiol rapidly down-regulated the level of mPR beta mRNA and protein in immature mice. The mPR gamma protein was down-regulated around the time of ovulation in cycling women, similar to the regulation observed in mice stimulated to ovulate via gonadotropin injections.</p> <p>Conclusion</p> <p>Our findings show the presence and hormonal regulation of two distinct mPRs associated with the cilia of the fallopian tubes in both mice and women. It is hypothesized that these receptors are involved in the control of ciliary movement and, thus, gamete transport in the fallopian tubes of mammals.</p

    Simultaneous inhibition of B7 and LFA-1 signaling prevents rejection of discordant neural xenografts in mice lacking CD40L.

    Transplantation of embryonic human neural tissue can restore dopamine neurotransmission and improve neurological function in patients with Parkinson's disease. Logistical and ethical factors limit the availability of human embryonic allogeneic tissue. Embryonic xenogeneic neural tissue from porcine donors is an alternative form of donor tissue, but effective immunomodulatory techniques are warranted for neural xenotransplantation to become clinically feasible. We transplanted embryonic porcine ventral mesencephalic tissue into the brains of adult untreated C57BL/6 mice, untreated CD40L-/-mice and CD40L-/-mice that received injections of anti-LFA-1, CTLA41g or both compounds. Double-treated CD40L-/-mice had large grafts with high numbers of dopaminergic neurons 4 wk after transplantation. The grafts were completely devoid of lymphocytes, macrophages and activated microglia. Untreated C57BL/6 mice had rejected their grafts. Untreated CD40L-/-mice and CD40L-/-mice treated with monotherapy of anti-LFA-1 or CTLA41g had smaller grafts and more microglial and lymphocytic infiltration than double-treated CD40L-/-mice. We conclude that immunomodulation with concomitant inhibition of LFA-1 and B7 signaling in the perioperative period in CD40L-/-mice prevented the rejection of discordant neural xenografts. The treatment most likely reduced antigen presenting capacity and interfered with the costimulatory signaling needed for T cell activation to occur

    Biogas Digestate in Vegetable Hydroponic Production: pH Dynamics and pH Management by Controlled Nitrification

    Soilless cultivation of vegetables with digestate fertilizer from biogas production as a nutrient source is a promising method for integrating food production and organic waste management. In this study, bok choy (Brassica rapa var. chinensis) was cultivated in a hydroponic nutrient film technique system with biogas digestate as the only fertilizer source. Nitrification in moving bed biofilm reactors (external and/or integrated into the hydroponic cultivation system) was employed to lower the high ammonium concentration in the digestate prior to use. Treatments with differing nitrification and digestate input strategies were compared with respect to pH dynamics, crop growth, shoot water content, and shoot mineral content. The results showed that < 20% longer cultivation time (< 1 week) gave similar yield in biogas digestate-based hydroponics as in conventional hydroponics based on synthetic fertilizers. Automatic pH control through addition of digestate resulted in similar shoot dry weight as in the mineral fertilizer reference system. It can be concluded that biogas digestate is a suitable plant nutrient source for hydroponic production of bok choy, considering productivity and circularity aspects. The combined impact of digestate ammonium content, digestate pH, and the nitrification step needs to be considered when designing a hydroponic system with biogas digestate as the fertilizer source.     Food waste to new food: Risk assessment and microbial community analysis of anaerobic digestate as a nutrient source in hydroponic production of vegetables

    In this study, the microbiological food safety of using anaerobic digestate as a fertilizer in hydroponic production of vegetables was evaluated. The used anaerobic digestate was a liquid residue obtained from the digestion of food waste in the production of biogas. Replacing the customary inorganic fertilizer used in hydroponic production with this recycled fertilizer (biofertilizer) could allow for sustainable urban food production close to retailers and consumers. However, in striving for circular food production, it is vital that the food safety of utilizing recycled resources is ensured. Especially in the application of hydroponic farming, where the nutrient loop is shorter than on arable land, a microbiological food safety risk assessment is crucial when adopting new and recycled fertilizers. The biofertilizer based on anaerobic digestate was therefore studied with regard to its microbial community (16S rRNA gene amplicon sequencing) during production of vegetables in a hydroponic system. The biofertilizer was also challenge tested with food borne pathogens (Salmonella enterica, Listeria monocytogenes and Bacillus cereus). Furthermore, the microbial activity of the biofertilizer was studied using isothermal calorimetry. The results showed that the microbial community of the biofertilizer changed distinctly through a necessary initial nitrification process, and that the most abundant genus was Mycobacterium. Deliberate contaminations with 5 log10 CFU mL−1 of either S. enterica or L. monocytogenes in the nitrified biofertilizer were no longer detectable with selective plating after 48 h of incubation at 20 °C. Selective plating for B. cereus revealed that the biofertilizer contained low levels (∌10 CFU mL−1) of the bacterium, and an inoculation of 5 log10 CFU mL−1 B. cereus decreased to these levels within 24 h of incubation at 20 °C. Analysis of the microbial activity of the biofertilizer indicated that the biofertilizer does not seem to support microbial activity without the addition of an external nutrient source that contains an accessible carbon source and trace elements. The type of biofertilizer investigated in this study is thus regarded as microbiologically safe for use in hydroponic cultivation. The constant presence of viable B. cereus, however, emphasizes the fundamental importance of continuous risk assessment in case of any modifications or supplementations of the biofertilizer, since it clearly can act as a reservoir for bacterial endospores
