    Poststructuralist theory focuses largely on describing how and why subjects reproduce the social conditions they have internalised. This is a deconstruction of the central idea of the Enlightenment, the human capacity for autonomous action. At the same time, however, it also denies all individuals any responsibility and ultimately leads criticism into a crisis. Pragmatist philosophy offers the possibility of determining the role of the mind in processes of becoming a subject without abandoning the achievements of the poststructuralist concept of subjectification. The concept of transaction describes how actors constitute each other as subjects within social situations. The relationships that arise through such processes depend, among other things, on the personalities of those people involved. Accordingly, it is possible to identify the responsibility of individuals to govern their social relations and personality development. Since these aspects can only be determined in localised individual cases, this offers a particularly suitable starting point for geographical critic.</p

    GesamtdissertationDie Ergebnisse nach LTX der Charite Berlin im Beobachtungszeitraum von 15 Jahren (7/1988-9/2003)bei leberassoziierten Stoffwechselerkrankungen sind mit anderen Transplantationszentren bzw. anderen Indikationsgruppen vergleichbar.Die Ergebnisse der 71 Patienten ist exzellent mit einer Überlebensrate von 80% nach 10 Jahren post LTX und einer deutlichen Verbesserung der Lebensqualität.Somit stellt die LTX eine adäquate und effiziente Therapieoption bei genetisch bedingten leberassoziierten Stoffwechselerkrankungen dar.The outcome after LTX in the Charite Berlin for 15 years observation (7/1988-9/2003) for hepatic associated metabolic disorders is with other transplantation centers and other indicationgroups comparable.The outcome of the 71 patients is excellent with a rate of survive of 80% after 10 years after LTX and a realy improved quality of life.Because of this facts, LTX is an appropriate and efficient treatment for genetic hepatic associated metabolic disorders

    Comparative Analysis of Acute Kidney Injury Models and Related Fibrogenic Responses: Convergence on Methylation Patterns Regulated by Cold Shock Protein

    Background: Fibrosis is characterized by excessive extracellular matrix formation in solid organs, disrupting tissue architecture and function. The Y-box binding protein-1 (YB-1) regulates fibrosis-related genes (e.g., Col1a1, Mmp2, and Tgfβ1) and contributes significantly to disease progression. This study aims to identify fibrogenic signatures and the underlying signaling pathways modulated by YB-1. Methods: Transcriptomic changes associated with matrix gene patterns in human chronic kidney diseases and murine acute injury models were analyzed with a focus on known YB-1 targets. Ybx1-knockout mouse strains (Ybx1ΔRosaERT+TX and Ybx1ΔLysM) were subjected to various kidney injury models. Fibrosis patterns were characterized by histopathological staining, transcriptome analysis, qRT-PCR, methylation analysis, zymography, and Western blotting. Results: Integrative transcriptomic analyses revealed that YB-1 is involved in several fibrogenic signatures related to the matrisome, the WNT, YAP/TAZ, and TGFß pathways, and regulates Klotho expression. Changes in the methylation status of the Klotho promoter by specific methyltransferases (DNMT) are linked to YB-1 expression, extending to other fibrogenic genes. Notably, kidney-resident cells play a significant role in YB-1-modulated fibrogenic signaling, whereas infiltrating myeloid immune cells have a minimal impact. Conclusions: YB-1 emerges as a master regulator of fibrogenesis, guiding DNMT1 to fibrosis-related genes. This highlights YB-1 as a potential target for epigenetic therapies interfering in this process
