Contribución al estudio de las algas patógenas para el hombre del género Prothoteca
Las algas del género Prototheca fueron descritas por primera vez
en 1.894 por Kmger y han aumentado en importancia al englobarlas
dentro de los agentes patógenos oportunistas humanos.
La entidad patógena que producen se la conoce como
Protothecosis, que es una infección rara y provoca infecciones cutáneas ó
bursitis óleocraneana y con menos frecuencia una patología sistémica.
Estas algas del género Prototheca con varias especies P. wickerhamii
(P.w), P. zopfii (P.z), P. stagnora (P.s) sólo la P.w. y P.z. son patógenos
humanos.
El objetivo de nuestro trabajo ha sido el estudio del comportamiento
de las dos especies de Protothecas patógenas humanas frente
a antirnicrobianos comercializados.
El material utilizado ha sido un total de 68 cepas de las que 44
son P.w. y 24 P.z. procedentes de distintas colecciones extranjeras cuyos
orígenes son cutáneos, de un hemocultivo, de heces y aguas residuales y
el resto de cepas aisladas en nuestro Departamento procedentes de aguas
residuales.
Los medios utilizados para probar la sensibilidad frente a los antimicrobianos
el Agar Sabouraud dextrosa. Los agentes antimicrobianos
ensayados fueron en dos modalidades: en disco y en polvo. En disco
fueron un total de 87 que distribuidos por familias 12 Aminoglucósidos,
11 B-lactárnicos (Penicilinas), 11 B-lactámicos (Cefalosporinas), 4
Macrólidos, 3 Polipeptídicos, 2 Sinergístinas, 2 Amsamínas, 2
Lincosamidas, 7 Tetraciclinas, 8 Antimicrobianos sin gmpo especial, 10
Quimioterápicos en síntesis 9 Sulfamidas y 6 Antifúngicos. Los
antibióticos en polvo fueron en total 9: Amikacina, DKB, Framicetina,
Kanamicina, Neomicina, Paromomicina, Ribostamicina, Sisomicina y
Tobramicina.
Con respecto a la Metodología utilizada, para estudio de las
sensibilidades se realizó por dos métodos, en primer lugar se hizo por el
método de Kirbi-Bauer modificado a continuación se hizo la CMI
mediante la dilución del antimicrobiano en Agar Sabouraud dextrosa.
Los resultados obtenidos por el método disco-placa de los Aminoglucósidos frente a Jos Protothecas, patógenos lnunanos, fueron
inhibidas Ja casi totalidad de las cepas estudiadas, si bien frente a Ja
Estreptomicina el nº de cepas inhibidas fue menor 48% para Pw, 11 %
para Pz. Los Polipeptídicos, colistina y polimixina inhibieron a Ja
totalidad de las cepas tanto de Pw como Pz, siendo rt!sistentes a la
bacitrina. De Jos Quimioterápicos de síntesis sólo Ja Mandelamina y Ja
Nitro-5-Hí<lrox-quinoleina inhibieron tanto a Ja Pw como Pz, el resto
fueron ineficaces. Los Antifúngicos salvo Ja 5 11uorocitosina inhibieron a
todas las cepas <le ProtoU1ecas probadas. El resto <le Jos Antibióticos
fueron ineficaces, B-lactámicos, Macrólidos, Sinergístinas, Ansaminas,
Lincosamidas, T etraciclina y Sulfamidas.
La Evolución di! Ja CMI de Jos Aminoglucósidos se obtuvieron
unos resultados variables. Para Ja Pw frente a Ja paromomicina se
bastaron concentrnciones de 16mcg/ml para el 95% de las cepas y para
Kanamicina y Ribostomicina precisaron 32mcg/ml para inhibir el 100%
de las cepas. Para el resto de Jos aminoglucósidos probados, percibieron
concentraciones mayores.
Para Ja Pz la Ribostomicina precisó de 4 mcg/ml para inhibir el
100% de las cepas probadas 16 mcg/ml para la Tobramicina y 32mcg/ml
para Amikacina, Kanamicina, Neomicina, Sisomicina.Algae of the genus Prototheca, closely related to the chlorellas

Seventy-eight strains were assayed: 44 Prototheca wickerhamii,
24 P zopfii. The strains came from our department's collections
intemational collections belonging to the following researchers. Wild
Strains isolated in our laboratory from residual waters were also assaye<l.
The outputs gotten by the method disk-badge of the Aminoglucosidos
opposite the Protothecas, human patogens, was inhibited
the almost entirety of the studied stumps, although opposite the
Estreptomicina the nwnber of stumps inhibited was minor 48% for Pw,
11% for Pz. The Polipeptidicos, colistina and polimixine inhibited to the
entirety of the slumps so much of Pw like Pz, being resistan! to the
bacitricina. From the Chemiotherapics of only synthesis the Mandelamine
and the Nitro-5-Hidrox-quinoleine so much inhibited to the Pw
like Pz, the remainder was inefTective. The Antifungics except for the 5
tluorocitosine inhibited to ali the stumps of proven Protothecas. The
remainder of the Antibiotics was inellective, B-lactamics, Macrolids,
Sinergistines, Ansamines, Lincosamides, Tetracicline and Sulfamides.
TI1e Evolution of the CMI of the Aminoglucosides was gotten
some variables outputs. For the Pw opposite the paromomicine
concentrations of 16 mcg were sulliced/ ml for the 95% of U1e stumps
and for Kanamicine and Ribostomicine specified 32 mg/ml for inhibited
the 100% of the stumps. For the remainder of the proven aminoglucosides,
they perceived old concentrations.
For the Pz the Ribostomicina specified of 4 mcg/ml for inhibited
the 100% of tl1e proven stumps 16 mcg/ml for the Tobramicina and 32
mcg/ml for Amikacine, Kanamicine, Neomicine, Sisomicine
Evaluation of a new automated Abbott RealTime MTB RIF/INH assay for qualitative detection of rifampicin/isoniazid resistance in pulmonary and extra-pulmonary clinical samples of Mycobacterium tuberculosis
A new automated real-time PCR assay for the detection of rifampicin (RIF) and
isoniazid (INH) resistance in Mycobacterium tuberculosis (MTB) was evaluated. A total of 163
clinical samples (128 pulmonary and 35 extra-pulmonary) were processed using four PCR assay
kits: Abbott RealTime MTB RIF/INH, Genotype MTBDRplus, Xpert/MTB RIF, and Anyplex
MTB/MDR. The results of phenotypic drug-susceptibility testing using BACTECMGIT 960
were used as reference. The sensitivity and specificity of the new Abbott RealTime MTB RIF/
INH assay in comparison with phenotypic testing was 96.3% (95%CI 87.32%–100%) for RIF
and 100% (95%CI 99.3%–100%) for INH; the sensitivity was 78.8% (95%CI 66.8%–90.9%) and
the specificity was 100% (95%CI 98.9%–100%). The Abbott RealTime MTB RIF/INH test could
be a valid method for detecting the most common mutations in strains resistant to RIF and INH
El virus de la hepatitis C como patógeno emergente I: diagnóstico mediante métodos de laboratorio
La infección por el virus de la hepatitis C se ha converti-
do en una enfermedad emergente en la población de pa-
cientes VIH+/SIDA. En los últimos años se han desarro-
llado multitud de técnicas de laboratorio para permitir
