    Comparative evaluation of therapy results in mild acne

    Introduction. Mild acne is the most underestimated form of the disease.The aim of the study was to compare the results of external combination therapy with 15% azelaic acid gel + 1% clindamycin gel and benzoyl peroxide + clindamycin for mild papulopustular acne.Material and methods. An open comparative prospective observational study in parallel groups (12 weeks) included 96 people, 65 patients with mild papulo-pustular acne and a control group of 30 healthy individuals. Group I received 15% azelaic acid gel +1% clindamycin gel, group II – benzoyl peroxide gel + clindamycin. The number of acne elements, morphofunctional indicators of the facial skin, side effects were recorded.Results. As a result of treatment, the indicators of sebumetry significantly decreased: in group I in the T-zone 47.5 ± 11.3 units (p = 0.043) and in the U-zone 57.5 ± 6.3 units (p = 0.037), and in group II in the T-zone 37.1 ± 5.8 units (p = 0.015) and in the U-zone 48.7 ± 5.6 units (p = 0.027). The pore sizes decreased significantly: in group I in the T-zone 0.052 ± 0.010 mm (p = 0.046) and in the U-zone 0.054 ± 0.009 mm (p = 0.049), and in group II in the T-zone 0.039 ± 0.011 mm (p = 0.064) and in the U-zone 0.047 ± 0.013 mm (p = 0.041). In patients in group I, pigmentation indicators in the T-zone of 17.0 ± 2.8 units (p < 0.001) and the U-zone of 17.0 ± 2.8 units (p = 0.048) were normalized. In patients in both groups, there was a significant (p >< 0.05) decrease in the number of papulopustules.>˂ 0.001) and the U-zone of 17.0 ± 2.8 units (p = 0.048) were normalized. In patients in both groups, there was a significant (p ˂ 0.05) decrease in the number of papulopustules.Conclusion. When comparing the results of therapy with 15% azelaic acid gel + 1% clindamycin gel and benzoyl peroxide + adapalene for mild papulopustular acne, comparable efficacy and safety were revealed

    Thermodynamic properties of myo-inositol

    © 2017 Elsevier Ltd In the present work, the temperature dependence of heat capacity of vitamin B8 (myo-inositol) has been measured for the first time over the range from 8 K to 340 K by precision adiabatic vacuum calorimetry. Based on the experimental data, the thermodynamic functions of the vitamin B8, namely, the heat capacity, enthalpy H°(T)–H°(0), entropy S°(T)–S°(0) and Gibbs function G°(T)–H°(0) have been determined for the range from T → 0 K to 340 K. The value of the fractal dimension D in the function of multifractal generalization of Debye's theory of the heat capacity of solids was estimated and the character of heterodynamics of structure was detected. The enthalpy of combustion (−2747.0 ± 2.1) kJ·mol−1 of the vitamin B8 was measured for the first time using high-precision combustion calorimeter. The standard molar enthalpy of formation in the crystalline state (−1329.3 ± 2.3) kJ·mol−1 of B8 at 298.15 K was derived from the combustion experiments. Using combination of the adiabatic and combustion calorimetry results the thermodynamic functions of formation of the myo-inositol at T = 298.15 K and p = 0.1 MPa have been calculated.     From Barbie to the oligarchs wife: Reading fantasy femininity and globalisation in post-Soviet Russian womens magazines

    This article shows how an analysis of fantasy femininity sheds light on how norms of gender, class and national identity reflect global and local cross-cultural currents in post-Soviet Russia. Drawing on a discourse analysis of women’s magazines and in-depth interviews with readers, it shows how, in the globalized post-Soviet cultural landscape, fantasy femininity represents both change and continuity. Feminine archetypes in women’s magazines, from fairytale princesses to Barbie dolls, reflect a wider post-Soviet cultural hybridisation, and show how Western women’s magazines have adapted to the Russian context. Furthermore, the article highlights readers’ ambiguous attitudes towards post-Soviet cultural trends linked to perceived Westernisation or globalisation, such as individualism, conspicuous consumption, and glamour

    Tight correlation between expression of the Forkhead transcription factor FOXM1 and HER2 in human breast cancer

    BACKGROUND: FOXM1 regulates expression of cell cycle related genes that are essential for progression into DNA replication and mitosis. Consistent with its role in proliferation, elevated expression of FOXM1 has been reported in a variety of human tumour entities. FOXM1 is a gene of interest because recently chemical inhibitors of FOXM1 were described to limit proliferation and induce apoptosis in cancer cells in vitro, indicating that FOXM1 inhibitors could represent useful anticancer therapeutics. METHODS: Using immunohistochemistry (IHC) we systematically analysed FOXM1 expression in human invasive breast carcinomas (n = 204) and normal breast tissues (n = 46) on a tissue microarray. Additionally, using semiquantitative realtime PCR, a collection of paraffin embedded normal (n = 12) and cancerous (n = 25) breast tissue specimens as well as benign (n = 3) and malignant mammary cell lines (n = 8) were investigated for FOXM1 expression. SPSS version 14.0 was used for statistical analysis. RESULTS: FOXM1 was found to be overexpressed in breast cancer in comparison to normal breast tissue both on the RNA and protein level (e.g. 8.7 fold as measured by realtime PCR). We found a significant correlation between FOXM1 expression and the HER2 status determined by HER2 immunohistochemistry (P < 0.05). Univariate survival analysis showed a tendency between FOXM1 protein expression and unfavourable prognosis (P = 0.110). CONCLUSION: FOXM1 may represent a novel breast tumour marker with prognostic significance that could be included into multi-marker panels for breast cancer. Interestingly, we found a positive correlation between FOXM1 expression and HER2 status, pointing to a potential role of FOXM1 as a new drug target in HER2 resistant breast tumour, as FOXM1 inhibitors for cancer treatment were described recently. Further studies are underway to analyse the potential interaction between FOXM1 and HER2, especially whether FOXM1 directly activates the HER2 promoter

    Peroxiredoxin 3 Is a Redox-Dependent Target of Thiostrepton in Malignant Mesothelioma Cells

    Thiostrepton (TS) is a thiazole antibiotic that inhibits expression of FOXM1, an oncogenic transcription factor required for cell cycle progression and resistance to oncogene-induced oxidative stress. The mechanism of action of TS is unclear and strategies that enhance TS activity will improve its therapeutic potential. Analysis of human tumor specimens showed FOXM1 is broadly expressed in malignant mesothelioma (MM), an intractable tumor associated with asbestos exposure. The mechanism of action of TS was investigated in a cell culture model of human MM. As for other tumor cell types, TS inhibited expression of FOXM1 in MM cells in a dose-dependent manner. Suppression of FOXM1 expression and coincidental activation of ERK1/2 by TS were abrogated by pre-incubation of cells with the antioxidant N-acetyl-L-cysteine (NAC), indicating its mechanism of action in MM cells is redox-dependent. Examination of the mitochondrial thioredoxin reductase 2 (TR2)-thioredoxin 2 (TRX2)-peroxiredoxin 3 (PRX3) antioxidant network revealed that TS modifies the electrophoretic mobility of PRX3. Incubation of recombinant human PRX3 with TS in vitro also resulted in PRX3 with altered electrophoretic mobility. The cellular and recombinant species of modified PRX3 were resistant to dithiothreitol and SDS and suppressed by NAC, indicating that TS covalently adducts cysteine residues in PRX3. Reduction of endogenous mitochondrial TRX2 levels by the cationic triphenylmethane gentian violet (GV) promoted modification of PRX3 by TS and significantly enhanced its cytotoxic activity. Our results indicate TS covalently adducts PRX3, thereby disabling a major mitochondrial antioxidant network that counters chronic mitochondrial oxidative stress. Redox-active compounds like GV that modify the TR2/TRX2 network may significantly enhance the efficacy of TS, thereby providing a combinatorial approach for exploiting redox-dependent perturbations in mitochondrial function as a therapeutic approach in mesothelioma

    Molecular Determinants and Dynamics of Hepatitis C Virus Secretion

    The current model of hepatitis C virus (HCV) production involves the assembly of virions on or near the surface of lipid droplets, envelopment at the ER in association with components of VLDL synthesis, and egress via the secretory pathway. However, the cellular requirements for and a mechanistic understanding of HCV secretion are incomplete at best. We combined an RNA interference (RNAi) analysis of host factors for infectious HCV secretion with the development of live cell imaging of HCV core trafficking to gain a detailed understanding of HCV egress. RNAi studies identified multiple components of the secretory pathway, including ER to Golgi trafficking, lipid and protein kinases that regulate budding from the trans-Golgi network (TGN), VAMP1 vesicles and adaptor proteins, and the recycling endosome. Our results support a model wherein HCV is infectious upon envelopment at the ER and exits the cell via the secretory pathway. We next constructed infectious HCV with a tetracysteine (TC) tag insertion in core (TC-core) to monitor the dynamics of HCV core trafficking in association with its cellular cofactors. In order to isolate core protein movements associated with infectious HCV secretion, only trafficking events that required the essential HCV assembly factor NS2 were quantified. TC-core traffics to the cell periphery along microtubules and this movement can be inhibited by nocodazole. Sub-populations of TC-core localize to the Golgi and co-traffic with components of the recycling endosome. Silencing of the recycling endosome component Rab11a results in the accumulation of HCV core at the Golgi. The majority of dynamic core traffics in association with apolipoprotein E (ApoE) and VAMP1 vesicles. This study identifies many new host cofactors of HCV egress, while presenting dynamic studies of HCV core trafficking in infected cells
