    MLVA genotyping of Chinese human Brucella melitensis biovar 1, 2 and 3 isolates

    <p>Abstract</p> <p>Background</p> <p>Since 1950, <it>Brucella melitensis </it>has been the predominant strain associated with human brucellosis in China. In this study we investigated the genotypic characteristics of <it>B. melitensis </it>isolates from China using a multiple-locus variable-number tandem-repeat analysis (MLVA) and evaluated the utility of MLVA with regards to epidemiological trace-back investigation.</p> <p>Results</p> <p>A total of 105 <it>B. melitensis </it>strains isolated from throughout China were divided into 69 MLVA types using MLVA-16. Nei's genetic diversity indices for the various loci ranged between 0.00 - 0.84. 12 out 16 loci were the low diversity with values < 0.2 and the most discriminatory markers were bruce16 and bruce30 with a diversity index of > 0.75 and containing 8 and 7 alleles, respectively. Many isolates were single-locus or double-locus variants of closely related <it>B. melitensis </it>isolates from different regions, including the north and south of China. Using panel 1, the majority of strains (84/105) were genotype 42 clustering to the 'East Mediterranean' <it>B. melitensis </it>group. Chinese <it>B. melitensis </it>are classified in limited number of closely related genotypes showing variation mainly at the panel 2B loci.</p> <p>Conclusion</p> <p>The MLVA-16 assay can be useful to reveal the predominant genotypes and strain relatedness in endemic or non-endemic regions of brucellosis. However it is not suitable for biovar differentiation of <it>B. melitensis</it>. Genotype 42 is widely distributed throughout China during a long time. Bruce 16 and bruce 30 in panel 2B markers are most useful for typing Chinese isolates.</p

    Isoflurane Preconditioning at Clinically Relevant Doses Induce Protective Effects of Heme Oxygenase-1 on Hepatic Ischemia Reperfusion in Rats

    <p>Abstract</p> <p>Background</p> <p>Activation of heme oxygenase-1 (HO-1) has been proved to reduce damages to the liver in ischemia reperfusion injury. The objective of present study was to determine whether clinic relevant doses of isoflurane treatment could be sufficient to activate HO-1 inducing, which confers protective effect against hepatic ischemia-reperfusion injury.</p> <p>Methods</p> <p>The hepatic artery and portal vein to the left and the median liver lobes of forty male Sprague-Dawley rats were occluded for 60 minutes. Reperfusion was allowed for 4 hours before the animal subjects were sacrificed. Six groups (n = 12) were included in the study. A negative control group received sham operation and positive control group a standard ischemia-reperfusion regimen. The third group was pretreated with isoflurane prior to the ischemia-reperfusion. The fourth group received an HO-1 inhibitor zinc protoporphyrin (Znpp) prior to the isoflurane pretreatment and the ischemia-reperfusion. The fifth group received Znpp alone before ischemia-reperfusion procedure, and the sixth group was administrated with a HO-1 inducer hemin prior to IR. HO-1 in the liver was measured using an enzymatic activity assay, a Western blot analysis, as well as immunohistochemical method. Extent of liver damage was estimated by determination of the serum transaminases, liver lipid peroxidation and hepatic histology. Infiltration of the liver by neutrophils was measured using a myeloperoxidase activity assay. TNFα mRNA in the liver was measured using RT-PCR.</p> <p>Results</p> <p>Isoflurane pretreatment significantly attenuated the hepatic injuries and inflammatory responses caused by the ischemia reperfusion. Selectively inhibiting HO-1 with ZnPP completed blocked the protective effects of isoflurane. Inducing HO-1 with hemin alone produced protective effects similar in magnitude to that of isoflurane.</p> <p>Conclusions</p> <p>Clinic relevant doses of isoflurane attenuate ischemia reperfusion injury in rats by increasing the HO-1 expression and activity.</p

    Annual (2023) taxonomic update of RNA-directed RNA polymerase-encoding negative-sense RNA viruses (realm Riboviria: kingdom Orthornavirae: phylum Negarnaviricota)

    55 PĂĄg.In April 2023, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by one new family, 14 new genera, and 140 new species. Two genera and 538 species were renamed. One species was moved, and four were abolished.     High-Order Semi-Lagrangian Schemes for the Transport Equation on Icosahedron Spherical Grids

    The transport process is an important part of the research of fluid dynamics, especially when it comes to tracer advection in the atmosphere or ocean dynamics. In this paper, a series of high-order semi-Lagrangian methods for the transport process on the sphere are considered. The methods are formulated entirely in three-dimensional Cartesian coordinates, thus avoiding any apparent artificial singularities associated with surface-based coordinate systems. The underlying idea of the semi-Lagrangian method is to find the value of the field/tracer at the departure point through interpolating the values of its surrounding grid points to the departure point. The implementation of the semi-Lagrangian method is divided into the following two main procedures: finding the departure point by integrating the characteristic equation backward and then interpolate on the departure point. In the first procedure, three methods are utilized to solve the characteristic equation for the locations of departure points, including the commonly used midpoint-rule method and two explicit high-order Runge&ndash;Kutta (RK) methods. In the second one, for interpolation, four new methods are presented, including (1) linear interpolation; (2) polynomial fitting based on the least square method; (3) global radial basis function stencils (RBFs), and (4) local RBFs. For the latter two interpolation methods, we find that it is crucial to select an optimal value for the shape parameter of the basis function. A Gauss hill advection case is used to compare and contrast the methods in terms of their accuracy, and conservation properties. In addition, the proposed method is applied to standard test cases, which include solid body rotation, shear deformation of twin slotted cylinders, and the evolution of a moving vortex. It demonstrates that the proposed method could simulate all test cases with reasonable accuracy and efficiency
