    Tyrosine and phenylalanine catabolism byLactobacillus cheese flavor adjuncts

    Bacterial metabolism of Tyr and Phe has been associated with the formation of aromatic compounds that impart barny-utensil and floral off-flavors in cheese. In an effort to identify possible mechanisms for the origin of these compounds in Cheddar cheese, we investigated Tyr and Phe catabolism by Lactobacillus casei and Lactobacillus helveticus cheese flavor adjuncts under simulated Cheddar cheese-ripening (pH 5.2, 4% NaCl, 15°C, no sugar) conditions. Enzyme assays of cell-free extracts indicated that L. casei strains catabolize Tyr and Phe by successive, constitutively expressed transamination and dehydrogenation reactions. Similar results were obtained with L. helveticus strains, except that the dehydrogenase enzymes were induced during incubation under cheese-ripening conditions. Micellar electrokinetic capillary chromatography of supernatants from L. casei and L. helveticus strains incubated under simulated cheese-ripening conditions confirmed that Tyr and Phe transamination and dehydrogenation pathways were active in both species and also showed these reactions were reversible. Major products of Tyr catabolism were p-hydroxy phenyl lactic acid and p-hydroxy phenyl acetic acid, while Phe degradation gave rise to phenyl lactic acid, phenyl acetic acid, and benzoic acid. However, some of these products were likely formed by nonenzymatic processes, since spontaneous chemical degradation of the Tyr intermediate p-hydroxy phenyl pyruvic acid produced p-hydroxy phenyl acetic acid, p-hydroxy phenyl propionic acid, and p-hydroxy benzaldehyde, while chemical degradation of the Phe intermediate phenyl pyruvic acid gave rise to phenyl acetic acid, benzoic acid, phenethanol, phenyl propionic acid, and benzaldehyde

    Microbial degradation of Trp is thought to promote the formation of aromatic compounds that impart putrid, fecal, or unclean flavors in cheese, but pathways for their production have not been established. This study investigated Trp catabolism by Lactobacillus casei and Lactobacillus helveticus cheese flavor adjuncts under carbohydrate starvation (pH 6.5, 30 or 37°C, no sugar) and near cheese-ripening (pH 5.2, 4% NaCl, 15°C, no sugar) conditions. Enzyme assays of cell-free extracts indicated that both species of Lactobacillus catabolized Trp to indole-3-lactic acid, and micellar electrokinetic capillary chromatography of culture supernatants showed this reaction occurred via successive transamination and dehydrogenation reactions. Tryptophan decarboxylase activity was also detected in all Lactobacillus cell-free extracts, but tryptamine was not detected in culture supernatants. Micellar electrokinetic capillary chromatography showed that Trp metabolism in Lactobacillus casei LC301 and LC202 was similar under both incubation conditions and that those catabolic reactions were reversible (i.e., conversion of indole-3-lactic acid to Trp). In contrast, Trp catabolism by Lactobacillus helveticus LH212 was only detected under near cheese ripening conditions. Cells of Lactobacillus helveticus CNRZ32 did not catabolize Trp in either condition but did convert indole-3-pyruvic acid to Trp in carbohydrate starvation medium and to Trp and indole-3-lactic acid under near cheese ripening conditions

    MAC Protocols for Wireless Mesh Networks with Multi-beam Antennas: A Survey

    Multi-beam antenna technologies have provided lots of promising solutions to many current challenges faced in wireless mesh networks. The antenna can establish several beamformings simultaneously and initiate concurrent transmissions or receptions using multiple beams, thereby increasing the overall throughput of the network transmission. Multi-beam antenna has the ability to increase the spatial reuse, extend the transmission range, improve the transmission reliability, as well as save the power consumption. Traditional Medium Access Control (MAC) protocols for wireless network largely relied on the IEEE 802.11 Distributed Coordination Function(DCF) mechanism, however, IEEE 802.11 DCF cannot take the advantages of these unique capabilities provided by multi-beam antennas. This paper surveys the MAC protocols for wireless mesh networks with multi-beam antennas. The paper first discusses some basic information in designing multi-beam antenna system and MAC protocols, and then presents the main challenges for the MAC protocols in wireless mesh networks compared with the traditional MAC protocols. A qualitative comparison of the existing MAC protocols is provided to highlight their novel features, which provides a reference for designing the new MAC protocols.     abd-A Regulation by the iab-8 Noncoding RNA

    The homeotic genes in Drosophila melanogaster are aligned on the chromosome in the order of the body segments that they affect. The genes affecting the more posterior segments repress the more anterior genes. This posterior dominance rule must be qualified in the case of abdominal-A (abd-A) repression by Abdominal-B (Abd-B). Animals lacking Abd-B show ectopic expression of abd-A in the epidermis of the eighth abdominal segment, but not in the central nervous system. Repression in these neuronal cells is accomplished by a 92 kb noncoding RNA. This “iab-8 RNA” produces a micro RNA to repress abd-A, but also has a second, redundant repression mechanism that acts only “in cis.” Transcriptional interference with the abd-A promoter is the most likely mechanism

    Initiator Elements Function to Determine the Activity State of BX-C Enhancers

    A >300 kb cis-regulatory region is required for the proper expression of the three bithorax complex (BX-C) homeotic genes. Based on genetic and transgenic analysis, a model has been proposed in which the numerous BX-C cis-regulatory elements are spatially restricted through the activation or repression of parasegment-specific chromatin domains. Particular early embryonic enhancers, called initiators, have been proposed to control this complex process. Here, in order to better understand the process of domain activation, we have undertaken a systematic in situ dissection of the iab-6 cis-regulatory domain using a new method, called InSIRT. Using this method, we create and genetically characterize mutations affecting iab-6 function, including mutations specifically modifying the iab-6 initiator. Through our mutagenesis of the iab-6 initiator, we provide strong evidence that initiators function not to directly control homeotic gene expression but rather as domain control centers to determine the activity state of the enhancers and silencers within a cis-regulatory domain

    Childhood lichen planus pemphigoides triggered by chickenpox

    Lichen planus pemphigoides (LPP) is a rare autoimmune bullous disorder that is rarer in children characterized by bullae on lichen planus like papules and the normal looking skin. Clinical, histopathological and direct immunoflourescence evaluation is important for the diagnosis of this entity. We report a case of LPP in a 5-year-old girl child probably triggered by an episode of preceding varicella. Diagnosis was confirmed by histopathology and immunofluorescence examination. Histological findings were typical of lichen planus from the biopsy taken from a violaceous to erythematous papule and characteristic of bullous pemphigoid in the biopsy taken from a bullous lesion with evidence of immunoglobulin G and C3 deposition along the basement membrane zone on direct immunofluorescence
