    Immobilization of Bacillus Subtilis E6-5 Protease and Commercial Protease in Nanofibrils Containing Different Amino Acids

    DergiPark: 633788trkjnatBu çalışmada, elektrospin yöntemiyle yüksek yüzey alanına sahip, glisin, tirozin ve glutamik asit aminoasitleri ile oluşturulmuş poliamid 6 polimer yüzeyler üretilmiş ve liyofilize Bacillus subtilis E6-5 proteaz ve ticari proteaz enzimleri nanofibriller üzerinde immobilize edilmiştir. Enzimlerin yeniden kullanılabilirliği araştırıldı. Enzimlerin immobilizasyon verimlilikleri yaklaşık olarak % 50-55 arasındaydı. Liyofilize Bacillus proteazı ile yapılan çalışmalarda glutaraldehitle aktifleştirilmiş PA6 nanolifler ve glutaraldehitle aktifleştirilmeyen PA6 nanoliflerde glutamik asit aminoasidi varlığında immobilizasyonun daha başarılı olduğu saptanmıştır. Glutaraldehit ile aktifleştirilmemiş ve aktifleştirilmiş yüzeylerde immobilize edilen liyofilize proteaz enziminin 4 kez kullanımı olmasına rağmen, en iyi işlevsel stabilite 2 kez kullanım ile elde edilmiştir. Saf PA6/glutamik asit nanoliflerinde iki tekrarlı kullanım sonucu enzimin immobilizasyon verimi % 38 olarak bulunmuştur. Glutaraldehitle aktifleştirilmiş PA6 nanoliflerde de PA6/glutamik asit nanolif yüzeyleri iki tekrarlı kullanım sonucu enzimin immobilizasyon verimi % 65 olarak bulunmuştur. Nanoliflerin glutaraldehitle aktifleştirmesi sonucu enzim immobilizasyon verimi iki kat arttırılmıştır. Ticari proteaz ile yapılan çalışmalarda ise glutaraldehitle aktifleştirilmemiş nanolif yüzeylerde enzimin 6 kez kullanımı olmasına rağmen en işlevsel stabilite 3 tekrarlı kullanımda elde edilmiştir. En başarılı immobilizasyon verimi PA6 nanoliflerde % 58 olarak bulunmuştur. Glutaraldehitle aktifleştirilmiş PA6 nanoliflerde de enzim 6 kez kullanım bulmuş fakat işlevsel stabilite 4 tekrarlı kullanıma kadar korunmuştur.In this study, polyamide 6 polymer surfaces that have a high surface area were produced by electrospinning method with the participation of Glycine, Tyrosine and Glutamic acid amino acids, and lyophilized Bacillus subtilis E6-5 protease and commercial protease enzymes were immobilized on nanofibrils. Enzyme reusability were investigated. The immobilization efficiencies of the enzymes were approximately between 50-55 %. In studies with lyophilized Bacillus protease, glutaraldehyde activated PA6 nanofibrils and glutaraldehyde unactivated PA6 nanofibrils were found to be more immobilized in the presence of Glutamic acid. Although the lyophilized protease enzyme immobilized on non-glutaraldehyde activated and activated surfaces has been used 4 times, the best functional stability has been achieved with 2 times use. In pure PA6/Glutamic acid nanofibrils, the immobilization yield of the two times used enzymes was found to be 38 %. In glutaraldehyde-activated PA6 nanofibrils, the PA6/Glutamic acid nanofibril surfaces were found to have 65 % immobilization yield of the two repetitive used enzymes. The enzyme immobilization efficiency has been doubled by glutaraldehyde activation of the nanofibrils. In studies with commercial protease, the most functional stability was obtained for 3 repeated uses, although the enzyme was used 6 times on the non-glutaraldehyde activated nanofibril surfaces. The most successful immobilization was found in 58 % of PA6 nanofibrils. In glutaraldehyde-activated PA6 nanofibrils, the enzyme was found to be used 6 times, but the functional stability was maintained as much as 4 times of repeated use

    Pre-operative Serum CEA and CA19-9 Levels in Patients with Gastric Cancer: A Single-Center Experience

    Introduction: To evaluate the preoperative CEA and CA19-9 levels and their association with clinicopathologic features and mortality. Methods: This retrospective study was conducted on patients who underwent gastric surgery due to gastric adenocarcinoma from 2008 to 2015. Preoperative serum CEA and CA19-9 levels were compared according to the TNM classification and survival data were compared. Results: A total of 335 patients were included in the study with the mean age of 66.1±11.0 years. CEA and CA19-0 positivity were 29.1% and 28.1%, respectively. According to the CEA groups, T stages of the cases were statistically significant (p = 0.013). The rates of T stage 1 in group 1 (CEA positive) were significantly higher than the group 2 (CEA negative) (p=0.007). According to the CA19-9 positivity; rate of stage 2 was significantly higher in the group 1 (CA19-9 positive) (p=0.001); however, rates of stage 2 and 3 were significantly higher in the group 2 (CA19-9 negative) (p=0.004 and p=0.007, respectively). Mortality information could be accessed for 309 cases and 108 deaths (36%) were observed. The mean survival time was 45.21±2.42 months. No significant difference was observed between the groups (p>0.05). Conclusion: Patients with higher levels of CEA and CA19-9 seem to have higher grades of gastric cancer. However, CEA and CA19-9 level does not seem to be in association in our study population

    A novel modified PAIR technique using a trocar catheter for percutaneous treatment of liver hydatid cysts: a six-year experience

    PURPOSEWe aimed to demonstrate the success and reliability of a novel puncture, aspiration, injection, and reaspiration (PAIR) technique in liver hydatid cysts.METHODSPercutaneous treatment with ultrasonographic guidance was performed in 493 hepatic hydatid cysts in 374 patients. Patients were treated with a new PAIR technique by single puncture method using a 6F trocar catheter. The results of this novel technique were evaluated with regards to efficacy and safety of the procedure and complication rates.RESULTSOut of 493 cysts, 317 were Gharbi type I (WHO CE 1) and 176 were Gharbi type II (WHO CE 3A). Of all cysts, 13 were referred to surgery because of cystobiliary fistulization. Recurrence was observed in 11 cysts one month later. Therefore, the success rate of the PAIR technique was 97.7% (469/480). Minor complications (fever, urticaria-like reactions, biliary fistula) were seen in 44 treated patients (12%, 44/374); the only major complication was reversible anaphylactic shock which was observed in two patients (0.5%, 2/374).CONCLUSIONThis novel modified PAIR technique may be superior to catheterization by Seldinger technique due to its efficiency, easier application, lower severe complication rate, and lower cost. Further comparative studies are required to confirm our observations

    Determination of the Young Modulus of Ti-TiAl3 Metallic Intermetallic Laminate Composites by Nano-Indentation

    Nano-indentation is an important technique to determine the Young modulus of multiphase materials where normal tensile tests are not appropriate. In this work, Ti-TiAl3 metallic-intermetallic laminate composites have been fabricated successfully in open atmosphere using commercial purity Al and Ti foils with 250 mu m and 500 mu m initial thicknesses, respectively. Sintering process was performed at 700 degrees C under 2 MPa pressure for 7.5 h. Mechanical properties including the Young modulus were determined after manufacturing. The Young moduli of metallic and intermetallic phases were determined as 89 GPa and 140 GPa, respectively. Microstructure analyses showed that aluminum foil was almost consumed by forming a titanium aluminide intermetallic compound. Titanium aluminides grow up through spherical shaped islands and metallic-intermetallic interface is a wavy form in Ti-Al system. Thus, the final microstructure consists of alternating layers of intermetallic compound and unreacted Ti metal. Microstructure and phase characterizations were performed by scanning electron microscopy, energy dispersive spectroscopy, and X-ray diffraction. Hardness of test samples was determined as 600 HV for intermetallic zone and 130 HV for metallic zone by the Vickers indentation method

    Quality of life in Turkish patients with Familial Mediterranean Fever: Association with fatigue, psychological status, disease severity and other clinical parameters

    Aim of the work: To evaluate the quality of life (QoL) in Familial Mediterranean Fever (FMF) patients, and determine its association with fatigue, depression, disease severity and other clinical parameters. Patients and methods: Sixty FMF patients were included. QoL was assessed by Short Form-36 (SF-36), depression by Hamilton Depression Scale (HDS), and fatigue by Fatigue severity scale (FSS). Disease severity score and Mutations of the Mediterranean fever (MEFV) gene were assessed. Results: The mean age of patients was 33.73 ± 9.81 years and disease duration 14.6 ± 12.1 years. They were 35 females and 25 males. FMF patients scored significantly higher in FSS (29.9 ± 17.6) and HDS (15.1 ± 8.5) compared to the control (10.6 ± 7.1 and 6.3 ± 9.4; p < 0.0001 respectively) while all SF36 sub-items except mental health were significantly lower (p < 0.05). MEFV gene mutation was present in 49 (81.7%) patients. The visual analogue scale of pain significantly negatively correlated with the FSS (p < 0.0001), HDS (p < 0.0001) and all SF36 sub-items except mental health (p < 0.0001). Disease duration, age of onset, and duration of attacks showed no significant correlation with FSS, HDS and SF36. Delay in diagnosis significantly correlated with FSS (p < 0.0001) and negatively with SF36 sub-items physical role (p = 0.02), general health (p = 0.01) and social functioning (p = 0.03). Age of diagnosis significantly correlated with FSS (p = 0.03) and negatively with SF36-vitality (p = 0.047). There was a significant effect of disease severity on QoL, fatigue and depression (p < 0.05). Conclusion: QoL is associated with fatigue, depression and disease severity in FMF patients.     A study on antioxidant capacities and fatty acid compositions of two daphne species from Turkey: New sources of antioxidants and essential fatty acids

    WOS: 000328062800002Daphne (Thymealaceae) species have been widely used in traditional Anatolian medicine. The antioxidant capacity and fatty acid composition of two Daphne taxa (D.oleoides subsp. oleoides and D.serica) were studied. The antioxidant properties of four solvent extracts of Daphne species were assessed using different chemical assays including phosphomolybdate assay, free radical-scavenging assay, -carotene/linoleic acid test system, ferric and cupric reducing power. The total phenolic and flavonoid content of the extracts also were determined. Ethyl acetate, methanol and ethanol extracts obtained from the Daphne species possess strong antioxidant potential. Polyunsaturated fatty acids in the studied oils were the highest level in the fatty acid components. Essential fatty acids accounted for 27.46-45.31% of total fatty acids. These results indicated that the extracts of studied Daphne taxa can be exploited as a source of natural antioxidants and essential fatty acids in pharmacological and food industry

    Craniosynostosis is the premature closure of the cranial sutures. We present the radiological findings of a six-month old case who presented with small head and frontal triangular shape and who was found to have generalized pitting and softness on palpation of the head. The three-dimensional images computed tomography (CT) showed a sagittal, coronal and metopic suture synostosis with exaggerated convolutional markings in the form of lacunar skull.
