    The forking effect

    This study introduces the concept of the forking effect in the cryptocurrency market,specifically focusing on the impact of forking events on bitcoin, also called parent coin.We use a modified exponential GARCH model to examine the bitcoin's response inreturns and volatility. Our findings reveal that forking events do not significantlyaffect the bitcoin's returns but have a strong positive impact on its volatility, especially when considering market dynamics. Our model accounts for key features likevolatility clustering and fat-tailed distributions. Additionally, we observe that following a fork event, volatility remains elevated for the next three days, regardless ofother forking events, and the volatility impact does not increase when multiple forksoccur simultaneously on the same day

    Toward coherent space–time mapping of seagrass cover from satellite data: an example of a Mediterranean lagoon

    Seagrass meadows are a highly productive and economically important shallow coastal habitat. Their sensitivity to natural and anthropogenic disturbances, combined with their importance for local biodiversity, carbon stocks, and sediment dynamics, motivate a frequent monitoring of their distribution. However, generating time series of seagrass cover from field observations is costly, and mapping methods based on remote sensing require restrictive conditions on seabed visibility, limiting the frequency of observations. In this contribution, we examine the effect of accounting for environmental factors, such as the bathymetry and median grain size (D50) of the substrate as well as the coordinates of known seagrass patches, on the performance of a random forest (RF) classifier used to determine seagrass cover. Using 148 Landsat images of the Venice Lagoon (Italy) between 1999 and 2020, we trained an RF classifier with only spectral features from Landsat images and seagrass surveys from 2002 and 2017. Then, by adding the features above and applying a time-based correction to predictions, we created multiple RF models with different feature combinations. We tested the quality of the resulting seagrass cover predictions from each model against field surveys, showing that bathymetry, D50, and coordinates of known patches exert an influence that is dependent on the training Landsat image and seagrass survey chosen. In models trained on a survey from 2017, where using only spectral features causes predictions to overestimate seagrass surface area, no significant change in model performance was observed. Conversely, in models trained on a survey from 2002, the addition of the out-of-image features and particularly coordinates of known vegetated patches greatly improves the predictive capacity of the model, while still allowing the detection of seagrass beds absent in the reference field survey. Applying a time-based correction eliminates small temporal variations in predictions, improving predictions that performed well before correction. We conclude that accounting for the coordinates of known seagrass patches, together with applying a time-based correction, has the most potential to produce reliable frequent predictions of seagrass cover. While this case study alone is insufficient to explain how geographic location information influences the classification process, we suggest that it is linked to the inherent spatial auto-correlation of seagrass meadow distribution. In the interest of improving remote-sensing classification and particularly to develop our capacity to map vegetation across time, we identify this phenomenon as warranting further research.</p

    Transcranial direct current stimulation (a-tCDS) after subacromial injections in patients with subacromial pain syndrome: a randomized controlled pilot study

    Background: Subacromial pain syndrome (SAPS) is a common complaint in orthopaedics. Subacromial corticosteroid injections (CSI) can relieve pain in the short term. Anodal transcranial direct current stimulation (a-tDCS) has been used for symptomatic pain relief in a variety of chronic pain conditions. The aim of this pilot study was to assess whether the application a-tDCS could enhance the symptomatic relief provided by CSI in patients affected by SAPS. Methods: Thirty-eight participants (18 to 65-year-old) suffering from SAPS were recruited to have a CSI and randomly allocated to receive, 1 weeks post CSI, real a-tDCS (r-tDCS), sham tDCS (s-tDCS) or no intervention (Control). Upper limb function was measured 1 week prior to the CSI, at the 2- and 4-week follow-ups using self-administered questionnaires and physical measures. Self-reported pain and activity during each day were logged by the participants using visual analog scales (VAS). Differences between groups were tested using repeated-measures ANOVAs. Results: Pain VAS and the Single Assessment Numeric Evaluation scale (SANE) showed significant improvement from baseline 2 weeks and 4 weeks after CSI in all groups (p < 0.05). There were no significant group X time interaction 2 weeks following tDCS treatment in any of the variables. Conclusion: All groups showed significant improvement in pain VAS and SANE scores following the CSI. One session of a-tDCS treatment 2 weeks following CSI did not result in any additive or potentializing effects when compared to a s-tDCS or a control group

    Cross-cultural translation and psychometric validation of the French version of the Fear- Avoidance Components Scale (FACS)

    Abstract: Background The Fear-Avoidance Components Scale (FACS) is a reliable and valid instrument widely used to assess fear-avoidance beliefs related to pain and disability. However, there is a scarcity of validated translations of the FACS in different cultural and linguistic contexts, including the French population. This study aimed to translate and validate the French ver- sion of the FACS (FACS-Fr/CF), examining its psychometric properties among French- speaking individuals. Methods A cross-cultural translation process–including forward translation, backward translation, expert committee review, and pre-testing–was conducted to develop the FACS-Fr/CF. The translated version was administered to a sample of French-speaking adults (n = 55) with chronic musculoskeletal pain. Internal consistency (including confirmatory analyses of the 2 factors identified in the Serbian version), test-retest reliability and convergent validity were then assessed. Results The FACS-Fr/CF demonstrated high global internal consistency (α = 0.94, 95% CI: 0.91– 0.96) as well as high internal consistency of the 2 factors identified in the Serbian version (α = 0.90, 95% CI: 0.86–0.94 and α = 0.90, 95% CI: 0.85–0.94, respectively). Test-retest analy- sis revealed a moderate (close to high) reliability (ICC = 0.89; 95% CI: 0.82–0.94 and r = 0.89; p<0.005). Convergent validity was supported by significant correlations between the FACS-Fr/CF scores and the Tampa Scale for Kinesiophobia (r = 0.82; p < 0.005), the Pain Catastrophizing Scale (r = 0.72; p < 0.005) and the Hospital Anxiety and Depression Scale (r = 0.66; p < 0.005). Conclusion The present study provides evidence for the cross-cultural translation and psychometric val- idation of the FACS-Fr/CF. The FACS-Fr/CF exhibits a high internal consistency, a moder- ate (close to high) test-retest reliability, and good construct validity, suggesting its utility in assessing fear-avoidance beliefs in the French-speaking population. This validated tool can enhance the assessment and understanding of fear-avoidance behaviors and facilitate cross-cultural research in pain-related studies

    Correlative Scanning-Transmission Electron Microscopy Reveals that a Chimeric Flavivirus Is Released as Individual Particles in Secretory Vesicles.

    International audienceThe intracellular morphogenesis of flaviviruses has been well described, but flavivirus release from the host cell remains poorly documented. We took advantage of the optimized production of an attenuated chimeric yellow fever/dengue virus for vaccine purposes to study this phenomenon by microscopic approaches. Scanning electron microscopy (SEM) showed the release of numerous viral particles at the cell surface through a short-lived process. For transmission electron microscopy (TEM) studies of the intracellular ultrastructure of the small number of cells releasing viral particles at a given time, we developed a new correlative microscopy method: CSEMTEM (for correlative scanning electron microscopy - transmission electron microscopy). CSEMTEM analysis suggested that chimeric flavivirus particles were released as individual particles, in small exocytosis vesicles, via a regulated secretory pathway. Our morphological findings provide new insight into interactions between flaviviruses and cells and demonstrate that CSEMTEM is a useful new method, complementary to SEM observations of biological events by intracellular TEM investigations

    Bone Marrow Mesenchymal Stem Cells Enhance the Differentiation of Human Switched Memory B Lymphocytes into Plasma Cells in Serum-Free Medium

    The differentiation of human B lymphocytes into plasma cells is one of the most stirring questions with regard to adaptive immunity. However, the terminal differentiation and survival of plasma cells are still topics with much to be discovered, especially when targeting switched memory B lymphocytes. Plasma cells can migrate to the bone marrow in response to a CXCL12 gradient and survive for several years while secreting antibodies. In this study, we aimed to get closer to niches favoring plasma cell survival. We tested low oxygen concentrations and coculture with mesenchymal stem cells (MSC) from human bone marrow. Besides, all cultures were performed using an animal protein-free medium. Overall, our model enables the generation of high proportions of CD38+CD138+CD31+ plasma cells (≥50%) when CD40-activated switched memory B lymphocytes were cultured in direct contact with mesenchymal stem cells. In these cultures, the secretion of CXCL12 and TGF-β, usually found in the bone marrow, was linked to the presence of MSC. The level of oxygen appeared less impactful than the contact with MSC. This study shows for the first time that expanded switched memory B lymphocytes can be differentiated into plasma cells using exclusively a serum-free medium

    Interoperable Semantic & Syntactic Service Matching for Ambient Computing Environments

    International audienceThe inherent heterogeneity of ambient computing environments and their constant evolution requires middleware platforms to manage networked components designed, developed and deployed independently. Such management must also be efficient to cater for resource-constrained devices and highly dynamic situations due to the spontaneous appearance and disappearance of networked resources. For service discovery protocols (SDP), one of the main functions of service-oriented architectures (SOA), the efficiency of the matching of syntactic service descriptions is most often opposed to the fullness of the semantic approach. As part of the PLASTIC middleware for ambient computing environments, we present in this paper an interoperable service matching and ranking platform, which is able to process service descriptions from both semantic and syntactic service description languages. To that end, we define a generic, modular description language able to record service functional properties, potentially extended with semantic annotations. An evaluation of the prototype implementation of our platform demonstrates that multi-protocols service matching supporting various levels of expressiveness can be achieved in ambient computing environments

    B lymphocyte differentiation into long-lived plasma cells is the keystone event for the production of long-term protective antibodies. CD40-CD154 and CD27-CD70 interactions are involved in human B lymphocyte differentiation into CD38hiCD138+ cells in vivo as well as in vitro. In this study, we have compared these interactions in their capacity to drive switched-memory B lymphocytes differentiation into CD38hiCD138+ plasma cells. The targeted B lymphocytes were isolated from human peripheral blood, expanded for 19 days, and then submitted to CD70 or CD154 interactions for 14 days. The expanded B lymphocytes were constitutively expressing CD39, whereas CD31’s expression was noticed only following the in vitro differentiation step (day 5) and was exclusively present on the CD38hi cell population. Furthermore, the generated CD38hiCD138+ cells showed a higher proportion of CD31+ cells than the CD38hiCD138- cells. Besides, analyses done with human blood and bone marrow plasma cells showed that in vivo and de novo generated CD38hiCD138+ cells have a similar CD31 expression profile but are distinct according to their reduced CD39 expression level. Overall, we have evidences that in vitro generated plasma cells are heterogeneous and appear as CD39+ precursors to the ones present in bone marrow niches
