    Synthesis of fucose derivatives with thiol motifs towards suicide inhibition of helicobacter pylori

    The syntheses of six thiol-exhibiting monosaccharides towards suicide inhibition of Helicobacter pylori are reported. Blood group Antigen Binding Adhesin (BabA), a bacterial membrane-bound lectin, binds to human ABO and Lewis b blood group structures displayed on the surface of host epithelial cells. Crystal structures of the carbohydrate-recognition domain revealed a conserved disulfide bonded loop that anchors a critical fucose residue in these blood group structures. Disruption of this loop by N-acetylcysteine results in reduced BabA-mediated adherence to human gastric tissue sections and attenuated virulence in Lewis b-expressing transgenic mice. With a view of creating specific inhibitors of the lectin, we designed and successfully synthesised six fucose-derived compounds with thiol motifs to engage in a thiol-disulfide exchange with this disulfide bond of BabA and form a glycan-lectin disulfide linkage. Branching and extending the fucose backbone with 2- and 3-carbon thiol motifs delivered a range of candidates to be tested for biological activity against BabA

    Automated data pre-processing via meta-learning

    The final publication is available at link.springer.comA data mining algorithm may perform differently on datasets with different characteristics, e.g., it might perform better on a dataset with continuous attributes rather than with categorical attributes, or the other way around. As a matter of fact, a dataset usually needs to be pre-processed. Taking into account all the possible pre-processing operators, there exists a staggeringly large number of alternatives and nonexperienced users become overwhelmed. We show that this problem can be addressed by an automated approach, leveraging ideas from metalearning. Specifically, we consider a wide range of data pre-processing techniques and a set of data mining algorithms. For each data mining algorithm and selected dataset, we are able to predict the transformations that improve the result of the algorithm on the respective dataset. Our approach will help non-expert users to more effectively identify the transformations appropriate to their applications, and hence to achieve improved results.Peer ReviewedPostprint (published version

    Frozen water waves over rough topographical bottoms

    The propagation of surface water waves over rough topographical bottoms is investigated by the multiple scattering theory. It is shown that the waves can be localized spatially through the process of multiple scattering and wave interference, a peculiar wave phenomenon which has been previously discussed for frozen light in optical systems (S. John, Nature {\bf 390}, 661, (1997)). We demonstrate that when frozen, the transmission of the waves falls off exponentially, and a cooperative behavior appears, fully supporting previous predictions. A phase diagram method is used to illustrate this distinct phase states in the wave propagation.Comment: 4 pages and 5 figure

    Gravity waves over topographical bottoms: Comparison with the experiment

    In this paper, the propagation of water surface waves over one-dimensional periodic and random bottoms is investigated by the transfer matrix method. For the periodic bottoms, the band structure is calculated, and the results are compared to the transmission results. When the bottoms are randomized, the Anderson localization phenomenon is observed. The theory has been applied to an existing experiment (Belzons, et al., J. Fluid Mech. {\bf 186}, 530 (1988)).     Immunotherapy is an emerging therapeutic strategy with a promising clinical outcome in some solid tumors, particularly metastatic melanoma. One approach to immunotherapy is immune checkpoint inhibitors, such as blockage of CTLA-4 and PD-1/PD-L1. This special report aims to describe the state of clinical trials of tremelimumab in patients with unresectable malignant mesothelioma (MM) in particular with regard to the clinical efficacy, safety and tolerability. Criticism and perspective of this treatment are also discussed. Biological and clinical considerations rule out the use of tremelimumab as single agent for MM and, more generally, the use of immune checkpoint inhibitors for MM is still largely questionable and not supported by evidences

    Adaptive filtering for removing nonstationary physiological noise from resting state fMRI BOLD signals

    fMRI is used to investigate brain functional connectivity after removing nonneural components by General Linear Model (GLM) approach with a reference ventricle-derived signal as covariate. Ventricle signals are related to low-frequency modulations of cardiac and respiratory rhythms, which are nonstationary activities. Herein, we employed an adaptive filtering approach to improve removing physiological noise from BOLD signals. Comparisons between filtering approaches were performed by evaluating the amount of removed signal variance and the connectivity between homologous contralateral regions of interest (ROIs). The global connectivity between ROIs was estimated with a generalized correlation named RV coefficient. The mean ROI decrease of variance was -52% and -11%, for adaptive filtering and GLM, respectively. Adaptive filtering led to higher connectivity between grey matter ROIs than that obtained with GLM. Thus, adaptive filtering is a feasible method for removing the physiological noise in the low frequency band and to highlight resting state functional networks

    Prolate and temperature-responsive self-assemblies of amphiphilic random copolymers with perfluoroalkyl and polyoxyethylene side chains in solution

    Two amphiphilic random copolymers, PEGMAx-co-FAy (x = 90 and 70 mol%), were synthesized by ATRP and their solutions were investigated as a function of solvent, concentration and temperature by DLS and SANS analyses. Both copolymers self-assembled in nanostructures by single-chain folding in water solutions over a wide range of temperatures. The values of the DLS hydrodynamic radius and the SANS radius of gyration were found to be ~4 nm and ~3.4–3.7 nm, respectively. Moreover, SANS showed the self-folded nanoassemblies to be prolated spheroids with ratio of polar/equatorial axes ~5:1 for PEGMA90-co-FA10 and ~2:1 for PEGMA70-co-FA30. On heating above a critical temperature Tc, multi-chain microassemblies were formed that reverted back to nanoassemblies on cooling below Tc. This temperature-responsive transition was fully and sharply reversible
