    SENSORY AND HEALTH-RELATED FRUIT QUALITY OF ORGANIC APPLES. A COMPARATIVE FIELD STUDY OVER THREE YEARS USING CONVENTIONAL AND HOLISTIC METHODS TO ASSESS FRUIT QUALITY

    In a 3 years lasting field study with the ‘Golden Delicious’ cultivar, we compared fruits of 5 pairs of organic/integrated fruit farms. The orchards were similar in microclimate, soil conditions and planting system. To assess inner fruit quality we investigated at the beginning and at the end of cold storage: (i) standard parameters (firmness, sugar, malic acid., mineral content); (ii) sensorial quality by panel tests; (iii) health related components (23 phenolic compounds, nutritional fibres; vitamins); and (v) fruit «vitality quality» by holistic approaches (crystallisation in copper chloride, self degradation tests, feeding preference tests with laboratory rats). The most significant differences were found in year one of the study, and were by tendency confirmed in the following two years. In year one all fruit samples of organic orchards had significantly firmer fruit flesh (14%), a 10 % higher index of inner quality (on basis of sugar and malic acid content and fruit flesh firmness), and 15% higher taste scores than conventional ones. Phosphorus content of the fruit flesh was 31% higher in organic apples and closely correlated (r2 = 0.93) with the index of inner quality and sensory score (r2 = 0.69). No extraction method of Phosphorus in the soil (water, NH4-EDTA, citric acid, CAL) correlated with the P-content in the fruits. However, P in the fruit flesh correlated by r2 = 0.72 with the microbial activity of the soil expressed as the ratio of microbialbound Nitrogen and Carbon in the soil. With a value of 3.85 the Cmic:Nmic ratio was 44.5 % lower (thus more favourable) in organic tree strips. Flavanols, with 65.7 % of the total polyphenol content were the dominant group of polyphenols. The content of flavonols was 22.7 % higher in organic apples in the first year and 15.6 % in the average of the three years. The self-degradation test didn't provide significant differences. Laboratory rats, showed a tendency to prefer IP apples probably due to their advanced ripeness. Thus rat behaviour did not correspond with the sensory panel judgement. The picture forming method provided a correct reproducibility with repeated blind samples before and after storage (r2 = 0.83), and distinguished 100 % correctly organic an IP fruit in the first year. In the second year there was one miss qualification. The average value over three dates of the index for «vitality quality», which was especially created for this study, was 44.6 % higher with organic apples The picture forming methods correlated well with sensory scores and standard quality (r2 = 0.63) in the first year. The study revealed interesting and consumer-relevant differences between organic and integrated apples with standard and holistic methods

    Genotoxicity of 2-amino-3-methylimidazo(4, 5-f)quinoline (IQ) and related compounds in Drosophila

    The potent food mutagen and carcinogen 2-amino-3-methyl-imidazo[4, 5-f]quinoline (IQ) and the structurally related heterocyclic aromatic amines 2-aminoimidazo[4, 5-f]quinoline (demethyl-IQ) and 2-amino-l-methylinudazo(4, 5-f)quinoline (iso-IQ) were assayed for genotoxicity in the wing somatic mutation and recombination test (SMART) as well as in the sex-linked recessive lethal (SLRL) test in Drosophila melanogaster. In addition, 3-methyl-2-nitroimidazo[4, 5-f]-quinoline (nitro-IQ), 2-nitrofluorene and 1, 8-dinitropyrene were also assayed in the wing spot test. IQ was clearly muta-genic in the SLRL test with highest activity in spermatids. Iso-IQ was more active than IQ whereas demethyl-IQ was inactive in this test. The same pattern of results was obtained in the wing SMART: iso-IQ produced >2-fold higher frequencies of spots than IQ and demethyl-IQ was clearly negative. In addition, nitro-IQ exhibited an approximately equal genotoxic activity as IQ. 2-Nitrofluorene and 1, 8-di-nitropyrene were both inactive in the wing spot test. These data provide good evidence for a correlation of genotoxic effects in germinal and somatic cells, and for the practical advantage of the wing spot test in Drosophila. Moreover, the results show structure-activity relationships among the heterocyclic aromatic amines and nitro compounds similar to those found in Salmonell

    Regulated overexpression of the survival factor bcl-2 in CHO cells increases viable cell density in batch culture and decreases DNA release in extended fixed-bed cultivation

    Using multicistronic expression technology we generated a stable Chinese hamster ovary (CHO) cell line (MG12) expressing a model secreted heterologous glycoprotein, the secreted form of the human placental alkaline phosphatase (SEAP), and bcl-2, best known as an apoptosis inhibitor, in a tetracycline-repressible dicistronic configuration. In batch cultivations in serum-containing medium, MG12 cells reached twice the final viable cell density when Bcl-2 was overexpressed (in the absence oftetracycline) compared to MG12 populations culturedunder tetracycline-containing conditions (bcl-2repressed). However, bcl-2-expressing MG12 cellsshowed no significant retardation of the decline phasecompared to batch cultures in which the dicistronicexpression unit was repressed.Genetic linkage of bcl-2 expression with the reporter protein SEAP in our multicistronic construct allowed online monitoring of Bcl-2 expression over an extended, multistage fixed-bed bioreactor cultivation. The cloned multicistronic expression unit proved to be stable over a 100 day bioreactor run. CHO MG12 cells in the fixed-bed reactor showed a drastic decrease in the release of DNA into the culture supernatant under conditions of reduced tetracycline (and hencederepressed SEAP and bcl-2 overexpression). This observation indicated enhanced robustness associated with bcl-2 overexpression, similar to recent findings for constitutive Bcl-2-overexpressing hybridoma cells under the same bioprocess conditions.
