The \u27Pseudomonas aeruginosa\u27 PSL Polysaccharide Is a Social but Noncheatable Trait in Biofilms
Extracellular polysaccharides are compounds secreted by microorganisms into the surrounding environment, and they are important for surface attachment and maintaining structural integrity within biofilms. The social nature of many extracellular polysaccharides remains unclear, and it has been suggested that they could function as either cooperative public goods or as traits that provide a competitive advantage. Here, we empirically tested the cooperative nature of the PSL polysaccharide, which is crucial for the formation of biofilms in Pseudomonas aeruginosa. We show that (i) PSL is not metabolically costly to produce; (ii) PSL provides populationlevel benefits in biofilms, for both growth and antibiotic tolerance; (iii) the benefits of PSL production are social and are shared with other cells; (iv) the benefits of PSL production appear to be preferentially directed toward cells which produce PSL; (v) cells which do not produce PSL are unable to successfully exploit cells which produce PSL. Taken together, this suggests that PSL is a social but relatively nonexploitable trait and that growth within biofilms selects for PSL-producing strains, even when multiple strains are on a patch (low relatedness at the patch level)
SPARTAN: a global network to evaluate and enhance satellite-based estimates of ground-level particulate matter for global health applications
Ground-based observations have insufficient spatial coverage to assess long-term human exposure to fine particulate matter (PM2.5) at the global scale. Satellite remote sensing offers a promising approach to provide information on both short-and long-term exposure to PM2.5 at local-to-global scales, but there are limitations and outstanding questions about the accuracy and precision with which ground-level aerosol mass concentrations can be inferred from satellite remote sensing alone. A key source of uncertainty is the global distribution of the relationship between annual average PM2.5 and discontinuous satellite observations of columnar aerosol optical depth (AOD). We have initiated a global network of ground-level monitoring stations designed to evaluate and enhance satellite remote sensing estimates for application in health-effects research and risk assessment. This Surface PARTiculate mAtter Network (SPARTAN) includes a global federation of ground-level monitors of hourly PM2.5 situated primarily in highly populated regions and collocated with existing ground-based sun photometers that measure AOD. The instruments, a three-wavelength nephelometer and impaction filter sampler for both PM2.5 and PM10, are highly autonomous. Hourly PM2.5 concentrations are inferred from the combination of weighed filters and nephelometer data. Data from existing networks were used to develop and evaluate network sampling characteristics. SPARTAN filters are analyzed for mass, black carbon, water-soluble ions, and metals. These measurements provide, in a variety of regions around the world, the key data required to evaluate and enhance satellite-based PM2.5 estimates used for assessing the health effects of aerosols. Mean PM2.5 concentrations across sites vary by more than 1 order of magnitude. Our initial measurements indicate that the ratio of AOD to ground-level PM2.5 is driven temporally and spatially by the vertical profile in aerosol scattering. Impact of Weddell Sea deep convection on natural and anthropogenic carbon in a climate model
A climate model is used to investigate the influence of Weddell Sea open ocean deep convection on anthropogenic and natural carbon uptake for the period 1860-2100. In a three-member ensemble climate change simulation, convection ceases on average by year 1981, weakening the net oceanic cumulative uptake of atmospheric CO2 by year 2100 (-4.3 Pg C) relative to an ocean that has continued convection. This net weakening results from a decrease in anthropogenic carbon uptake (-10.1 Pg C), partly offset by an increase in natural carbon storage (+5.8 Pg C). Despite representing only 4% of its area, the Weddell Sea is responsible for 22% of the Southern Ocean decrease in total climate-driven carbon uptake and 52% of the decrease in the anthropogenic component of oceanic uptake. Although this is a model-specific result, it illustrates the potential of deep convection to produce an inter-model spread in future projections of ocean carbon uptake
Brain health: the importance of recognizing cognitive impairment: an IAGG consensus conference
Cognitive impairment creates significant challenges for patients, their families and friends, and clinicians who provide their health care. Early recognition allows for diagnosis and appropriate treatment, education, psychosocial support, and engagement in shared decision-making regarding life planning, health care, involvement in research, and financial matters. An IAGG-GARN consensus panel examined the importance of early recognition of impaired cognitive health. Their major conclusion was that case-finding by physicians and health professionals is an important step toward enhancing brain health for aging populations throughout the world. This conclusion is in keeping with the position of the United States' Centers for Medicare and Medicaid Services that reimburses for detection of cognitive impairment as part the of Medicare Annual Wellness Visit and with the international call for early detection of cognitive impairment as a patient's right. The panel agreed on the following specific findings: (1) validated screening tests are available that take 3 to 7 minutes to administer; (2) a combination of patient- and informant-based screens is the most appropriate approach for identifying early cognitive impairment; (3) early cognitive impairment may have treatable components; and (4) emerging data support a combination of medical and lifestyle interventions as a potential way to delay or reduce cognitive decline
Repeated measures of inflammation, blood pressure, and heart rate variability associated with traffic exposures in healthy adults
Abstract Background Previous human exposure studies of traffic-related air pollutants have demonstrated adverse health effects in human populations by comparing areas of high and low traffic, but few studies have utilized microenvironmental monitoring of pollutants at multiple traffic locations while looking at a vast array of health endpoints in the same population. We evaluated inflammatory markers, heart rate variability (HRV), blood pressure, exhaled nitric oxide, and lung function in healthy participants after exposures to varying mixtures of traffic pollutants. Methods A repeated-measures, crossover study design was used in which 23 healthy, non-smoking adults had clinical cardiopulmonary and systemic inflammatory measurements taken prior to, immediately after, and 24 hours after intermittent walking for two hours in the summer months along three diverse roadways having unique emission characteristics. Measurements of PM2.5, PM10, black carbon (BC), elemental carbon (EC), and organic carbon (OC) were collected. Mixed effect models were used to assess changes in health effects associated with these specific pollutant classes. Results Minimal associations were observed with lung function measurements and the pollutants measured. Small decreases in BP measurements and rMSSD, and increases in IL-1β and the low frequency to high frequency ratio measured in HRV, were observed with increasing concentrations of PM2.5 EC. Conclusions Small, acute changes in cardiovascular and inflammation-related effects of microenvironmental exposures to traffic-related air pollution were observed in a group of healthy young adults. A Non-Specific Effect Associated with Conditional Transgene Expression Based on Cre-loxP Strategy in Mice
Transgenes flanked by loxP sites have been widely used to generate transgenic mice where the transgene expression can be controlled spatially and temporally by Cre recombinase. Data from this approach has led to important conclusions in cancer, neurodevelopment and neurodegeneration. Using this approach to conditionally express micro RNAs (miRNAs) in mice, we found that Cre-mediated recombination in neural progenitor cells caused microcephaly in five of our ten independent transgenic lines. This effect was not associated with the types or the quantity of miRNAs being expressed, nor was it associated with specific target knockdown. Rather, it was correlated with the presence of multiple tandem transgene copies and inverted (head-to-head or tail-to-tail) transgene repeats. The presence of these inverted repeats caused a high level of cell death in the ventricular zone of the embryonic brain, where Cre was expressed. Therefore, results from this Cre-loxP approach to generate inducible transgenic alleles must be interpreted with caution and conclusions drawn in previous reports may need reexamination

From bits to bites: Advancement of the Germinate platform to support prebreeding informatics for crop wild relatives
Management and distribution of experimental data from prebreeding projects
is important to ensure uptake of germplasm into breeding and research programs.
Being able to access and share this data in standard formats is essential.
The adoption of a common informatics platform for crops that may have limited
resources brings economies of scale, allowing common informatics components
to be used across multiple species. The close integration of such a platform with
commonly used breeding software, visualization, and analysis tools reduces the
barrier for entry to researchers and provides a common framework to facilitate
collaborations and data sharing. This work presents significant updates to the
Germinate platform and highlights its value in distributing prebreeding data for
14 crops as part of the project ‘Adapting Agriculture to Climate Change: Collecting,
Protecting and Preparing Crop Wild Relatives’ (hereafter Crop Trust Crop
Wild Relatives project) led by the Crop Trust (https://www.cwrdiversity.org). The
addition of data on these species compliments data already publicly available in
Germinate. We present a suite of updated Germinate features using examples
from these crop species and their wild relatives. The use of Germinate within the
Crop TrustCropWildRelatives project demonstrates the usefulness of the system
and the benefits a shared informatics platform provides. These data resources
provide a foundation on which breeding and research communities can develop
additional online resources for their crops, harness new data as it becomes available,
and benefit collectively from future developments of the Germinate platform
