    Culturally Responsive Distance Counselor Education for International Counseling Students

    Counselor education programs have increasingly offered distance education during the last decades. As one of the growing student populations in counselor education programs, international students need to be given more attention pertaining to their cultural and educational experiences as counseling students. Although distance learning in counselor education has gained empirical support for its effectiveness, it may still pose challenges to international counseling students due to their unique characteristics such as language barriers and cultural differences, which may render differing qualities of studentsā€™ learning experiences. Aiming to enhance our understanding of this student population to provide culturally responsive distance counselor education, the present article introduces international students in the context of distance counselor education and discusses implications for supporting international students in distance counselor education

    Dynamics of Online Engagement: Counseling Studentsā€™ Experiences and Perceptions in Distance Learning

    In this article, the authors present counseling studentsā€™ personal accounts of their experiences in distance education with a focus on online engagement based on an extensive review of the counseling literature. In addition, an evaluation tool was introduced for gathering qualitative and quantitative information regarding patterns of instructor engagement efforts and styles. Facilitating factors for experiential online learning were also identified from the studentsā€™ perceptions of online learning experience. Finally, the results were used to make specific recommendations for counselor educators to enhance their teaching efficacy and engagement

    Multivalent electrostatic piā€“cation interaction between synaptophysin and synapsin is responsible for the coacervation

    We recently showed that synaptophysin (Syph) and synapsin (Syn) can induce liquidā€“liquid phase separation (LLPS) to cluster small synaptic-like microvesicles in living cells which are highly reminiscent of SV cluster. However, as there is no physical interaction between them, the underlying mechanism for their coacervation remains unknown. Here, we showed that the coacervation between Syph and Syn is primarily governed by multivalent piā€“cation electrostatic interactions among tyrosine residues of Syph C-terminal (Ct) and positively charged Syn. We found that Syph Ct is intrinsically disordered and it alone can form liquid droplets by interactions among themselves at high concentration in a crowding environment in vitro or when assisted by additional interactions by tagging with light-sensitive CRY2PHR or subunits of a multimeric protein in living cells. Syph Ct contains 10 repeated sequences, 9 of them start with tyrosine, and mutating 9 tyrosine to serine (9YS) completely abolished the phase separating property of Syph Ct, indicating tyrosine-mediated pi-interactions are critical. We further found that 9YS mutation failed to coacervate with Syn, and since 9YS retains Syphs negative charge, the results indicate that piā€“cation interactions rather than simple charge interactions are responsible for their coacervation. In addition to revealing the underlying mechanism of Syph and Syn coacervation, our results also raise the possibility that physiological regulation of piā€“cation interactions between Syph and Syn during synaptic activity may contribute to the dynamics of synaptic vesicle clustering.This work was supported by grants from the National Research Foundation of Korea (Grants 2019R1A2C2089182 to S.C.).     A nanohybrid system for taste masking of sildenafil

    A nanohybrid was prepared with an inorganic clay material, montmorillonite (MMT), for taste masking of sildenafil (SDN). To further improve the taste-masking efficiency and enhance the drug-release rate, we coated the nanohybrid of SDNā€“MMT with a basic polymer, polyvinylacetal diethylaminoacetate (AEA). Powder X-ray diffraction and Fourier transform infrared experiments showed that SDN was successfully intercalated into the interlayer space of MMT. The AEA-coated SDNā€“MMT nanohybrid showed drug release was much suppressed at neutral pH (release rate, 4.70 Ā± 0.53%), suggesting a potential for drug taste masking at the buccal cavity. We also performed in vitro drug release experiments in a simulated gastric fluid (pH = 1.2) and compared the drug-release profiles of AEA-coated SDNā€“MMT and ViagraĀ®, an approved dosage form of SDN. As a result, about 90% of SDN was released from the AEA-coated SDNā€“MMT during the first 2 hours while almost 100% of drug was released from ViagraĀ®. However, an in vivo experiment showed that the AEA-coated SDNā€“MMT exhibited higher drug exposure than ViagraĀ®. For the AEA-coated SDNā€“MMT, the area under the plasma concentrationā€“ time curve from 0 hours to infinity (AUC0-āˆž) and maximum concentration (Cmax) were 78.8 Ā± 2.32 Ī¼g Ā· hour/mL and 12.4 Ā± 0.673 Ī¼g/mL, respectively, both of which were larger than those obtained with ViagraĀ® (AUC0-āˆž = 69.2 Ā± 3.19 Ī¼g Ā· hour/mL; Cmax = 10.5 Ā± 0.641 Ī¼g/mL). Therefore, we concluded that the MMT-based nanohybrid is a promising delivery system for taste masking of SDN with possibly improved drug exposure

    The immediate upstream region of the 5 0 -UTR from the AUG start codon has a pronounced effect on the translational efficiency in Arabidopsis thaliana

    ABSTRACT The nucleotide sequence around the translational initiation site is an important cis-acting element for post-transcriptional regulation. However, it has not been fully understood how the sequence context at the 5 0 -untranslated region (5 0 -UTR) affects the translational efficiency of individual mRNAs. In this study, we provide evidence that the 5 0 -UTRs of Arabidopsis genes showing a great difference in the nucleotide sequence vary greatly in translational efficiency with more than a 200-fold difference. Of the four types of nucleotides, the A residue was the most favourable nucleotide from positions ƀ1 to ƀ21 of the 5 0 -UTRs in Arabidopsis genes. In particular, the A residue in the 5 0 -UTR from positions ƀ1 to ƀ5 was required for a high-level translational efficiency. In contrast, the T residue in the 5 0 -UTR from positions ƀ1 to ƀ5 was the least favourable nucleotide in translational efficiency. Furthermore, the effect of the sequence context in the ƀ1 to ƀ21 region of the 5 0 -UTR was conserved in different plant species. Based on these observations, we propose that the sequence context immediately upstream of the AUG initiation codon plays a crucial role in determining the translational efficiency of plant genes

    Oral immunization of haemaggulutinin H5 expressed in plant endoplasmic reticulum with adjuvant saponin protects mice against highly pathogenic avian influenza A virus infection

    Pandemics in poultry caused by the highly pathogenic avian influenza (HPAI) A virus occur too frequently globally, and there is growing concern about the HPAI A virus due to the possibility of a pandemic among humans. Thus, it is important to develop a vaccine against HPAI suitable for both humans and animals. Various approaches are underway to develop such vaccines. In particular, an edible vaccine would be a convenient way to vaccinate poultry because of the behaviour of the animals. However, an edible vaccine is still not available. In this study, we developed a strategy of effective vaccination of mice by the oral administration of transgenic Arabidopsis plants (HA-TG) expressing haemagglutinin (HA) in the endoplasmic reticulum (ER). Expression of HA in the ER resulted in its high-level accumulation, N-glycosylation, protection from proteolytic degradation and long-term stability. Oral administration of HA-TG with saponin elicited high levels of HA-specific systemic IgG and mucosal IgA responses in mice, which resulted in protection against a lethal influenza virus infection with attenuated inflammatory symptoms. Based on these results, we propose that oral administration of freeze-dried leaf powders from transgenic plants expressing HA in the ER together with saponin is an attractive strategy for vaccination against influenza A virus.X111411Ysciescopu

    Operation of national coordinating service for interhospital transfer from emergency departments: experience and implications from Korea

    Background Since 2014, Korea has been operating the National Emergency Medical Situation Room (NEMSR) to provide regional emergency departments (EDs) with coordination services for the interhospital transfer of critically ill patients. The present study aimed to describe the NEMSRs experience and interhospital transfer pattern from EDs nationwide, and investigate the factors related to delayed transfers or transfers that could not be arranged by the NEMSR. Methods This study was a retrospective cross-sectional analysis of the NEMSRs coordination registry from 2017 to 2019. The demographic and hospital characteristics related to emergency transfers were analyzed with hierarchical logistic models. Results The NEMSR received a total of 14,003 requests for the arrangement of the interhospital transfers of critically ill patients from 2017 to 2019. Of 10,222 requests included in the analysis, 8297 (81.17%) successful transfers were coordinated by the NEMSR. Transfers were requested mainly due to a shortage of medical staff (59.79%) and ICU beds (30.80%). Delayed transfers were significantly associated with insufficient hospital resources. The larger the bed capacity of the sending hospital, the more difficult it was to coordinate the transfer (odds ratio [OR] for transfer not arrangedā€‰=ā€‰2.04; 95% confidence interval [CI]: 1.48ā€“2.82, ā‰„ 1000 beds vs. <ā€‰300 beds) and the longer the transfer was delayed (OR for delays of more than 44ā€‰minutesā€‰=ā€‰2.08; 95% CI: 1.57ā€“2.76, ā‰„ 1000 beds vs. <ā€‰300 beds). Conclusions The operation of the NEMSR has clinical importance in that it could efficiently coordinate interhospital transfers through a protocolized process and resource information system.     Persistent Anemia in a Patient with Diffuse Large B Cell Lymphoma: Pure Red Cell Aplasia Associated with Latent Epstein-Barr Virus Infection in Bone Marrow

    We report a case of pure red cell aplasia (PRCA), which was initially suspected as a result of bone marrow involvement of diffuse large B cell lymphoma. Persistent anemia without an obvious cause was observed in a 47-yr-old man diagnosed with relapsed diffuse large B cell lymphoma. The bone marrow study showed only erythroid hypoplasia without the evidence of bone marrow involvement with lymphoma cells, thus PRCA was suggested. However, parvovirus infection was excluded as a potential cause of PRCA because of negative IgM anti-parvovirus B19 antibody and negative parvovirus PCR in the serum. Latent Epstein-Barr virus (EBV) infection of bone marrow was suggested by in situ hybridization with EBV-encoded small RNA (EBER) that showed a strong positive expression in bone marrow cells. Thus, PRCA was thought to be associated with latent EBV infection in bone marrow cells. Although the finding of unexplained anemia is a possible predictor of bone marrow involvement with lymphoma cells, PRCA as a result of a viral infection including EBV should be considered in lymphoma patients. This is the first report of the occurrence of PRCA associated with latent EBV infection in a patient with non-Hodgkin's lymphoma
