SelbstÀndigkeitserklÀrungDas Ziel dieser Arbeit war es, die Bewertung der Wirkung von Anionenrationen
anhand von Herdendaten erfassbar zu machen und den Einsatz der Anionen
sicherer zu gestalten. Im ersten Teil der Arbeit sollte die praktische
DurchfĂŒhrung der Messung des Harn-pH zur Bewertung der Wirkung von Anionen auf
den SĂ€uren-Basen-Haushalt untersucht werden. Dazu wurde in 2 Betrieben ĂŒber
einem Zeitraum von 8 Wochen jede Woche der Harn-pH der Vorbereiter mit einem
elektrischen pH-Meter im Stall und mit pH-Streifen direkt nach der Entnahme
gemessen. AuĂerdem wurde der Harn eingefroren und nach dem Auftauen mittels
eines elektrischen pH-Meters im Labor gemessen. Die pH-Streifen lieferten
nahezu dieselben Ergebnisse wie das elektronische pH-Meter direkt nach der
Harnentnahme. Die pH-Messung nach dem Auftauen ergeben leicht höhere pH-Werte.
Im Rahmen der Bestandsbetreuung wurden 50 Betriebe untersucht, wobei einige
Betriebe mehrmals besucht wurden. Bei diesen Untersuchungen wurden Blut- und
Harnproben der Vorbereiter und der Frischkalber sowie Rationsanalysen der
Vorbereiterration durchgefĂŒhrt. Von den Blut- und Harnproben wurden aus zehn
Einzelproben sogenannte Poolproben hergestellt. FĂŒr den Harn-pH, die NSBA und
die Kalziumausscheidung mit dem Harn wurden die Poolwerte mit den Mittelwerten
der Einzelproben verglichen. Es wurden fĂŒr alle 3 Parameter hohe Korrelationen
festgestellt, so dass die Aussagekraft eines Poolwertes mit der eines
gemittelten Wertes vergleichbar ist. Die Rationsanalysen von 2002 bis 2004
enthielten die DCAB sowie die Mengenelemente Kalium, Natrium, Kalzium, Chlorid
und Schwefel. Es wurden Betriebe mit und ohne Anionenrationen
gegenĂŒbergestellt. Die Gruppe mit Anionenrationen wies vor allem eine
niedrigere DCAB sowie einen höheren Schwefelgehalt in allen drei Jahren auf.
Jedoch werden die NĂ€hrstoffempfehlungen des NRC in beiden Gruppen nicht
erreicht. Dies spricht beim Einsatz von Anionen fĂŒr eine nicht ausreichenden
Menge an sauren Salzen in Bezug auf die Rationsinhalte. Zur Beurteilung
weiterer Faktoren der Ration auf die Wirkung der Anionen wurden dem Harn-pH,
der NSBA und der Kalziumausscheidung mit dem Harn die Rationsparameter
gegenĂŒbergestellt. Bei einer Reduzierung der DCAB und des Kaliumgehaltes der
Ration sowie bei Erhöhung des Kalzium- und Schwefelgehaltes der Ration kommt
es zu einer milden Azidose gekoppelt mit einer Erhöhung der
Kalziumausscheidung mit dem Harn. Genau dies beschreibt die Wirkung der
Anionen. Indem Anionen zugefĂŒttert werden, soll eine Verschiebung der
Mengenelemente erfolgen. Dies fĂŒhrt zu einer Senkung des DCAB und zu den
genannten Folgen beim Tier. Derselbe Effekt auf den SĂ€uren-Basen-Haushalt und
die Kalziumausscheidung wurde durch die Erhöhung des StÀrkegehaltes sowie die
Senkung der Rohfaser einer Ration erzielt. Also könnte eine Wirkung der
Anionen vorgetÀuscht werden. Die Kennzahlen der Energiekonzentration einer
Ration wiesen Korrelationen mit den Parametern des SĂ€uren-Basen-Gehaltes auf.
Bei niedrigen Werten kommt es zu einer azidotischen Belastung der Tiere. Dies
muss beim Einsatz von Anionen vermieden werden, um den Effekt auf den SĂ€uren-
Basen-Haushalt nicht zu potenzieren, und so eine klinische Azidose mit ihren
negativen Folgen zu provozieren. Der Rohaschegehalt und die Salzmenge zeigten
die Effekte, die unter anderen fĂŒr die DCAB beschrieben wurden. Die Rohasche,
als Kennzahl des Mineralstoffgehaltes einer Ration, beinhaltet in gewissem
MaĂe die Salzmenge. Beim Vergleich von Betrieben mit und ohne Anionen wurden
die Rationsinhalte der Vorbereiterration sowie der SĂ€uren-Basen-Haushalt, die
Mengenelemente und ausgewÀhlte Stoffwechselparameter der Vorbereiter und
Frischkalber beurteilt. In Betrieben mit Anionen zeigten die Rationen eine
niedrigere DCAB und Kaliumkonzentration sowie eine höhere Schwefel- und
Kalziumkonzentration. Dies hatte eine metabolische Azidose der Vorbereiter zur
Folge, die sich vor allem in den Harnparametern widerspiegelte. Zudem erhöhte
sich die Kalziumausscheidung der Vorbereiter im Harn, so dass von einer
Erhöhung der Kalziumhomöostase ausgegangen werden kann. Bei nÀherer
Betrachtung zeigt sich, dass die empfohlenen Bereiche sowohl fĂŒr die
Rationsinhalte als auch fĂŒr die Stoffwechselparameter nicht erreicht werden.
So wurden im Folgenden die Betriebe mit Anionen nach der Wirkung der Anionen
eingeteilt. Dies wurde zum einen anhand der Aussage der Betriebsleiter ĂŒber
die Inzidenz der GebÀrparese versucht. Dabei wurden keine auswertbaren
Ergebnisse erzielt. Die Beurteilung der Anionen erscheint fĂŒr die Landwirte
relativ schwierig zu sein, da die meisten keine genaue Auflistung der
vorkommenden Erkrankungen im Betrieb durchfĂŒhren. Doch kann anhand der
Inzidenzen von Erkrankungen der Gesundheitszustand einer Herde und auch die
Wirkung von Anionen sehr gut beurteilt werden. Zum anderen wurden die Betriebe
durch den Harn-pH und die Kalziumausscheidung mit dem Harn eingeteilt. Es
zeigte sich in Betrieben mit wirkenden Anionen eine deutlich niedrigere DCAB.
So mĂŒssen Werte zwischen 0 und 50 meq/kg TS in der Ration der Vorbereiter
erreicht werden. Durch die Einteilung mittels des Harn-pH war eine Absenkung
der Parameter des SĂ€uren-Basen-Haushaltes bei Wirkung der Anionen zu erwarten.
Die NSBA lag im empfohlenen Bereich. Jedoch zeigten auch die Frischkalber
deutlich niedrigere Werte der Harnparameter des SĂ€uren-Basen-Haushaltes. Als
Fehlerquellen wurden eine zu hohe DCAB und ein zu hoher Kaliumgehalt sowie ein
zu niedriger Kalzium- und Schwefelgehalt der Ration angesehen. Die
NĂ€hrstoffempfehlungen des NRC erwiesen sich als ungeeignet zur Einteilung der
Betriebe mit ausreichendem oder unzureichendem NĂ€hrstoffgehalt, da diese Werte
von wenigen oder keinem Betrieb erreicht wurden. So wurde fĂŒr die DCAB, dem
Kaliumgehalt und dem Kalziumgehalt die Medianwerte der Gruppe mit Wirkung der
Anionen verwendet. Nach der Einteilung wurden die Rationsinhalte sowie der
Harn-pH, die NSBA und die Kalziumkonzentration im Harn gemessen. Bei
ausreichenden Gehalten der genannten Rationsinhalte optimierten sich die
Harnparameter. Bei der Optimierung der Rationen mĂŒssen dem Kalium- und dem
Schwefelgehalt der Rationen besondere Aufmerksamkeit gewidmet werden.
Hauptziel beim Einsatz von Anionen ist die Optimierung der DCAB. AbschlieĂend
wurden drei einzelne Betriebe beschrieben. Die ersten beiden Betriebe streuten
die Anionen ĂŒber die TMR und mischten diese mit Hilfe einer Heugabel unter. Es
wurde ĂŒber 8 Wochen jede Woche der Harn-pH, die NSBA und die
Kalziumausscheidung mit dem Harn gemessen. Bei beiden Betrieben fiel die
breite Streuung der Werte auf. DafĂŒr könnte eine unterschiedliche
Futteraufnahme oder eine individuelle Reaktion der Tiere verantwortlich sein.
Dies erklĂ€rt auch die Berichte der Landwirte ĂŒber ein Versagen der Anionen
oder auch plötzliche TodesfÀlle beim Einsatz von Anionen. Der dritte Betrieb
begann mit der FĂŒtterung von Anionen. Dieser Betrieb konnte nach Optimierung
der Ration die Anionen erfolgreich einsetzen. Die Vorbereiterration
beinhaltete einen Schwefelgehalt ĂŒber der empfohlenen Grenze von 0,4% der
Ration ohne nachteilige Effekte an den Tieren. Bei Verwendung hochwertiger
Futtermittel und genauer Kalkulation der Rationsinhalte können die toxischen
Eigenschaften von Schwefel toleriert werden. Der Einsatz von Anionenrationen
erfordert also ein sehr gutes Management. Die Menge der Anionen sowie die
Ration selbst mĂŒssen optimal berechnet sein. Trotzdem kann auf eine Kontrolle
nicht verzichtet werden. Hierzu mĂŒssen der SĂ€uren-Basen-Haushalt anhand des
Harn-pH oder der NSBA sowie der Kalziumhaushalt mittels der
Kalziumausscheidung im Harn ĂŒberprĂŒft werden. ZusĂ€tzlich muss die Ration
regelmĂ€Ăig untersucht und die HĂ€ufigkeit von Erkrankungen festgehalten werden.
Diese Arbeit beantwortet die Frage, wann welche Untersuchung durchgefĂŒhrt
werden sollte und gibt so eine Anleitung zur Kontrolle der Anionen. So kann
der Einsatz der Anionen sicherer gestaltet werden.The aim of this study was to find a solution how to valuate the effect of
anionic rations with herd datas and to make the use of anions safer. The first
part of the study shows the examinations about the practical use of the
measurement of urine ph to valuate the effects of anions on the acid-base
status. For that purpose, samples of urine were taken for eight weeks from the
two weeks prepartum cows of two different farms. The ph was measured with an
electrical ph-meter in the cowshed and with ph-stripes directly after the
winning. Apart from that, the urine was frozen and after defrosting measured
with an electrical ph-meter in the lab. The measurement of the ph by ph-
stripes showed nearly the similar results than the measurement of the urine ph
by electrical ph-meter directly after the winning. After defrosting the urine,
the results of the urine ph were slightly increased. Within the scope of herd
management fifty farms were visited and some of them more than one time. Blood
and urine samples were taken from close up dry cows and recently calved cows.
The ration of the close up dry cows was analysed. Ten single samples of blood
and urine were necessary to produce a pool sample. The values of the pool
samples of urine ph, net acid-base excretion and urine calcium excretion were
compared with the mean values of the single samples. There were high
correlations between the pool and the mean values for each of the parameters.
Because of that, the pool and the mean values have nearly the same content of
information. The analyses of the rations from 2002 to 2004 contain the dcad
and the amount of potassium, sodium, chloride and sulphur. Herds with and
without anionic rations were compared. The analysis of the feed of herds with
anionic rations showed a lower dcad and a higher amount of sulphur in all of
the three years. But the recommendations of the NRC were not achieved. That is
an indication that not enough anionic salts were used refer to the rations
content. To examine the influence of more factors of the ration on the effect
of the anions the urine ph, the net acid-base excretion and the urine calcium
excretion were compared with the parameters of the rations. The effect of
reducing the dcad and the amount of potassium as well as the effect of
increasing the amount of calcium and sulphur in the ration is a mild acidosis
and an increased urine calcium excretion. These are the effects of the anions.
Within feeding anions there should be a shift of the major elements. This
leads to a decreased dcad and to the metabolic effects as described. The same
effects on the acid-base status and the urine calcium excretion were observed
by feeding an increased amount of starch and a decreased amount of crude
fibre. This could simulate the effect of anionic rations. The parameters of
the energy concentration in the rations have a slight correlation with the
parameters of the acid-base status. At low amounts of energy there is an
acidotic stress of the animals. This has to be avoided when farmers use
anionic rations because it could raise the effect on the acid-base status and
provoke a clinical acidosis with its negative results. The crude ash and the
amount of salt showed the same effects as described for the dcad. The crude
ash as a characteristic number for the major elements contains a certain
degree of the amount of anions. The comparison of farms with or without
anionic rations includes the rations analysis, the acid-base status, the major
elements and some metabolic parameters from close up dry cows and from
recently calved cows. Farms with anionic rations had a lower dcad and a lower
amount of potassium as well as higher amounts of sulphur and calcium in their
rations. This result in a mild metabolic acidosis above all reflected in the
urine parameters of the cows two weeks prepartum. Besides the urine calcium
excretion of the close up dry cows were increased, so that a heighten calcium
homeostasis can be assumed. The closer inspection showed that the recommended
values for the rations and the metabolic parameters are not achieved. So the
farms feeding anionic rations were divided into farms with or without an
effect of the anions. This was attempted by the statement of the farmers about
the incidence of milk fever. But there were no useable results. The assessment
of the effect of the anions seemed to be difficult for the most farmers
because the most farmers don t list the diseases in their herd. However the
incidence of diseases is useful for the assessment of the state of health of
the herd and the effect of the anionic ration. Moreover the farms were divided
on the basis of the urine ph and the urine calcium excretion. The farms with
an effect of the anions showed a lower dcad. There should be a dcad between 0
50 meq/kg DM in the ration of close up dry cows. Because of the
classification by the urine ph was a reduction of the parameters of the acid-
base status in the group with an effect of the anions expected. The net acid-
base excretion was in the recommended area. The recently calved cows in farms
with an effect of anions showed lower values of the parameters of the acid-
base status. A high dcad and a high amount of potassium as well as a low
amount of calcium and sulphur in the rations were considered as sources of
errors. The nutrient recommendations of the NRC were unsuitable for the
classification of the farms sufficient or insufficient nutrient amount because
the recommended values were not achieved. So the values of the dcad, the
amount of potassium and the amount of calcium were taken from the group of
farms with an effect of anions. After this classification the ration analysis
as well as the urine ph, the net acid-base excretion and the urine calcium
excretion were studied. With sufficient amounts of the rations contents the
urine parameter were optimized. To optimize the ration you have to give
special attention to the amount of potassium and sulphur. The main target by
using anionic rations is to optimize the dcad. Finally there were three farms
described. Two farms sprinkled the anions over the ration and mixed it with a
pitchfork. The urine ph, the net acid-base excretion and the urine calcium
excretion were measured over eight weeks. In both farms is a great variation
of the values conspicuous. For this could be a varying feed intake or an
individual reaction of each cow responsible. This also explained the farmers
report about a failure or sudden death by using anionic rations. The third
farm started using anionic rations. After optimizing the ration the farm uses
the anions with success. The close up dry cow ration had an amount of sulphur
over the recommended 0,4% without any negative effect at the animals. By using
high quality food and a well calculated ration the toxic effects of sulphur
can be tolerated. The use of anionic rations needs a very good management. The
amount of anions as well as the ration should be optimal calculated. Also the
check of the effect of the anions is essential. For this the acid-base status
has to be controlled on the basis of urine ph and the net acid-base excretion.
The calcium homeostasis has to be controlled on the basis of the urine calcium
excretion. Additionally the ration has to be analysed regularly and the
incidence of the diseases in the herd has to be listed. This study describes
when and which analyse should be made and leads to a guidance to control the
effect of anions. This makes the use of anionic rations safer
Myelin dysfunction drives amyloid-ÎČ deposition in models of Alzheimer's disease
The incidence of Alzheimer's disease (AD), the leading cause of dementia, increases rapidly with age, but why age constitutes the main risk factor is still poorly understood. Brain ageing affects oligodendrocytes and the structural integrity of myelin sheaths(1), the latter of which is associated with secondary neuroinflammation(2,3). As oligodendrocytes support axonal energy metabolism and neuronal health(4-7), we hypothesized that loss of myelin integrity could be an upstream risk factor for neuronal amyloid-beta (A beta) deposition, the central neuropathological hallmark of AD. Here we identify genetic pathways of myelin dysfunction and demyelinating injuries as potent drivers of amyloid deposition in mouse models of AD. Mechanistically, myelin dysfunction causes the accumulation of the A beta-producing machinery within axonal swellings and increases the cleavage of cortical amyloid precursor protein. Suprisingly, AD mice with dysfunctional myelin lack plaque-corralling microglia despite an overall increase in their numbers. Bulk and single-cell transcriptomics of AD mouse models with myelin defects show that there is a concomitant induction of highly similar but distinct disease-associated microglia signatures specific to myelin damage and amyloid plaques, respectively. Despite successful induction, amyloid disease-associated microglia (DAM) that usually clear amyloid plaques are apparently distracted to nearby myelin damage. Our data suggest a working model whereby age-dependent structural defects of myelin promote A beta plaque formation directly and indirectly and are therefore an upstream AD risk factor. Improving oligodendrocyte health and myelin integrity could be a promising target to delay development and slow progression of AD
Split-Cre Complementation Indicates Coincident Activity of Different Genes In Vivo
Cre/LoxP recombination is the gold standard for conditional gene regulation in mice in vivo. However, promoters driving the expression of Cre recombinase are often active in a wide range of cell types and therefore unsuited to target more specific subsets of cells. To overcome this limitation, we designed inactive âsplit-Creâ fragments that regain Cre activity when overlapping co-expression is controlled by two different promoters. Using transgenic mice and virus-mediated expression of split-Cre, we show that efficient reporter gene activation is achieved in vivo. In the brain of transgenic mice, we genetically defined a subgroup of glial progenitor cells in which the Plp1- and the Gfap-promoter are simultaneously active, giving rise to both astrocytes and NG2-positive glia. Similarly, a subset of interneurons was labelled after viral transfection using Gad67- and Cck1 promoters to express split-Cre. Thus, split-Cre mediated genomic recombination constitutes a powerful spatial and temporal coincidence detector for in vivo targeting
Prozessverbesserung ĂŒber Fehlerstrommessung bei einem mittelstĂ€ndischen Unternehmen
Eine konstant hohe ProduktqualitĂ€t ist insbesondere fĂŒr kleine und mittelstĂ€ndische Unternehmen (KMUs) wichtig, um Kundenzufriedenheit gewĂ€hrleisten zu können. Dabei ist ein guter Entwicklungs- und QualitĂ€tssicherungsprozess maĂgeblich fĂŒr die Erreichung einer hohen SoftwarequalitĂ€t verantwortlich. Die systematische Fehlermessung nimmt dabei eine SchlĂŒsselrolle ein, da nur so empirisch zu ermitteln ist, wie effektiv bestehende QualitĂ€tssicherungs-Prozesse sind, welche Fehlerarten durch welche Prozesse gefunden werden und welches Verbesserungspotential gegeben ist. In diesem Beitrag werden das Vorgehen und die Erfahrungen bei der Definition und EinfĂŒhrung eines Messprogramms zur Erfassung eines Fehlerstrommodells (FSM) bei einem mittelstĂ€ndischen Unternehmen beschrieben. Das Messprogramm ist eingefĂŒhrt und wird aktiv betrieben. Dies ist unseres Wissens die erste Dokumentation der EinfĂŒhrung eines FSM bei einer KMU. Durch die gewonnenen Erfahrungen konnte der Definitions- und EinfĂŒhrungsprozess fĂŒr KMUs verfeinert werden. Trotz des frĂŒhen Zeitpunkts und des durch die statistische Evaluierung aufgezeigten Verbesserungspotentials hinsichtlich der QualitĂ€t des Schemas und der Fehlererfassung können schon erste interessante Ergebnisse hinsichtlich der Fehlerkorrekturkosten unterschiedlicher Fehlerarten und Entdeckungszeitpunkte prĂ€sentiert werden
A framework for the balanced optimization of quality assurance strategies focusing on small and medium sized enterprises
The Quality Improvement Paradigm (QIP) offers a general framework for systematically improving an organization's development processes in a continuous manner. In the context of the LifeCycleQM project, the general QIP framework was concretized for a specific application area, namely, the balanced improvement of quality assurance (QA) strategies, i.e., a set of systematically applied QA activities. Especially with respect to small and medium-sized enterprises, the encompassing QIP framework presents limited guidance for easy and concrete application. Therefore, individual activities within the QIP framework were refined focusing on QA strategies, and proven measurement procedures such as the defect flow model and knowledge from the area of process improvement (e.g., with regard to individual QA procedures) was integrated for reuse in a practice-oriented manner. The feasibility of the developed approach was initially explored by its application at IBS AG, a medium-sized enterprise, where improvement goals were defined, a corresponding measurement program was established, improvement potential was identified, and concrete improvement suggestions for the QA strategy were derived, assessed, and implemented
