    HST astrometry in the Orion Nebula Cluster: census of low-mass runaways

    We present a catalog of high-precision proper motions in the Orion Nebula Cluster (ONC), based on Treasury Program observations with the Hubble Space Telescope's (HST) ACS/WFC camera. Our catalog contains 2,454 objects in the magnitude range of 14.2<mF775W<24.714.2<m_{\rm F775W}<24.7, thus probing the stellar masses of the ONC from \sim0.4 MM_\odot down to \sim0.02 MM_\odot over an area of \sim550 arcmin2^2. We provide a number of internal velocity dispersion estimates for the ONC that indicate a weak dependence on the stellar location and mass. There is good agreement with the published velocity dispersion estimates, although nearly all of them (including ours at σv,x=0.94\sigma_{v,x}=0.94 and σv,y=1.25\sigma_{v,y}=1.25 mas yr1^{-1}) might be biased by the overlapping young stellar populations of Orion A. We identified 4 new ONC candidate runaways based on HST and the Gaia DR2 data, all with masses less than \sim1 MM_\odot. The total census of known candidate runaway sources is 10 -- one of the largest samples ever found in any Milky Way open star cluster. Surprisingly, none of them has the tangential velocity exceeding 20 km s1^{-1}. If most of them indeed originated in the ONC, it may compel re-examination of dynamical processes in very young star clusters.     koopmans: an open-source package for accurately and efficiently predicting spectral properties with Koopmans functionals

    Over the past decade we have developed Koopmans functionals, a computationally efficient approach for predicting spectral properties with an orbital-density-dependent functional formulation. These functionals address two fundamental issues with density functional theory (DFT). First, while Kohn-Sham eigenvalues can loosely mirror experimental quasiparticle energies, they are not meant to reproduce excitation energies and there is formally no connection between the two (except for the HOMO for the exact functional). Second, (semi-)local DFT deviates from the expected piecewise linear behavior of the energy as a function of the total number of electrons. This can make eigenvalues an even poorer proxy for quasiparticle energies and, together with the absence of the exchange-correlation derivative discontinuity, contributes to DFT's underestimation of band gaps. By enforcing a generalized piecewise linearity condition to the entire electronic manifold, Koopmans functionals yield molecular orbital energies and solid-state band structures with comparable accuracy to many-body perturbation theory but at greatly reduced computational cost and preserving a functional formulation. This paper introduces "koopmans", an open-source package that contains all of the code and workflows needed to perform Koopmans functional calculations without requiring expert knowledge.     Inulin from Globe Artichoke Roots: A Promising Ingredient for the Production of Functional Fresh Pasta

    Globe artichoke roots represent an alternative and sustainable source for inulin extraction and are well-noted for their technological and functional properties. Therefore, the aim of our study was to exploit inulin with high degree of polymerization as a replacement of durum wheat semolina for the production of functional fresh pasta. The effect of increased level of substitution (5, 10, 15%) on cooking, structural, sensory, and nutritional properties were evaluated and compared with a control sample consisting exclusively of durum wheat semolina. Inulin addition caused changes to internal structure as evaluated by scanning electron microscopy. The enriched samples showed a lower swelling index, an increasing cooking time, and values of cooking loss (2.37-3.62%), mainly due to the leaching of inulin into the cooking water. Cooked and raw enriched pasta was significantly darker and firmer than the control, but the sensory attributes were not negatively affected, especially at 5 and 10% of substitution levels. The increase of dietary fiber content in enriched pasta (3.44-12.41 g/100 g) resulted in a significant reduction of glycaemic index (pGI) and starch hydrolysis (HI).     Perioperative characterization of anastomotic doughnuts with high-resolution probe-based confocal laser endomicroscopy in colorectal cancer surgery: a feasibility study.

    BACKGROUND: Confocal laser enables in vivo real-time histopathology of the mucosa layer of gastrointestinal tract. The aim of this study was to assess the feasibility and the role of probe-based confocal laser endomicroscopy in extemporary examination of staple rings of patients with colorectal cancer. METHODS: This was a prospective, single-center pilot study. Patients who underwent end-to-end stapled surgical resection for colorectal cancer were included. Confocal imaging was analyzed with great attention to image quality evaluation of cellular morphology and cellular structures of the serosal, muscular, and mucosal layers of the doughnuts than comparing results with definitive histopathology. RESULTS: Twenty-six doughnuts were analyzed. Real-time video sequences were obtained in all patients, with a total of 204 mosaic images. For each doughnut, most of the images were adequate for evaluation of cellular morphology and cellular structure of the serosal, muscular, and mucosal layers. CONCLUSIONS: Perioperative assessment of doughnut tissues in patients with colorectal cancer by confocal laser endomicroscopy is feasible and safe. Prospective studies are warranted for further evaluation of the clinical impact of this technology during surgery

    In vitro and in vivo evaluation of BMAP-derived peptides for the treatment of cystic fibrosis-related pulmonary infections

    Patients with cystic fibrosis require pharmacological treatment against chronic lung infections. The alpha-helical antimicrobial peptides BMAP-27 and BMAP-28 have shown to be highly active in vitro against planktonic and sessile forms of multidrug-resistant Pseudomonas aeruginosa, Staphylococcus aureus, and Stenotrophomonas maltophilia cystic fibrosis strains. To develop small antibacterial peptides for therapeutic use, we tested shortened/modified BMAP fragments, and selected the one with the highest in vitro antibacterial activity and lowest in vivo acute pulmonary toxicity. All the new peptides have shown to roughly maintain their antibacterial activity in vitro. The 1-18 N-terminal fragment of BMAP-27, showing MIC90 of 16 A mu g/ml against P. aeruginosa isolates and strain-dependent anti-biofilm effects, showed the lowest pulmonary toxicity in mice. However, when tested in a murine model of acute lung infection by P. aeruginosa, BMAP-27(1-18) did not show any curative effect. If exposed to murine broncho-alveolar lavage fluid BMAP-27(1-18) was degraded within 10 min, suggesting it is not stable in pulmonary environment, probably due to murine proteases. Our results indicate that shortened BMAP peptides could represent a starting point for antibacterial drugs, but they also indicate that they need a further optimization for effective in vivo use

    MC profiling: a novel approach to analyze DNA methylation heterogeneity in genome-wide bisulfite sequencing data

    : DNA methylation is an epigenetic mark implicated in crucial biological processes. Most of the knowledge about DNA methylation is based on bulk experiments, in which DNA methylation of genomic regions is reported as average methylation. However, average methylation does not inform on how methylated cytosines are distributed in each single DNA molecule. Here, we propose Methylation Class (MC) profiling as a genome-wide approach to the study of DNA methylation heterogeneity from bulk bisulfite sequencing experiments. The proposed approach is built on the concept of MCs, groups of DNA molecules sharing the same number of methylated cytosines. The relative abundances of MCs from sequencing reads incorporates the information on the average methylation, and directly informs on the methylation level of each molecule. By applying our approach to publicly available bisulfite-sequencing datasets, we individuated cell-to-cell differences as the prevalent contributor to methylation heterogeneity. Moreover, we individuated signatures of loci undergoing imprinting and X-inactivation, and highlighted differences between the two processes. When applying MC profiling to compare different conditions, we identified methylation changes occurring in regions with almost constant average methylation. Altogether, our results indicate that MC profiling can provide useful insights on the epigenetic status and its evolution at multiple genomic regions

    What’s the best assessment of preload after cardiac surgery?

    Objective: The assessment of the role of transesophageal echocardiography and invasive tests with pulmonary modified catheter to monitor the preload indexes in patients in intensive- care-unit after cardiac coronary surgery. Materials and Methods: Between January and December 2004 24 patients (14 male, 10 female) with coronary artery disease were prospectively enrolled for preload assessment during off-pump myocardial revascularization. Pulmonary Capillary Wedge Pressure (PCWP), Left Ventricular End Diastolic Indexed Area (LVEDAI), Δ Aortic Velocity (ΔVAo), Right Ventricular End Diastolic Volume (RVEDVI) as preload indexes were evaluated. Transesophageal echocardiography and pulmonary modified catheter monitoring were performed during the preoperative period at T1 and after fluid infusion (T2). Patients were considered Responders (R) or No Responders (NR) if the Stroke Volume Index increase at T2 was >20% with respect to T1. Results: Mean T1 PCWP was similar in both groups (12.8±2.2 in R vs. 11.4±3 mmHg in NR; p=NS) and mean increase of PCWP at T2 was similar in both groups (1.5±0.3% in R vs. 1.2±3% in NR; p=NS). Mean T1 RVEDVI was similar in both groups (97.33±34 in R vs. 101±21 ml/m2 in NR; p=NS); T2 RVEDVI was similar in R and NR Groups (122.11±49 vs. 138.54±30 ml/m2; p=NS); mean T1 and T2 LVEDAI was similar in R and NR (11.2±3.5 vs. 10.2±2.3 at T1 and 14.04±3.35 vs. 14.67±2.1 cm2/m2 at T2 respectively; p=NS). Higher mean value of T1 ΔVAo (20±7% in R vs. 10±2% in NR; p=0.006) were recorded while similar mean value of T2 ΔVAo were observed (11±3% in R vs. 5±2% in NR; p=0.743). Correlation index between T1 and T2 ΔVAo (R=0.82) in R was significant (p=0.0002), while correlation index between T1 and T2 ΔVAo (R=0.11) in NR was not significant. Conclusions: Our study showed in patients soon after coronary cardiac surgery ΔVAo is the only predictor of “fluid responsiveness” and of ventricular compliance

    Naples Prognostic Score Predicts Tumor Regression Grade in Resectable Gastric Cancer Treated with Preoperative Chemotherapy

    : Despite recent progresses, locally advanced gastric cancer remains a daunting challenge to embrace. Perioperative chemotherapy and D2-gastrectomy depict multimodal treatment of gastric cancer in Europe, shows better results than curative surgery alone in terms of downstaging, micrometastases elimination, and improved long-term survival. Unfortunately, preoperative chemotherapy is useless in about 50% of cases of non-responder patients, in which no effect is registered. Tumor regression grade (TRG) is directly related to chemotherapy effectiveness, but its understanding is achieved only after surgical operation; accordingly, preoperative chemotherapy is given indiscriminately. Conversely, Naples Prognostic Score (NPS), related to patient immune-nutritional status and easily obtained before taking any therapeutic decision, appeared an independent prognostic variable of TRG. NPS was calculated in 59 consecutive surgically treated gastric cancer patients after neoadjuvant FLOT4-based chemotherapy. 42.2% of positive responses were observed: all normal NPS and half mild/moderate NPS showed significant responses to chemotherapy with TRG 1-3; while only 20% of the worst NPS showed some related benefits. Evaluation of NPS in gastric cancer patients undergoing multimodal treatment may be useful both in selecting patients who will benefit from preoperative chemotherapy and for changing immune-nutritional conditions in order to improve patient's reaction against the tumor
