    Wind turbine condition assessment through power curve copula modeling

    Power curves constructed from wind speed and active power output measurements provide an established method of analyzing wind turbine performance. In this paper it is proposed that operational data from wind turbines are used to estimate bivariate probability distribution functions representing the power curve of existing turbines so that deviations from expected behavior can be detected. Owing to the complex form of dependency between active power and wind speed, which no classical parameterized distribution can approximate, the application of empirical copulas is proposed; the statistical theory of copulas allows the distribution form of marginal distributions of wind speed and power to be expressed separately from information about the dependency between them. Copula analysis is discussed in terms of its likely usefulness in wind turbine condition monitoring, particularly in early recognition of incipient faults such as blade degradation, yaw and pitch errors

    Functional characterization of a glutamate/aspartate transporter from the mosquito Aedes aegypti

    Glutamate elicits a variety of effects in insects, including inhibitory and excitatory signals at both neuromuscular junctions and brain. Insect glutamatergic neurotransmission has been studied in great depth especially from the standpoint of the receptor-mediated effects, but the molecular mechanisms involved in the termination of the numerous glutamatergic signals have only recently begun to receive attention. In vertebrates, glutamatergic signals are terminated by Na^+/K^+-dependent high-affinity excitatory amino acid transporters (EAAT), which have been cloned and characterized extensively. Cloning and characterization of a few insect homologues have followed, but functional information for these homologues is still limited. Here we report a study conducted on a cloned mosquito EAAT homologue isolated from the vector of the dengue virus, Aedes aegypti. The deduced amino acid sequence of the protein, AeaEAAT, exhibits 40–50% identity with mammalian EAATs, and 45–50% identity to other insect EAATs characterized thus far. It transports l-glutamate as well as l- and d-aspartate with high affinity in the micromolar range, and demonstrates a substrate-elicited anion conductance when heterologously expressed in Xenopus laevis oocytes, as found with mammalian homologues. Analysis of the spatial distribution of the protein demonstrates high expression levels in the adult thorax, which is mostly observed in the thoracic ganglia. Together, the work presented here provides a thorough examination of the role played by glutamate transport in Ae. aegypti

    Wildlife contraception is only now emerging as a wildlife policy issue It will emerge into a sociopolitical environment that is already polarized from a clash of ideologies. The wildlife conservation/hunting community strives to preserve the status quo while animal welfare and animal rights activists struggle to change wildlife management philosophy and practice to conform to their respective beliefs. Recent professional and popular literature reveal at least four major areas of conflict: (1) anti-management sentiment, (2) anti-hunting sentiment, (3) animal rights sentiment, and (4) animal welfare sentiment. Wildlife managers anticipate that the conflict over the use of contraceptives will involve value and belief conflicts between traditional wildlife management and animal rights proponents. We believe instead that the primary conflicts will revolve around pragmatic issues such as when, where, and in which circumstances managers will LISP the contraceptive tool. In this context, wildlife contraception will be regarded as a mixed bag. Given the nature and potential polarity of the wildlife contraception issue, wildlife agencies will have to behave proactively by projecting themselves into their future. Currently, wildlife agencies respond to many policy challenges reactively and defensively in an attempt to preserve their past. If a productive compromise can be reached over the issue of if, how, when, and where to use wildlife contraception, the wildlife policy decision process must be visionary, wise, bold, accessible, adaptable, and, most of all, fair

    The transporter-like protein inebriated mediates hyperosmotic stimuli through intracellular signaling

    We cloned the inebriated homologue MasIne from Manduca sexta and expressed it in Xenopus laevis oocytes. MasIne is homologous to neurotransmitter transporters but no transport was observed with a number of putative substrates. Oocytes expressing MasIne respond to hyperosmotic stimulation by releasing intracellular Ca(2+), as revealed by activation of the endogenous Ca(2+)-activated Cl(-) current. This Ca(2+) release requires the N-terminal 108 amino acid residues of MasIne and occurs via the inositol trisphosphate pathway. Fusion of the N terminus to the rat gamma-aminobutyric acid transporter (rGAT1) also renders rGAT1 responsive to hyperosmotic stimulation. Immunohistochemical analyses show that MasIne and Drosophila Ine have similar tissue distribution patterns, suggesting functional identity. Inebriated is expressed in tissues and cells actively involved in K(+) transport, which suggests that it may have a role in ion transport, particularly of K(+). We propose that stimulation of MasIne releases intracellular Ca(2+) in native tissues, activating Ca(2+)-dependent K(+) channels, and leading to K(+) transport

    This article outlines a collaboration between the Foundation Centre and the Education PGCE department at Durham University to support student teachers on a particular academic writing assignment (the Self Directed Study assignment). This project involved designing online academic writing activities tailored especially to the assignment. These activities made use of excerpts of exemplar student writing as well as the FOCUS corpus, a Foundation Centre initiated database of “good” (2:1 or First) Durham University student writing. The article outlines the challenges for the students with this particular assignment, details of the collaboration, student feedback from the first roll out of the activities as well as future steps in this ongoing project

    Immobilization of anode-attached microbes in a microbial fuel cell

    Current-generating (exoelectrogenic) bacteria in bioelectrochemical systems (BESs) may not be culturable using standard in vitro agar-plating techniques, making isolation of new microbes a challenge. More in vivo like conditions are needed where bacteria can be grown and directly isolated on an electrode. While colonies can be developed from single cells on an electrode, the cells must be immobilized after being placed on the surface. Here we present a proof-of-concept immobilization approach that allows exoelectrogenic activity of cells on an electrode based on applying a layer of latex to hold bacteria on surfaces. The effectiveness of this procedure to immobilize particles was first demonstrated using fluorescent microspheres as bacterial analogs. The latex coating was then shown to not substantially affect the exoelectrogenic activity of well-developed anode biofilms in two different systems. A single layer of airbrushed coating did not reduce the voltage produced by a biofilm in a microbial fuel cell (MFC), and more easily applied dip-and-blot coating reduced voltage by only 11% in a microbial electrolysis cell (MEC). This latex immobilization procedure will enable future testing of single cells for exoelectrogenic activity on electrodes in BESs

    Identification of missing persons from mass disasters is based on evaluation of a number of variables and observations regarding the combination of features derived from these variables. DNA typing now is playing a more prominent role in the identification of human remains, and particularly so for highly decomposed and fragmented remains. The strength of genetic associations, by either direct or kinship analyses, is often quantified by calculating a likelihood ratio. The likelihood ratio can be multiplied by prior odds based on nongenetic evidence to calculate the posterior odds, that is, by applying Bayes' Theorem, to arrive at a probability of identity. For the identification of human remains, the path creating the set and intersection of variables that contribute to the prior odds needs to be appreciated and well defined. Other than considering the total number of missing persons, the forensic DNA community has been silent on specifying the elements of prior odds computations. The variables include the number of missing individuals, eyewitness accounts, anthropological features, demographics and other identifying characteristics. The assumptions, supporting data and reasoning that are used to establish a prior probability that will be combined with the genetic data need to be considered and justified. Otherwise, data may be unintentionally or intentionally manipulated to achieve a probability of identity that cannot be supported and can thus misrepresent the uncertainty with associations. The forensic DNA community needs to develop guidelines for objectively computing prior odds
