    The preservation of quartz grain surface textures following vehicle fire and their use in forensic enquiry

    During a terrorist trial, dispute arose as to whether the temperature produced in a car fire was sufficient to destroy quartz grain surface textures. A series of seven sequential experiments showed that the temperature for quartz surface texture modification/destruction and the production of vugs, vesicles and glassy precipitation ('snowdrifting') occurred at 1200 degrees C under normal atmospheric conditions. By adding a number of man-made and natural substances, it was found that only the presence of salts depressed this modification temperature (to 900 degrees C). Experiments to determine the temperature of fire in a car indicated that the maximum temperature produced under natural conditions (810 degrees C) was insufficient to affect the quartz grain Surface textures. These results confirm the use of surface texture analysis of quartz grains recovered from the remains of cars Subjected to fire and their use as a forensic indicator.     Peripheral blood eosinophils: a surrogate marker for airway eosinophilia in stable COPD.

    INTRODUCTION: Sputum eosinophilia occurs in approximately one-third of stable chronic obstructive pulmonary disease (COPD) patients and can predict exacerbation risk and response to corticosteroid treatments. Sputum induction, however, requires expertise, may not always be successful, and does not provide point-of-care results. Easily applicable diagnostic markers that can predict sputum eosinophilia in stable COPD patients have the potential to progress COPD management. This study investigated the correlation and predictive relationship between peripheral blood and sputum eosinophils. It also examined the repeatability of blood eosinophil counts. METHODS: Stable COPD patients (n=141) were classified as eosinophilic or noneosinophilic based on their sputum cell counts (≥3%), and a cross-sectional analysis was conducted comparing their demographics, clinical characteristics, and blood cell counts. Receiver operating characteristic curve analysis was used to assess the predictive ability of blood eosinophils for sputum eosinophilia. Intraclass correlation coefficient was used to examine the repeatability of blood eosinophil counts. RESULTS: Blood eosinophil counts were significantly higher in patients with sputum eosinophilia (n=45) compared to those without (0.3×10(9)/L vs 0.15×10(9)/L; P<0.0001). Blood eosinophils correlated with both the percentage (ρ=0.535; P<0.0001) and number of sputum eosinophils (ρ=0.473; P<0.0001). Absolute blood eosinophil count was predictive of sputum eosinophilia (area under the curve =0.76, 95% confidence interval [CI] =0.67-0.84; P<0.0001). At a threshold of ≥0.3×10(9)/L (specificity =76%, sensitivity =60%, and positive likelihood ratio =2.5), peripheral blood eosinophil counts enabled identification of the presence or absence of sputum eosinophilia in 71% of the cases. A threshold of ≥0.4×10(9)/L had similar classifying ability but better specificity (91.7%) and higher positive likelihood ratio (3.7). In contrast, ≥0.2×10(9)/L offered a better sensitivity (91.1%) for ruling out sputum eosinophilia. There was a good agreement between two measurements of blood eosinophil count over a median of 28 days (intraclass correlation coefficient =0.8; 95% CI =0.66-0.88; P<0.0001). CONCLUSION: Peripheral blood eosinophil counts can help identify the presence or absence of sputum eosinophilia in stable COPD patients with a reasonable degree of accuracy

    While Quine’s linguistic behaviorism is well-known, his Kant Lectures contain one of his most detailed discussions of behaviorism in psychology and the philosophy of mind. Quine clarifies the nature of his psychological commitments by arguing for a modest view that is against ‘excessively restrictive’ variants of behaviorism while maintaining ‘a good measure of behaviorist discipline…to keep [our mental] terms under control’. In this paper, I use Quine’s Kant Lectures to reconstruct his position. I distinguish three types of behaviorism in psychology and the philosophy of mind: ontological behaviorism, logical behaviorism, and epistemological behaviorism. I then consider Quine’s perspective on each of these views and argue that he does not fully accept any of them. By combining these perspectives we arrive at Quine’s surprisingly subtle view about behaviorism in psychology

    Reagentless Affimer- and antibody-based impedimetric biosensors for CEA-detection using a novel non-conducting polymer

    Polyoctopamine (POct), an amine-functionalised non-conducting polymer, as the transducer layer in an electrochemical biosensor, is presented. This polymer offers versatile covalent coupling either through thiol linker conjugation, carboxyl or aldehyde functional groups without the requirement of pre- or post-surface activation. The colorectal cancer biomarker carcinoembryonic antigen (CEA) was selected as the target analyte, whilst an antibody and a synthetic binding protein, an Affimer, were used as distinct bioreceptors to demonstrate the versatility of polyoctopamine as a transducer polymer layer for oriented immobilisation of the bioreceptors. The electrodeposited polymer layer was characterised using cyclic voltammetry, electrochemical impedance spectroscopy, and on-sensor chemiluminescent blotting. The performance of optimised POct-based biosensors were tested in spiked human serum. Results showed that the electropolymerisation of octopamine on screen printed gold electrode generates a thin polymer film with low resistance. Close proximity of the immobilised bioreceptors to the transducer layer greatly enhanced the sensitivity detection. The sensitivity of the smaller monomeric bioreceptor (Affimer, 12.6 kDa) to detect CEA was comparable to the dimeric antibody (150 kDa) with limit of detection at 11.76 fM which is significantly lower than the basal clinical levels of 25 pM. However, the Affimer-based sensor had a narrower dynamic range compared to the immunosensor (1–100 fM vs. 1 fM – 100 nM, respectively). All electrochemical measurements were done in less than 5 min with small sample volumes (10 μl). Hence, polyoctopamine features a simple fabrication of impedimetric biosensors using amine-functionalisation technique, provides rapid response time with enhanced sensitivity and label-free detection

    VLT/FORS2 comparative transmission spectroscopy II: confirmation of a cloud-deck and Rayleigh scattering in WASP-31b, but no potassium?

The observations cover a wavelength range of ≈400–840 nm. The light curves are corrupted by significant systematics, but these were to first order invariant with wavelength and could be removed using a commonmode correction derived from the white light curves. We reach a precision in the transit depth of ≈140 ppm in 15 nm bins, although the precision varies significantly over the wavelength range. Our FORS2 observations confirm the cloud-deck previously inferred using HST/STIS. We also re-analyse the HST/STIS data using a Gaussian process model, finding excellent agreement with earlier measurements. We reproduce the Rayleigh scattering signature at short wavelengths (. 5300 ˚A) and the cloud-deck at longer wavelengths. However, our FORS2 observations appear to rule out the large potassium feature previously detected using STIS, yet it is recovered from the HST/STIS data, although with reduced amplitude and significance (≈ 2.5σ). The discrepancy between our results and the earlier STIS detection of potassium (≈ 4.3σ) is either a result of telluric contamination of the ground-based observations, or an underestimate of the uncertainties for narrow-band features in HST/STIS when using linear basis models to account for the systematics. Our results further demonstrate the use of ground-based multi-object spectrographs for the study of exoplanet atmospheres, and highlight the need for caution in our interpretation of narrow-band features in low-resolution spectra of hot-Jupiters.This work is based on observations collected at the European Organisation for Astronomical Research in the Southern Hemisphere under ESO programme 096.C-0765.
