FEASIBILITY OF AN OKLAHOMA FRESH GREENS AND COWPEAS PACKING COOPERATIVE

Oklahoma's green producers are not benefiting from a growing fresh market. In order to seize the opportunities offered by the growing fresh market for leafy greens, investment in packing facilities have been evaluated. To make use of these facilities during summer months, the addition of a cowpea shelling enterprise is considered. A business plan for a new generation cooperative is estimated using an updated version of "The Packing Simulation Model" (PACKSIM) The business associates PACKSIM with @RISK®, to incorporate risks in the financial analysis.Agribusiness,

3-hydroxykynurenine suppresses CD4+ T-cell proliferation, induces T-regulatory-cell development, and prolongs corneal allograft survival

IDO (indoleamine 2,3-dioxygenase) modulates the immune response by depletion of the essential amino acid tryptophan, and IDO overexpression has been shown to prolong corneal allograft survival. This study was conducted to examine the effect of kynurenines, the products of tryptophan breakdown and known to act directly on T lymphocytes, on corneal graft survival. Methods. The effects of kynurenines on T-cell proliferation and death, T-regulatory-cell development, and dendritic cell function, phenotype, and viability were analyzed in vitro. The effect of topical and systemic administration of 3-hydroxykynurenine (3HK) on orthotopic murine corneal allograft survival was examined. Results. T-lymphocyte proliferation was inhibited by two of the four different kynurenines: 3HK and 3-hydroxyanthranilic acid (3HAA). This effect was accompanied by significant T-cell death. Neither 3HK nor 3HAA altered dendritic cell function, nor did they induce apoptosis or pathogenicity to corneal endothelial cells. Administration of systemic and topical 3HK to mice receiving a fully mismatched corneal graft resulted in significant prolongation of graft survival (median survival of control grafts, 12 days; of treated, 19 and 15 days, respectively; P < 0.0003). While systemic administration of 3HK was associated with a significant depletion of CD4+ T, CD8+ T, and B lymphocytes in peripheral blood, no depletion was found after topical administration. Conclusions. The production of kynurenines, in particular 3HK and 3HAA, may be one mechanism (in addition to tryptophan depletion) by which IDO prolongs graft survival. These molecules have potential as specific agents for preventing allograft rejection in patients at high rejection risk.Fight for Sight and the Wellcome Trust

Protein Antigens Increase the Protective Efficacy of a Capsule-Based Vaccine against Staphylococcus aureus in a Rat Model of Osteomyelitis

Staphylococcus aureus is an invasive bacterial pathogen, and antibiotic resistance has impeded adequate control of infections caused by this microbe. Moreover, efforts to prevent human infections with single-component S. aureus vaccines have failed. In this study, we evaluated the protective efficacy in rats of vaccines containing both S. aureus capsular polysaccharides (CPs) and proteins. The serotypes 5 CP (CP5) and 8 CP (CP8) were conjugated to tetanus toxoid and administered to rats alone or together with domain A of clumping factor A (ClfA) or genetically detoxified alpha-toxin (dHla). The vaccines were delivered according to a preventive or a therapeutic regimen, and their protective efficacy was evaluated in a rat model of osteomyelitis. Addition of dHla (but not ClfA) to the CP5 or CP8 vaccine induced reductions in bacterial load and bone morphological changes compared with immunization with either conjugate vaccine alone. Both the prophylactic and therapeutic regimens were protective. Immunization with dHla together with a pneumococcal conjugate vaccine used as a control did not reduce staphylococcal osteomyelitis. The emergence of unencapsulated or small-colony variants during infection was negligible and similar for all of the vaccine groups. In conclusion, addition of dHla to a CP5 or CP8 conjugate vaccine enhanced its efficacy against S. aureus osteomyelitis, indicating that the inclusion of multiple antigens will likely enhance the efficacy of vaccines against both chronic and acute forms of staphylococcal disease.Fil: Lattar, Santiago Martín. LIMEADE: A General Framework for Explanation-Based Human Tuning of Opaque Machine Learners

Research in human-centered AI has shown the benefits of systems that can explain their predictions. Methods that allow humans to tune a model in response to the explanations are similarly useful. While both capabilities are well-developed for transparent learning models (e.g., linear models and GA2Ms), and recent techniques (e.g., LIME and SHAP) can generate explanations for opaque models, no method for tuning opaque models in response to explanations has been user-tested to date. This paper introduces LIMEADE, a general framework for tuning an arbitrary machine learning model based on an explanation of the model's prediction. We demonstrate the generality of our approach with two case studies. First, we successfully utilize LIMEADE for the human tuning of opaque image classifiers. Second, we apply our framework to a neural recommender system for scientific papers on a public website and report on a user study showing that our framework leads to significantly higher perceived user control, trust, and satisfaction. Analyzing 300 user logs from our publicly-deployed website, we uncover a tradeoff between canonical greedy explanations and diverse explanations that better facilitate human tuning.Comment: 16 pages, 7 figure

Solving Linux Upgradeability Problems Using Boolean Optimization

Managing the software complexity of package-based systems can be regarded as one of the main challenges in software architectures. Upgrades are required on a short time basis and systems are expected to be reliable and consistent after that. For each package in the system, a set of dependencies and a set of conflicts have to be taken into account. Although this problem is computationally hard to solve, efficient tools are required. In the best scenario, the solutions provided should also be optimal in order to better fulfill users requirements and expectations. This paper describes two different tools, both based on Boolean satisfiability (SAT), for solving Linux upgradeability problems. The problem instances used in the evaluation of these tools were mainly obtained from real environments, and are subject to two different lexicographic optimization criteria. The developed tools can provide optimal solutions for many of the instances, but a few challenges remain. Moreover, it is our understanding that this problem has many similarities with other configuration problems, and therefore the same techniques can be used in other domains.Comment: In Proceedings LoCoCo 2010, arXiv:1007.083
