    Relação genética entre as cepas de Yersinia pestis isoladas durante epizootia no foco da chapada do Araripe, Pernambuco, Brasil, por MLVA

    A set of twenty strains of Yersinia pestis isolated during the investigation of an outbreak in a focus of the Araripe, municipality of Exu, state of Pernambuco, Brazil, was analyzed for six VNTR (variable number of tandem repeats) loci. The strains, conserved in the bacterioteca SRP (bacterial collection of the Reference Service in Plague), were reactivated and the identification confirmed by bacteriological bacteriophage susceptibility to anti-plague. Changes in the genome were screened for the presence or absence of virulence genes in plasmids prototypical Y. pestis and High-Pathogenicity Island (HPI) of yersinias by multiplex-PCR. The strains proved to be genetically related by MLVA (analysis of multiple loci of variable number of tandem repeats), which reflects the epidemiological relationship of these isolates.Um conjunto de vinte cepas de Yersinia pestis isoladas durante a investigação de uma epizootia em um foco da Chapada do Araripe, município de Exu, Pernambuco, Brasil, foi analisado em seis locos VNTR (número variável de repetições em tandem). As cepas, conservadas na bacterioteca do SRP (Serviço de Referência em Peste), foram reativadas e a identificação bacteriológica confirmada pela suscetibilidade ao bacteriófago antipestoso. Alterações no genoma foram pesquisadas pela presença ou ausência de genes de virulência nos plasmídeos prototípicos da Y. pestis e na Ilha de Alta Patogenicidade (HPI) das yersínias por meio da reação em cadeia da polimerase multiplex (multiplexPCR). As cepas revelaram-se geneticamente relacionadas, por MLVA (análise de múltiplos locos do número variável de repetições em tandem), o que reflete a relação epidemiológica desses isolados

    Apesar de sua fundamentação clínico-epidemiológica, numerosos casos suspeitos de peste nos focos brasileiros têm sido descartados por serem negativos pelo teste de hemaglutinação para detecção de anticorpos contra o antígeno F1 da Yersinia pestis. A transcendência da peste justifica estudar se tais resultados decorrem da falta de resposta ao F1, e se outras proteínas da Yersinia pestis poderiam ser reconhecidas nos soros suspeitos, sendo desta forma candidatas como alvo diagnóstico alternativo ao F1. Assim sendo, cepas de Yersinia pestis e de Yersinia pseudotuberculosis, uma proteína YopH recombinante e a F1 foram utilizadas para analisar soros de pacientes e soros imunes de coelhos. A F1 e a YopH não foram reconhecidas pelos soros humanos HA- e nenhuma proteína majoritária, comum a todos os soros humanos e coelhos, foi identificada, o que permite concluir que os casos suspeitos devem ser submetidos a uma avaliação clínico-laboratorial mais rigorosa, aprofundando a investigação epidemiológica em busca de outras etiologias

    Seroprevalence of hantavirus and Yersinia pestis antibodies in professionals from the Plague Control Program

    Introduction Professionals who handle rodents in the field and in the laboratory are at risk of infection by the microorganisms harbored by these animals. Methods Serum samples from professionals involved in rodent and Yersinia pestis handling in field or laboratory work were analyzed to determine hantavirus and plague seroprevalence and to establish a relationship between these activities and reports of illnesses. Results Two individuals had antibodies against hantavirus, and two harbored antibodies against the plague; none of the individuals had experienced an illness related to their duties. Conclusions These results confirm the risks of hantavirus- and plague-related field and laboratory activities and the importance of protective measures for such work

    Toxicity and Anti-Inflammatory Activity of Phenolic-Rich Extract from Nopalea cochenillifera (Cactaceae): A Preclinical Study on the Prevention of Inflammatory Bowel Diseases

    Phenolic compounds have been scientifically recognized as beneficial to intestinal health. The cactus Nopalea cochenillifera, used as anti-inflammatory in traditional medicine, is a rich source of these bioactive compounds. The present study aimed to investigate the phytochemical profile of N. cochenillifera extract and evaluate its acute toxicity and anti-inflammatory effect on 2,4-dinitrobenzenesulfonic acid (DNBS)-induced colitis in rats. The total phenolic content per gram of dry extract was 67.85 mg. Through HPLC-IES-MSn, a total of 25 compounds such as saccharides, organic acids, phenolic acids and flavonoids were characterized. The dose of 2000 mg/kg of extract by an oral route showed no signs of toxicity, mortality or significant changes in biochemical and hematological parameters. Regarding intestinal anti-inflammatory effects, animals were treated with three different doses of extract or sulfasalazine. Macroscopic analysis of the colon indicated that the extract decreased the disease activity index. Levels of IL-1β and TNF-α decreased, IL-10 increased and MDA and MPO enzyme levels decreased when compared with the control group. In addition, a down-regulation of MAPK1/ERK2 and NF-κB p65 pathway markers in colon tissue was observed. The epithelial integrity was improved according to histopathological and immunohistological analysis. Thus, the extract provided strong preclinical evidence of being effective in maintaining the remission of colitis

    The Pequi fruit (Caryocar Brasiliense cambess), typical of the Brazilian cerrado or savannah, is a source of essential fatty acids, carotenoids, and phenolic compounds. The aim of this study was to analyze the effects of consuming this fruit on anxiety behavior and lipid peroxidation in the brains of rats whose mothers were treated (by gavage) during pregnancy and lactation with Pequi fruit (pulp or nuts) at 2000 mg/kg of body weight. Anxiety parameters were assessed using the open field (OF), elevated plus maze (EPM), and light/dark box (LDB) tests. The brain was removed to measure malondialdehyde (MDA) levels. Data were analyzed using One-way Anova (p < 0.05). In the OF, the animals in the pulp group presented more time spent in the central area (20.37 ± 0.73 vs Control: 12.51 ± 0.39; Nuts: 8.28 ± 0.40) and increased locomotion (159.7 ± 6.10) compared to the other groups (Control: 127.3 ± 5.54; Nuts: 139.08 ± 6.57). In the EPM, the pulp group entered into the open arms (8.57 ± 0.36) and stayed more time in the central area (19.44 ± 1.17) compared to the Nuts group (7.14 ± 0.34; 13.00 ± 1.57). In the LDB the pulp group entered more (8.00 ± 0.42 vs Control: 7.16 ± 0.16 and Nuts: 7.42 ± 0.75) and stayed longer in the clear light side (92.18 ± 6.42) than all the other groups (Control: 71.44 ± 3.53; Nuts: 80.57 ± 6.50), respectively. Pulp group presented lower MDA in the brain (55.34 ± 3.04) compared to Control (72.06 ± 4.66) and Nuts (66.57 ± 2.45). We conclude that Pequi pulp consumption during pregnancy and lactation reduces lipid peroxidation in brain tissue and induces anxiolytic-like behavior in rat offspring. These effects were not observed in the Pequi nuts group
