    Rapid evolutionary turnover underlies conserved lncRNA-genome interactions

    Many long noncoding RNAs (lncRNAs) can regulate chromatin states, but the evolutionary origin and dynamics driving lncRNA-genome interactions are unclear. We adapted an integrative strategy that identifies lncRNA orthologs in different species despite limited sequence similarity, which is applicable to mammalian and insect lncRNAs. Analysis of the roX lncRNAs, which are essential for dosage compensation of the single X chromosome in Drosophila males, revealed 47 new roX orthologs in diverse Drosophilid species across ∼40 million years of evolution. Genetic rescue by roX orthologs and engineered synthetic lncRNAs showed that altering the number of focal, repetitive RNA structures determines roX ortholog function. Genomic occupancy maps of roX RNAs in four species revealed conserved targeting of X chromosome neighborhoods but rapid turnover of individual binding sites. Many new roX-binding sites evolved from DNA encoding a pre-existing RNA splicing signal, effectively linking dosage compensation to transcribed genes. Thus, dynamic change in lncRNAs and their genomic targets underlies conserved and essential lncRNA-genome interactions

    Collaborative Deep Learning for Recommender Systems

    Collaborative filtering (CF) is a successful approach commonly used by many recommender systems. Conventional CF-based methods use the ratings given to items by users as the sole source of information for learning to make recommendation. However, the ratings are often very sparse in many applications, causing CF-based methods to degrade significantly in their recommendation performance. To address this sparsity problem, auxiliary information such as item content information may be utilized. Collaborative topic regression (CTR) is an appealing recent method taking this approach which tightly couples the two components that learn from two different sources of information. Nevertheless, the latent representation learned by CTR may not be very effective when the auxiliary information is very sparse. To address this problem, we generalize recent advances in deep learning from i.i.d. input to non-i.i.d. (CF-based) input and propose in this paper a hierarchical Bayesian model called collaborative deep learning (CDL), which jointly performs deep representation learning for the content information and collaborative filtering for the ratings (feedback) matrix. Extensive experiments on three real-world datasets from different domains show that CDL can significantly advance the state of the art

    High H2 Sorption Energetics in Zeolitic Imidazolate Frameworks

    A combined experimental and theoretical study of H2 sorption was carried out on two isostructural zeolitic imidazolate frameworks ZIFs , namely ZIF 68 and ZIF 69. The former consists of Zn2 ions that are coordinated to two 2 nitroimidazolate and two benzimidazolate linkers in a tetrahedral fashion, while 5 chlorobenzimidazolate is used in place of benzimidazolate in the latter compound. H2 sorption measurements showed that the two ZIFs display similar isotherms and isosteric heats of adsorption Qst . The experimental initial H2 Qst value for both ZIFs was determined to be 8.1 kJ mol 1, which is quite high for materials that do not contain exposed metal centers. Molecular simulations of H2 sorption in ZIF 68 and ZIF 69 confirmed the similar H2 sorption properties between the two ZIFs, but also suggest that H2 sorption is slightly favored in ZIF 68 with regards to uptake at 77 K 1.0 atm. This work also presents inelastic neutron scattering INS spectra for H2 sorbed in ZIFs for the first time. The spectra for ZIF 68 and ZIF 69 show a broad range of intensities starting from about 4 meV. The most favorable H2 sorption site in both ZIFs corresponds to a confined region between two adjacent 2 nitroimidazolate linkers. Two dimensional quantum rotation calculations for H2 sorbed at this site in ZIF 68 and ZIF 69 produced rotational transitions that are in accord with the lowest energy peak observed in the INS spectrum for the respective ZIFs. We found that the primary binding site for H2 in the two ZIFs generates high barriers to rotation for the adsorbed H2, which are greater than those in several metal organic frameworks MOFs which possess open metal sites. H2 sorption was also observed for both ZIFs in the vicinity of the nitro groups of the 2 nitroimidazolate linkers. This study highlights the constructive interplay of experiment and theory to elucidate critical details of the H2 sorption mechanism in these two isostructural ZIF

    RDGB , a PtdIns-PtdOH transfer protein, regulates G-protein-coupled PtdIns(4,5)P2 signalling during Drosophila phototransduction

    Many membrane receptors activate phospholipase C (PLC) during signalling, triggering changes in the levels of several plasma membrane lipids including phosphatidylinositol (PtdIns), phosphatidic acid (PtdOH) and phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2]. It is widely believed that exchange of lipids between the plasma membrane and endoplasmic reticulum (ER) is required to restore lipid homeostasis during PLC signalling, yet the mechanism remains unresolved. RDGBα (hereafter RDGB) is a multi-domain protein with a PtdIns transfer protein (PITP) domain (RDGB-PITPd). We find that, in vitro, the RDGB-PITPd binds and transfers both PtdOH and PtdIns. In Drosophila photoreceptors, which experience high rates of PLC activity, RDGB function is essential for phototransduction. We show that binding of PtdIns to RDGB-PITPd is essential for normal phototransduction; however, this property is insufficient to explain the in vivo function because another Drosophila PITP (encoded by vib) that also binds PtdIns cannot rescue the phenotypes of RDGB deletion. In RDGB mutants, PtdIns(4,5)P2 resynthesis at the plasma membrane following PLC activation is delayed and PtdOH levels elevate. Thus RDGB couples the turnover of both PtdIns and PtdOH, key lipid intermediates during G-protein-coupled PtdIns(4,5)P2 turnover

    Optical monitoring of the z=4.40 quasar Q 2203+292

    We report Cousins R-band monitoring of the high-redshift (z=4.40) radio quiet quasar Q 2203+292 from May 1999 to October 2007. The quasar shows maximum peak-to-peak light curve amplitude of ~0.3 mag during the time of our monitoring, and ~0.9 mag when combined with older literature data. The rms of a fit to the light curve with a constant is 0.08 mag and 0.2 mag, respectively. The detected changes are at ~3-sigma level. The quasar was in a stable state during the recent years and it might have undergone a brightening event in the past. The structure function analysis concluded that the object shows variability properties similar to those of the lower redshift quasars. We set a lower limit to the Q 2203+292 broad line region mass of 0.3-0.4 M_odot. Narrow-band imaging search for redshifted Ly_alpha from other emission line objects at the same redshift shows no emission line objects in the quasar vicinity.Comment: 9 pages, 8 figures, accepted for publication in MNRA

    Adipose-Derived Stem Cells in Crohn's Rectovaginal Fistula

    Therapeutic options for recto-vaginal fistula in the setting of Crohn's disease are limited and many data are available in the literature. The manuscript describes the history of a patient who has been the pioneer of our Clinical Trials in treating this disease in fistulizing Crohn's disease environment. We believe it is the first time that a patient with this disease has been treated by adipose-derived stem cells in allogeneic form. The conclusion of our study with Mary is that the use of mesenchymal stem cells derived from adipose tissue is secure, either in autologous or allogeneic form. Furthermore, we have proved that if we use multi-dose and multiple applications on a patient, it does not produce any adverse effect, which confirms us the safety of using these cells in patients at least in the fistulizing Crohn's disease environment

    p53 suppresses type II endometrial carcinomas in mice and governs endometrial tumour aggressiveness in humans

    Type II endometrial carcinomas are a highly aggressive group of tumour subtypes that are frequently associated with inactivation of the TP53 tumour suppressor gene. We show that mice with endometrium-specific deletion of Trp53 initially exhibited histological changes that are identical to known precursor lesions of type II endometrial carcinomas in humans and later developed carcinomas representing all type II subtypes. The mTORC1 signalling pathway was frequently activated in these precursor lesions and tumours, suggesting a genetic cooperation between this pathway and Trp53 deficiency in tumour initiation. Consistent with this idea, analyses of 521 human endometrial carcinomas identified frequent mTORC1 pathway activation in type I as well as type II endometrial carcinoma subtypes. mTORC1 pathway activation and p53 expression or mutation status each independently predicted poor patient survival. We suggest that molecular alterations in p53 and the mTORC1 pathway play different roles in the initiation of the different endometrial cancer subtypes, but that combined p53 inactivation and mTORC1 pathway activation are unifying pathogenic features among histologically diverse subtypes of late stage aggressive endometrial tumours

    The process of managing students' motor level requires solving tasks, such as measuring and evaluating basic qualities, and is the basis of pedagogical activity. The purpose of the study is to evaluate and compare the level of physical preparedness of newly admitted Bulgarian and foreign students. They chose to practice table tennis or volleyball. As a result of a conducting ascertaining pedagogical experiment, the input level of indicators, such as upper and lower extremity explosive strength, grip strength, abdominal muscle strength, the state of sensory systems and the neuromuscular apparatus, the ability of the spine to fold and etc. The results of the sports-pedagogical testing were obtained using mathematical-statistical processing, through a specialized program IBM "SPSS" 19 and "Excel".
