    Design Features to Accelerate the Higher-Order Assembly of DNA Origami on Membranes

    Nanotechnology often exploits DNA origami nanostructures assembled into even larger superstructures up to micrometer sizes with nanometer shape precision. However, large-scale assembly of such structures is very time-consuming. Here, we investigated the efficiency of superstructure assembly on surfaces using indirect cross-linking through low-complexity connector strands binding staple strand extensions, instead of connector strands binding to scaffold loops. Using single-molecule imaging techniques, including fluorescence microscopy and atomic force microscopy, we show that low sequence complexity connector strands allow formation of DNA origami superstructures on lipid membranes, with an order-of-magnitude enhancement in the assembly speed of superstructures. A number of effects, including suppression of DNA hairpin formation, high local effective binding site concentration, and multivalency are proposed to contribute to the acceleration. Thus, the use of low-complexity sequences for DNA origami higher-order assembly offers a very simple but efficient way of improving throughput in DNA origami design.Published as part of The Journal of Physical Chemistry virtual special issue “W.     Isokinetic profile of Elite Serbian female judoists

    Elite judo athletes undergo vigorous training to achieve outstanding results. In pursuit of achieving competitive success, the occurrence of injuries amongst judo athletes is not rare. The study aimed to perform a knee flexors and extensors isokinetic torque analysis in elite female judo athletes. Fifty\u2010eight elite female judo athletes of the Serbian national team (21.02 \ub1 3.11 years; 62.36 \ub1 11.91 kg, 165.04 \ub1 10.24 cm, training experience 12.72 \ub1 2.98 years) volunteered to participate in this study. The range of motion (ROM) was set at 90\u2070. Testing was performed in a concentric\u2013concentric mode for the testing speed of 60 \u2070/s. Five maximal voluntary contractions of knee extensors and knee flexors muscle groups were measured for both legs. The obtained data showed a statistically significant difference in absolute torque values among different categories as heavier athletes demonstrated higher values. Post hoc analysis showed a significant difference between weight categories, as heavier athletes demonstrated higher values, while no significant differences in normalized torque values for different weight categories were observed. The implementation of new elements and training modalities may improve performance and prevent lateral asymmetry, thus reducing the risk of injury

    Ibrutinib Unmasks Critical Role of Bruton Tyrosine Kinase in Primary CNS Lymphoma.

    Bruton tyrosine kinase (BTK) links the B-cell antigen receptor (BCR) and Toll-like receptors with NF-ÎșB. The role of BTK in primary central nervous system (CNS) lymphoma (PCNSL) is unknown. We performed a phase I clinical trial with ibrutinib, the first-in-class BTK inhibitor, for patients with relapsed or refractory CNS lymphoma. Clinical responses to ibrutinib occurred in 10 of 13 (77%) patients with PCNSL, including five complete responses. The only PCNSL with complete ibrutinib resistance harbored a mutation within the coiled-coil domain of CARD11, a known ibrutinib resistance mechanism. Incomplete tumor responses were associated with mutations in the B-cell antigen receptor-associated protein CD79B

    Midterm follow-up after embolization of intracranial aneurysms proximal to the circle of Willis with the Silk Vista flow diverter: the I-MAMA registry

    Purpose: The aim of this registry was to assess technical success, procedural safety and mid- to long-term follow-up results of the Silk Vista “Mama” (SVM) flow diverter (BALT, Montmorency, France) for the treatment of proximal intracranial aneurysms. Methods: Between August 2020 and March 2022, data from nine Italian neurovascular centres were collected. Data included patients’ clinical presentation, aneurysms’ size, location and status, technical details, overall complications and mid- to long-term angiographic follow-up. Results: Forty-eight aneurysms in 48 patients were treated using the SVM. Most aneurysms were small (≀ 10 mm: no. 29, 60%) and unruptured (no. 31, 65%); 13 aneurysms were recurrent after coiling or clipping. 37/48 aneurysms involved the internal carotid artery (77%). Optimal opening and complete wall apposition of the device were achieved in 46 out of 48 cases (96%). Four intra- or periprocedural complications occurred (two thrombotic complications successfully resolved, one cerebellar ischemia, one perirenal hematoma), without new neurological deficit. No significant intra-stent stenosis or stent displacement was observed during follow-up. No FD-related morbidity nor mortality was reported. At midterm (6–12 months) to long-term (> 12 months) follow-up, complete aneurysm occlusion (OKM D) was achieved in 76% of cases. Eighty-eight percent of patients had complete aneurysm occlusion or entry remnant (OKM D + C). Conclusions: Our experience suggests that the new generation of low-profile SVM flow diverter for the treatment of proximal intracranial aneurysms is safe and effective, with low rates of intraprocedural complications and acceptable mid- to long-term occlusion rate

    Occupational Therapy Intervention with Children Survivors of War

    A preventive occupational therapy program with children surviving the Kosovo conflict is examined. The objective of the program was to facilitate the emotional expression of traumatic experiences in order to prevent the development of future psychological problems. The intervention was based on a community-centred approach with spirituality as a central focus of the intervention.The Model of Human Occupation and the Occupational Performance Process Model were utilized to guide the identification and intervention of occupational performance issues.The children’s return from a land of war to a land of children demonstrates the potential of occupational therapy intervention in this field. With increasing awareness of populations facing social and political challenges, there is a growing importance of the concept of occupational justice and the need to work against occupational apartheid.Cet article dĂ©crit un programme de prĂ©vention en ergothĂ©rapie qui Ă©tait destinĂ© aux enfants ayant survĂ©cu au conflit du Kosovo. L’objectif du programme Ă©tait d’aider les enfants Ă  exprimer les Ă©motions qu’ils avaient ressenties lors d’expĂ©riences traumatiques afin de prĂ©venir l’apparition de problĂšmes psychologiques. L’intervention Ă©tait basĂ©e sur une approche communautaire s’articulant autour de la spiritualitĂ©. Le ModĂšle de l’occupation humaine et le ModĂšle du processus d’intervention dans le rendement occupationnel ont Ă©tĂ© utilisĂ©s pour cibler les difficultĂ©s en matiĂšre de rendement occupationnel et pour dĂ©terminer les interventions requises. Le retour des enfants d’un monde de guerre vers le monde de l’enfance dĂ©montre la possibilitĂ© de proposer une intervention ergothĂ©rapique dans ce domaine.     Internet-based Self-Assessment after the Tsunami: lessons learned

    BACKGROUND: In the aftermath of the Tsunami disaster in 2004, an online psychological self-assessment (ONSET) was developed and made available by the University of Zurich in order to provide an online screening instrument for Tsunami victims to test if they were traumatized and in need of mental health care. The objective of the study was to report the lessons learnt that were made using an Internet-based, self-screening instrument after a large-scale disaster and to discuss its outreach and usefulness. METHODS: Users of the online self-assessment decided after finishing the procedure whether their dataset could be used for quality control and scientific evaluation Their answers were stored anonymously only if they consented (which was the case in 88% of the sample), stratified analyses according to level of exposure were conducted. RESULTS: A total of 2,914 adult users gave their consent for analysis of the screenings. Almost three quarter of the sample filled out the ONSET questionnaire within the first four weeks. Forty-one percent of the users reported direct exposure to the Tsunami disaster. Users who were injured by the Tsunami and users who reported dead or injured family members showed the highest degree of PTSD symptoms. CONCLUSION: ONSET was used by a large number of subjects who thought to be affected by the catastrophe in order to help them decide if they needed to see a mental health professional. Furthermore, men more frequently accessed the instrument than women, indicating that Internet-based testing facilitates reaching out to a different group of people than "ordinary" public mental health strategies
