    Reproductive biology of Clerodendrum splendens (Verbenaceae)

    Clerodendrum splendens (Verbenaceae) is an ornamental climbing shrub. Flowering occur during the period between December – April. Flowers are red, small complete pedicellate, pentamerous, zygomorphic, hermaphrodite and hypogynous. Anthesis occur between 06:00 – 06:30hrs followed by anther dehiscence at 07:00 – 09:00hrs. Stigma become receptive at 11:00 – 14:00hrs. There are 2160 ± 380 pollens/flower. There are 4 ovules/flower and pollen – ovule ratio is 540 : 1. Nearly 2% flower show open pollination. There is no fruitset in bagged inflorescence indicating cross pollinating nature. Fruit and seedset percentage is very low. Xylocopa Eumenes sp. and Componotus campertris (Black ant) are most effective visitors

    Background: Identification of breast cancer stem cells as the chemo-resistant and tumor-initiating population represents an important milestone in approaching anticancer therapies. Targeting this minor subpopulation of chemo- and radio-resistant stem-like cells, termed as the cancer stem cells (CSCs) and their eradication could significantly enhance clinical outcomes. Most of the presently administered chemotherapeutics target the tumor bulk but are ineffective against the CSCs. We report here that diosgenin (DG), a naturally occurring steroidal saponin, could effectively inhibit CSCs from three breast cancer cell lines, MCF7, T47D and MDA-MB-231, by inducing apoptosis and inhibiting the CSC associated phenotypes. Methods: CSCs were enriched in these cells lines, characterized for CSC traits by immunocytochemistry and flow cytometry. Proliferation and apoptosis assays were performed in these breast CSCs in the presence of DG to obtain the inhibitory concentration. Apoptosis was confirmed with gene expression analysis, Western blotting and propidium iodide staining. TCF-LEF reporter assay, sFRP overexpression and RNAi silencing studies were performed to study regulation of the Wnt pathway. Statistical significance was evaluated by a two-sided Student's t-test. Results: Using the TCF-LEF reporter system, we show the effect of DG on CSCs is predominantly through the network regulating CSC self renewal, the Wnt ß-catenin pathway. Specifically, the Wnt antagonist, the secreted frizzled related protein 4, (sFRP4), had a defining role in the action of DG. Gain-of-function of sFRP4 in CSCs could improve the response to DG wherein CSC mediators were inhibited, ß-catenin was down regulated and the effectors of epithelial to mesenchymal transition and pro-invasive markers were repressed. Conversely, the loss-of-function of sFRP4 had a reverse effect on the CSC population which therein became enriched, their response to DG treatment was modest, ß-catenin levels increased, GSK3ß expression decreased and the expression of epithelial markers of CSC was completely abrogated. Conclusion: These findings demonstrate the effect of DG on inhibiting the resilient breast CSCs which could provide a benchmark for the development of DG-based therapies in breast cancer treatment. © 2017 Bhuvanalakshmi, Basappa, Rangappa, Dharmarajan, Sethi, Kumar and Warrier

    Understanding sustainable operation of micro-hydropower:a field study in Nepal

    Off-grid renewable energy technologies are important in improving electricity access for rural communities. However, methods for ensuring their sustainable operation are often poorly understood. In this article, existing approaches for the assessment of off-grid projects are examined. Reliability of the technology, financial viability and community engagement are identified as the 3 key areas governing the sustainability of projects. Focusing on these areas, a methodology is proposed to understand the sustainability of micro-hydropower plants. A mixed-methods approach including a maintenance assessment and interviews with managers, operators and consumers is used to evaluate 24 sites in Nepal. Technically, the results of the study showed that trained operators delivered a higher standard of maintenance, however, technical issues were identified that arise during the design, manufacture and installation phases. The financial viability of plants was aided by charging consumers based on consumption, whilst plants with a higher rated capacity tended to benefit from a larger number of productive end uses. Community engagement was fostered through the local identity of the plant however this was threatened by societal changes. Inherent features of the site, such as rated power and the population density, internal resilience to short-term shock events (failure of components, insufficient collection of tariffs and departure of trained operators) and long-term external development (increased incomes, increased energy consumptions and growth in rural settlements) were found to affect the sustainability of plants

    Prevalence of metabolic syndrome in a rural population- a cross sectional study from Western Uttar Pradesh, India

    Background: Prevalence of non-communicable diseases like hypertension, diabetes mellitus and coronary artery disease is on the rise due to the change in lifestyle, unfavourable dietary habits and obesity. Metabolic syndrome is a simple tool by which we can predict the future risk of diabetes mellitus and cardiovascular disease. Studies showed that prevalence of metabolic syndrome is rising in Indian population, but majority of them were done in urban population. This study was conducted to look into the current status of the metabolic syndrome in rural population.Methods: The study was conducted among a population of 2982. Each participant was subjected to clinical examination, anthropometric measurements and necessary laboratory investigations. Metabolic syndrome was diagnosed based on modified NCEP: ATP III criteria.Results: The prevalence of metabolic syndrome was found to be 11.7% and was higher among female population (13.8%) as compared to males (9.6%). The prevalence of metabolic syndrome increased with increasing age. 28.3% of the participants over the age of 50 years had metabolic syndrome whereas it was only 0.4% below the age of 20 years. Nearly half (47.1%) of the obese individuals were suffering from metabolic syndrome implicating obesity as one of the most important risk factors in the etiopathogenesis of metabolic syndrome. The prevalence was only 1.1% among the underweight group.Conclusions: Present study has shown moderate prevalence of metabolic syndrome among the rural population of Western Uttar Pradesh, India with a more female predisposition

    Targeting the PI3K/Akt signaling pathway in gastric carcinoma: A reality for personalized medicine?

    Frequent activation of phosphatidylinositol-3 kinases (PI3K)/Akt/mTOR signaling pathway in gastric cancer (GC) is gaining immense popularity with identification of mutations and/or amplifications of PIK3CA gene or loss of function of PTEN, a tumor suppressor protein, to name a few; both playing a crucial role in regulating this pathway. These aberrations result in dysregulation of this pathway eventually leading to gastric oncogenesis, hence, there is a need for targeted therapy for more effective anticancer treatment. Several inhibitors are currently in either preclinical or clinical stages for treatment of solid tumors like GC. With so many inhibitors under development, further studies on predictive biomarkers are needed to measure the specificity of any therapeutic intervention. Herein, we review the common dysregulation of PI3K/Akt/mTOR pathway in GC and the various types of single or dual pathway inhibitors under development that might have a superior role in GC treatment. We also summarize the recent developments in identification of predictive biomarkers and propose use of predictive biomarkers to facilitate more personalized cancer therapy with effective PI3K/Akt/mTOR pathway inhibition

    Cost estimation of micro-hydropower equipment in Nepal

    Selecting the appropriate technology for providing electricity to rural communities depends upon evaluating the cost of a potential installation. For some rural communities, locally manufactured technology, in the form of wind and hydropower, can be effective. However, often the cost of these locally manufactured technologies is largely unknown. Access to costing data allows the economic viability of a site to be compared with other options. Furthermore, it enables benchmarking, allowing the expected total cost of an installation, or individual sub-systems, to be compared with quotations. This paper attempts to address the current lack of publicly available costing information for locally manufactured micro-hydropower equipment. A methodology is presented where quotations are provided by micro-hydropower manufacturing companies in Nepal for randomly generated sites. Using that information, they provided a quotation for various sub-systems. This data allows comparison of the cost of major components and the influence of turbine type. Through a linear regression model, expression have been developed that can be used to determine the expected cost for both Pelton and Crossflow turbine installations. The accuracy of these expressions is compared with previous costing models, the outcomes of the work and their significance in the context of Nepal and elsewhere is discussed. The key contribution of this work is establishing numerical expressions which allow proposed costs of micro-hydropower equipment to be rapidly evaluated

    State-specific Prevalence and Factors Associated With Current Marijuana, ENDS, and Cigarette use Among US Adults With Asthma

    Background The use of marijuana (MJ), combustible cigarettes (hereafter cigarettes), and electronic nicotine delivery systems (ENDS) is widespread among United States (US) adults and linked to worsening respiratory symptoms, especially among adults with asthma. This study examined state-specific prevalence and factors associated with MJ, ENDS, and cigarette use among US adults with asthma. Methods We analyzed data of 41 974 adults aged ≥18 years having self-reported current asthma from the 2018 Behavioral Risk Factor Surveillance System (BRFSS). We reported weighted prevalence to account for complex survey design and performed multivariable logistic regression models to examine factors associated with current use of MJ, ENDS, and cigarettes. Results Overall prevalence of current MJ, ENDS, and cigarette use among adults with asthma was 14.5%, 6.6%, and 27.2%, respectively. Our results showed the US states and territories with highest and lowest use prevalence for MJ (California: 23.6% vs Guam: 3.2%), ENDS (Indiana: 12.8% vs North Dakota: 4.0%), and cigarettes (West Virginia: 42.1% vs Guam: 12.3%). Both MJ and ENDS users were more likely to be male, younger, and live in an urban area, but MJ users were more likely and ENDS users less likely to be Non-Hispanic (NH) American Indian/Alaskan Native. Cigarette users were more likely to be older, have at least 1 health condition, and were less likely to be NH Black or Hispanic and college-educated. Conclusion Many US adults with asthma use MJ, ENDS, and cigarettes. Our findings provide insights for clinicians about the urgent need for effective interventions to reduce tobacco and MJ use among adults with asthma

    Microcontroller-based Low-cost Seed Metering Module Retrofit on Cultivator

    Planting is one of critical operations in agriculture. Ground wheel drives conventional metering mechanism, and results in improper placement of seeds because of skidding and vibration of the ground wheel. Electronically equipped seed metering system in the planter have potential to reduces losses associated with friction and wheel slippage. In this study, a retrofit type microcontroller based low-cost seed metering module was developed for commonly available cultivator. Basically, it can be called as mechatronics due to combination of both electronics and mechanical components. Seed metering plate’scell size was designed based on physical properties of black and green gram seeds. The microcontroller controlled motor's speed through Pulse Width Modulation (PWM) based on a signal obtained from a rotary encoder mounted on ground wheel. Under laboratory test, the developed module with a cell size of 5.5 mm found highest quality of feed index of 90 % for green gram and 86 % for black gram when operated at speed of 1.56 km/h. Average plant to plant distance was observed to be 6.73 cm, 6.55 cm for green and black gram during field evaluation. These observed spacing were close to desired spacing (6.28 cm) which intended in the precision metering mechanism. A unit's cost is approximately INR 2000. It facilitates sowing of pulse crop’s seeds without investing extra cost on purchasing separate planter required for it. This retrofit type proposed module will make cultivator as a precision planter suitable for sowing different crops at a very low cost
