    FICD acts bifunctionally to AMPylate and de-AMPylate the endoplasmic reticulum chaperone BiP

    Protein folding homeostasis in the endoplasmic reticulum (ER) is defended by an unfolded protein response that matches ER chaperone capacity to the burden of unfolded proteins. As levels of unfolded proteins decline, a metazoan-specific FIC-domain-containing ER-localized enzyme (FICD) rapidly inactivates the major ER chaperone BiP by AMPylating T518. Here we show that the single catalytic domain of FICD can also release the attached AMP, restoring functionality to BiP. Consistent with a role for endogenous FICD in de-AMPylating BiP, FICD/_{-/-} hamster cells are hypersensitive to introduction of a constitutively AMPylating, de-AMPylation-defective mutant FICD. These opposing activities hinge on a regulatory residue, E234, whose default state renders FICD a constitutive de-AMPylase in vitro\textit{in vitro}. The location of E234 on a conserved regulatory helix and the mutually antagonistic activities of FICD in vivo\textit{in vivo}, suggest a mechanism whereby fluctuating unfolded protein load actively switches FICD from a de-AMPylase to an AMPylase.Supported by Wellcome Trust Principal Research Fellowship to D.R.     Diminishing benefits of urban living for children and adolescents’ growth and development

    AbstractOptimal growth and development in childhood and adolescence is crucial for lifelong health and well-being1–6. Here we used data from 2,325 population-based studies, with measurements of height and weight from 71 million participants, to report the height and body-mass index (BMI) of children and adolescents aged 5–19 years on the basis of rural and urban place of residence in 200 countries and territories from 1990 to 2020. In 1990, children and adolescents residing in cities were taller than their rural counterparts in all but a few high-income countries. By 2020, the urban height advantage became smaller in most countries, and in many high-income western countries it reversed into a small urban-based disadvantage. The exception was for boys in most countries in sub-Saharan Africa and in some countries in Oceania, south Asia and the region of central Asia, Middle East and north Africa. In these countries, successive cohorts of boys from rural places either did not gain height or possibly became shorter, and hence fell further behind their urban peers. The difference between the age-standardized mean BMI of children in urban and rural areas was &lt;1.1 kg m–2 in the vast majority of countries. Within this small range, BMI increased slightly more in cities than in rural areas, except in south Asia, sub-Saharan Africa and some countries in central and eastern Europe. Our results show that in much of the world, the growth and developmental advantages of living in cities have diminished in the twenty-first century, whereas in much of sub-Saharan Africa they have amplified.</jats:p

    Worldwide trends in underweight and obesity from 1990 to 2022: a pooled analysis of 3663 population-representative studies with 222 million children, adolescents, and adults

    Background Underweight and obesity are associated with adverse health outcomes throughout the life course. We estimated the individual and combined prevalence of underweight or thinness and obesity, and their changes, from 1990 to 2022 for adults and school-aged children and adolescents in 200 countries and territories. Methods We used data from 3663 population-based studies with 222 million participants that measured height and weight in representative samples of the general population. We used a Bayesian hierarchical model to estimate trends in the prevalence of different BMI categories, separately for adults (age ≥20 years) and school-aged children and adolescents (age 5–19 years), from 1990 to 2022 for 200 countries and territories. For adults, we report the individual and combined prevalence of underweight (BMI 2 SD above the median). Findings From 1990 to 2022, the combined prevalence of underweight and obesity in adults decreased in 11 countries (6%) for women and 17 (9%) for men with a posterior probability of at least 0·80 that the observed changes were true decreases. The combined prevalence increased in 162 countries (81%) for women and 140 countries (70%) for men with a posterior probability of at least 0·80. In 2022, the combined prevalence of underweight and obesity was highest in island nations in the Caribbean and Polynesia and Micronesia, and countries in the Middle East and north Africa. Obesity prevalence was higher than underweight with posterior probability of at least 0·80 in 177 countries (89%) for women and 145 (73%) for men in 2022, whereas the converse was true in 16 countries (8%) for women, and 39 (20%) for men. From 1990 to 2022, the combined prevalence of thinness and obesity decreased among girls in five countries (3%) and among boys in 15 countries (8%) with a posterior probability of at least 0·80, and increased among girls in 140 countries (70%) and boys in 137 countries (69%) with a posterior probability of at least 0·80. The countries with highest combined prevalence of thinness and obesity in school-aged children and adolescents in 2022 were in Polynesia and Micronesia and the Caribbean for both sexes, and Chile and Qatar for boys. Combined prevalence was also high in some countries in south Asia, such as India and Pakistan, where thinness remained prevalent despite having declined. In 2022, obesity in school-aged children and adolescents was more prevalent than thinness with a posterior probability of at least 0·80 among girls in 133 countries (67%) and boys in 125 countries (63%), whereas the converse was true in 35 countries (18%) and 42 countries (21%), respectively. In almost all countries for both adults and school-aged children and adolescents, the increases in double burden were driven by increases in obesity, and decreases in double burden by declining https://researchonline.ljmu.ac.uk/images/research_banner_face_lab_290.jpgunderweight or thinness. Interpretation The combined burden of underweight and obesity has increased in most countries, driven by an increase in obesity, while underweight and thinness remain prevalent in south Asia and parts of Africa. A healthy nutrition transition that enhances access to nutritious foods is needed to address the remaining burden of underweight while curbing and reversing the increase in obesity

The objective of this study was to evaluate in a real-life setting, the efficacy of intravitreal Ziv-aflibercept in the treatment of DME after the administration of three consecutive monthly doses.Methods: A single arm, prospective cohort study. We included patients with DME who received three doses of intravitreal Ziv-aflibercept. Data such as best corrected visual acuity (BCVA) and tomographic biomarkers before treatment and a month after the third dose were collected. DME was staged using the Panozzo classification.Results: Thirty-eight patients participated for a total of 53 eyes. The mean age was 59 ± 8.1 years. We observed significant changes after the third dose in the parameters studied (BCVA in LogMAR pre-treatment (0.6 ± 0.33) and post-treatment (0.4 ± 0.29) [p< 0.001], macular thickness pre-treatment (501 ± 167 μm) and post-treatment (324 ± 114 μm) [p< 0.001], macular volume pre-treatment 10.8 (7.5– 17.8) mm3 and post-treatment 9.3 (0– 13.6) mm3 [p< 0.005]). And 73.6% of the patients presented an advanced severe stage during their pre-treatment evaluation and after post-treatment, 64.2% of the patients no longer presented edema. No systemic or ocular adverse events occurred.Conclusion: The use of three consecutive monthly doses of intravitreal Ziv-aflibercept in a real-life setting is effective and safe in the management of diabetic macular edema.Keywords: antiangiogenic, diabetes mellitus, intravitreal injection, macular edema, optical coherence tomography, Ziv-aflibercep

    Atmospheric Plasma Supported by TiO2 Catalyst for Decolourisation of Reactive Orange 16 Dye in Water

    Purpose Every advanced oxidation process (AOP) has its limitations in water purification. Novel designs with simultaneous application of different AOPs can offer better solutions for cleaner water. Methods We have comparatively studied two advanced oxidation processes (AOPs) on decolourisation of Reactive Orange 16 (RO 16) azo dye pollutant from water: gas plasma treatment by low power atmospheric pressure plasma using novel plasma needle configuration, and semiconductor heterogeneous photocatalysis using titanium dioxide (TiO2) nanopowders. Additionally, simultaneous application of two advanced oxidation processes on azo dye decolourisation was studied. Results It was found that plasma treatment is very efficient system for the dye removal even for low flow rates (1 slm) of the Ar as feed gas. The presence of 10% of O-2 in Ar flow intensified dye oxidation process and shortened required time for total decolourisation. When plasma and catalyst were simultaneously applied, TiO2 was activated with a few Watts plasma source as well as 300 W UV lamp source. The synergic effect of two AOPs was more pronounced for higher feed gas flow rates, resulting in improved decolourisation efficiency. Conclusion Plasma needle can efficiently remove Reactive Orange 16 azo dye from water with a power consumption of only few Watts. With the addition of TiO2 the removal efficiency is significantly improved. [GRAPHICS]

    Population biology of Trichomycterus sp. (Siluriformes, Trichomycteridae) in Passa Cinco stream, Corumbataí River sub-basin, São Paulo State, southeastern Brazil

    The aim of this work was to characterise the population of Trichomycterus sp. in Passa Cinco stream, regarding length structure, sex ratio, diet and reproductive aspects, in accordance with drought and rainy season periods and longitudinal gradient, as well as to analyse its corporal condition in a temporal dimension. Six samplings were accomplished with the use of electric fishing equipment in three different sites in Passa Cinco stream, contemplating sites of order two, three and four, during the months of May, July, September and November of 2005; and January and March of 2006. Three hundred and forty one individuals were captured, composed of 203 males, 99 females and 39 immatures. The smallest captured individual, an immature, presented 28 mm of standard length and the largest, a male, 85 mm. There was not significant variation in repletion degree and accumulated fat in the visceral cavity of the individuals analysed during the considered periods. Eleven different food items were found in the stomach contents. Considering the whole sampling period, immature Diptera was a preferential item and other items were found as occasionally ingested. Spearman and Friedman tests did not find significant differences in the diet of Trichomycterus sp. in the periods and sampling sites, respectively. The average of absolute fecundity was of 73 oocytes, and the parceled type of spawning was performed. Both males and females of Trichomycterus sp. presented significant differences in their corporal conditions in the considered periods, and in the rainy season, these fishes were in better condition
