    On the viability of a CSO architecture for on-demand virtualized cloud planning and provisioning

    As bandwidth requirements and computing capacity for future applications have been predicted to exceed current network and IT infrastructure capabilities, providers face the need to adapt their provisioning models. This article presents the benefits of Cross Stratum Optimized architectures (provision of network and IT resources in a coordinated way) in support of Cloud-based applications. We also present the architecture's potential impact and benefits for operators, based on MACTOR methodology. MACTOR results show the interactions among value-chain actors and identify their business convergences and divergences, revealing the architecture feasibility

    This paper proposes a next generation ubiquitous converged infrastructure to support Cloud and mobile Cloud computing services. The proposed infrastructure facilitates interconnection of fixed and mobile end users with IT resources through a heterogeneous network integrating optical metro and wireless access networks. To support the Infrastructure as a Service paradigm, the proposed architecture adopts the concept of virtualization across the technology domains involved. To optimally plan the proposed virtual infrastructures a holistic approach considering jointly the presence of all network technology domains and the IT resources is applied. Our modelling results identify trends and trade-offs relating to end-to-end service delay, resource requirements and energy consumption levels of the infrastructure across the various technology domains. © 2013 Authors

    GEYSERS: a novel architecture for virtualization and co-provisioning of dynamic optical networks and IT services

    GEYSERS aims at defining an end-to-end network architecture that offers a novel planning, provisioning and operational framework for optical network and IT infrastructure providers and operators. In this framework, physical infrastructure resources (network and IT) are dynamically partitioned to virtual resources and then composed into a Virtual Infrastructure offered to operators as a service. The operator will be able to operate a virtual infrastructure with an enhanced Network Control Plane (NCP) able to offer coupled, optimized and dynamic on-demand Network and IT provisioning services (i.e., interconnections between multiple IT resources and end-users). The scope and novelty of the GEYSERS architecture is focused on the functional definition and description of two innovative layers, the enhanced Network Control Plane (NCP) and the Logical Infrastructure Composition Layer (LICL). The Service Middleware Layer (SML) and the physical layer (PHY) are part of the overall GEYSERS architecture but are based on existing developments that will be appropriately adapted or extended

    Germline pathogenic variants in BRCA1 confer a high risk of developing breast and ovarian cancer. The BRCA1 exon 11 (formally exon 10) is one of the largest exons and codes for the nuclear localization signals of the corresponding gene product. This exon can be partially or entirely skipped during pre-mRNA splicing, leading to three major in-frame isoforms that are detectable in most cell types and tissue, and in normal and cancer settings. However, it is unclear whether splicing imbalance of this exon is associated with cancer risk. Here we identify a common genetic variant in intron 10, rs5820483 (NC_000017.11:g.43095106_43095108dup), which is associated with exon 11 isoform expression and alternative splicing, and with the risk of breast cancer, but not ovarian cancer, in BRCA1 pathogenic variant carriers. The identification of this genetic effect was confirmed by analogous observations in mouse cells and tissue in which a loxP sequence was inserted in the syntenic intronic region. The prediction that the rs5820483 minor allele variant would create a binding site for the splicing silencer hnRNP A1 was confirmed by pull-down assays. Our data suggest that perturbation of BRCA1 exon 11 splicing modifies the breast cancer risk conferred by pathogenic variants of this gene. This article is protected by copyright. All rights reserved
