    Genotoxicity evaluation of individual cigarette smoke toxicants using the in vitro γH2AX assay by High Content Screening

    AbstractCigarette smoke is a complex mixture consisting of more than 5600 identified chemical constituents of which approximately 150 have been identified so far as “tobacco smoke toxicants”. Proposals made by the World Health Organisation Framework Convention on Tobacco Control mandate the lowering of nine tobacco smoke priority toxicants, including 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), N-nitrosonornicotine (NNN), and benzo[a]pyrene (B[a]P) and monitoring the levels of a further nine including cadmium.Here, we evaluated the genotoxic potential in human bronchial epithelial BEAS-2B cells of four cigarette smoke toxicants; NNK, NNN, B[a]P and cadmium using the novel in vitro γH2AX assay by High Content Screening (HCS). We also examined the genotoxicity of binary mixtures of NNK and NNN reporting their relative contribution to the genotoxic end-point.The results of this preliminary assessment showed that the in vitro γH2AX assay by HCS could be used as a pre-screening tool to detect and quantify the genotoxicity effect of cigarette smoke toxicants individually and in binary mixture. Moreover, the data produced could contribute to the prioritisation of toxicant reduction research in modified tobacco products

    Isoscalar off-shell effects in threshold pion production from pd collisions

    We test the presence of pion-nucleon isoscalar off-shell effects in the pdπ+tpd\to \pi^+ t reaction around the threshold region.     AbstractIn a previous work we reported the molecular characterization of a glutamine synthetase (GS; EC 6.3.1.2.) complementary DNA from a woody plant (Cantón et al. (1993) Plant Mol. Biol. 22, 819–828). The isolated cDNA (pGSP114) encoding a Scots pine (Pinus sylvestris) cytosolic subunit, has been subcloned into the expression vector pET3c to overproduce the GS polypeptide in Escherichia coli cells. The recombinant GS protein showed the same molecular size as a native Scots pine GS subunit. Antibodies against the pET3c-GSP114 encoded protein were raised in rabbits by injecting purified preparations and specificity was determined by immunoprecipitation of GS activity present in pine crude extracts. In spite of the antibodies were able to recognize both cytosolic and chloroplastic GS in tomato plants, they were unable to immunodetect chloroplastic GS in green cotyledons of pine seedlings and cytosolic GS was the unique recognized polypeptide. Unlike to that found in other plant species, cytosolic GS was strongly expressed in green tissues as determined by protein and Northern analysis.     Epidemiology and echinocandin susceptibility of Candida parapsilosis sensu lato species isolated from bloodstream infections at a Spanish University Hospital

    Objectives: The aims of this work were to study the epidemiological profiles, differences in echinocandin susceptibilities and clinical relevance of the Candida parapsilosis sensu lato species isolated from proven fungaemia cases at La Fe University Hospital of Valencia (Spain) from 1995 to 2007.The aims of this work were to study the epidemiological profiles, differences in echinocandin susceptibilities and clinical relevance of the Candida parapsilosis sensu lato species isolated from proven fungaemia cases at La Fe University Hospital of Valencia (Spain) from 1995 to 2007. Results: The prevalence of these species was: C. parapsilosis sensu stricto, 74.4%; Candida orthopsilosis, 23.54%; and Candida metapsilosis, 2.05%. The incidence of the species complex as agents of fungaemia remained stationary until 2005 and doubled in 2006. The incidence of C. orthopsilosis showed an increasing trend during the study period, while C. parapsilosis sensu stricto incidence diminished. Also, an important epidemiological change was observed starting in 2004, when 86.5% of the C. parapsilosis sensu lato strains were found in adult patients, while before that year only 13.5% of the isolates were found in this population. Conclusions: Echinocandin drug susceptibility testing using the CLSI M27-A3 document showed a wide range of MIC values (0.015?4 mg/L), with micafungin being the most potent in vitro inhibitor followed by anidulafungin and caspofungin (MIC geometric mean of 0.68, 0.74 and 0.87 mg/L, respectively). C. metapsilosis was the most susceptible species of the complex to anidulafungin and micafungin in vitro (MIC50 for anidulafungin and micafungin: 0.06 mg/L), while there were no differences between C. parapsilosis sensu lato species when caspofungin MIC50s were compared (MIC50 1.00 mg/L).     A Wearable Textile 3D Gesture Recognition Sensor Based on Screen-Printing Technology

    [EN] Research has developed various solutions in order for computers to recognize hand gestures in the context of human machine interface (HMI). The design of a successful hand gesture recognition system must address functionality and usability. The gesture recognition market has evolved from touchpads to touchless sensors, which do not need direct contact. Their application in textiles ranges from the field of medical environments to smart home applications and the automotive industry. In this paper, a textile capacitive touchless sensor has been developed by using screen-printing technology. Two different designs were developed to obtain the best configuration, obtaining good results in both cases.     Pathology of cattle experimentally intoxicated with ground Ricinus communis seeds

    Five Aberdeen Angus calves were inoculated intra-ruminally with ground seeds of Ricinus communis at doses of 1, 1.5, 2 or 3 gr per kg of body weight, or with saline solution (control), respectively. Grossly, all intoxicated animals showed hemorrhages in abdominal serosas, epicardium, endocardium, spleen, pre-stomachs, abomasum, and small and large intestine, and diffuse edema of the ruminal mucosa. Microscopically, in all animals inoculated with R. communis seeds, the main feature was the presence of pyknotic and karyorrhectic nuclei in the endothelium of central nervous system, hepatic, ruminal, intestinal, glomerular and alveolar capillaries, and in lymphoid cells of multiple organs. Apoptosis, confirmed by activated caspase-3 immunohistochemistry, was observed in these cells. No gross or microscopic lesions were observed in the control animal. The results of this study suggest that apoptosis is the main mechanism of cell death in cattle intoxicated with R. communis seeds.EEA BalcarceFil: Marin, Raúl.     Retrospective analysis of cattle poisoning in Argentina (2000-2013)

    A retrospective analysis (2000 to 2013) of cattle poisoning caused by toxic plants and other compounds was carried out in the Pampas region of Argentina by the Animal Health Group of INTA-EEA, Balcarce. During this period, 1263 reports of diseases of different etiologies (infectious, parasitic, toxic, metabolic and miscellaneous) were recorded in cattle, by collecting anamnestic, clinical and pathological information. A toxic etiology was diagnosed in 21.1% of these reports. Iatrogenic poisoning caused by ionophores was the most frequently recorded etiology. Consumption of toxic plants (Wedelia glauca, Solanum glaucophyllum, among others), mycotoxins (Claviceps purpurea, Claviceps paspali, Epichloë coenophiala, among others), and plants producing cyanide and nitrates/nitrites were also commonly diagnosed. The high frequency of toxic episodes and the difficulties in their diagnosis by practitioners in our livestock production systems emphasizes the importance of this report.EEA BalcarceFil: García, Juan Agustín.
