    Surveillance studies of Lymphocystis disease virus in farmed gilthead seabream (Sparus aurata) by real-time PCR

    Lymphocystis disease (LCD) is the main viral infection reported to affect cultured gilthead seabream (Sparus aurata) in Southern Atlantic and Mediterranean aquaculture. Its etiological agent is the Lymphocystis disease virus (LCDV), a member of the family Iridoviridae (genus Lymphocystivirus). The only adequate measures for LCD prevention in the aquaculture systems are general prophylactic practices, such as the control of fish to be introduced in the farm facilities in order to detect carrier fish. These animals may pose a risk for the introduction of LCDV in fish farms, as direct contact between fish specimens is considered the main route of LCDV spreading. More recently, asymptomatic carrier breeders, as well as virus contaminated-live food, have been involved in LCDV transmission to fish larvae. The detection of subclinical viral infections in carrier fish requires the use of sensitive diagnostic methods. In this context, the objective of this study was to establish the applicability of a real-time PCR assay for LCDV diagnosis in surveillance studies. In addition, the assay has been evaluated with samples from a gilthead seabream hatchery, in order to prove its utility to trace the origin of LCDV in fish farms. Juvenile fish were collected at four farms with different background regarding to LCD. LCDV was detected in all farms, and 30 to 100% of fish were identified as LCDV-infected. Estimated viral load in caudal fin of asymptomatic fish was two to five orders of magnitude lower than in diseased fish. Carrier fish were also identified in the broodstock from a farm with LCD records by analysing caudal fin samples by qPCR. In this farm, the q-PCR assay developed in this study allowed the quantitative detection of LCDV in all samples collected in the hatchery, including fertilized eggs, larvae and fingerlings, and also rotifer cultures and artemia metanauplii and cysts used for larval rearing.Universidad de Málaga.     Lymphocystis disease virus (LCDV) is the etiological agent of lymphocystis disease (LCD), a pathology that affects a wide variety of fish species. Data about LCDV pathogenesis are very short, and mainly limited to histopathological studies of skin lesions. Recent studies on viral genome detection (both by PCR or DNA-DNA in situ hybridization) suggest that LCDV establish a systemic and persistent infection in gilthead seabream (Sparus aurata), but further studies are necessary to prove if this infection is productive or not. In the present study viral quantification and viral mRNA detection (by qPCR and RT-qPCR) have been used to investigate LCDV multiplication in different organs of juvenile gilthead seabream. In addition, a histopathological study was carried out. Animals were collected from two commercial farms in Southwestern Spain. In one farm, where no LCD outbreaks have been recorded, apparently healthy fish were collected, whereas in the other farm, diseased and recovered (two months after LCD symptoms disappearance) fish were sampled. All the animals were LCDV-infected, and viral gene expression was detected in every organ analysed (caudal fin, intestine, liver, spleen, kidney and brain). In asymptomatic animals, both apparently healthy and recovered, a low-titre infection was observed, with the highest viral copy numbers detected in brain and kidney. In diseased fish, viral loads were significantly higher than in subclinical infected animals, being maximal in caudal fin, where lymphocysts were present in the dermis. Different histological alterations were observed in the internal organs from diseased fish analysed, although no hypertrophied cells were detected in any of them. In recovered fish, most of the organs examined presented similar histological features to those in healthy animals. Thus, pathological changes were only detected in the intestine and liver, although they were less severe than those observed in diseased fish. The results presented showed that LCDV establishes a systemic infection in juvenile gilthead seabream, which can be subclinical. In addition, although the disease is self-limiting, the virus is not removed after disease recovery, but produces a persistent infection.Universidad de Málaga.     Non-equilibrium Goldstone phenomenon in tachyonic preheating

    The dominance of the direct production of elementary Goldstone waves is demonstrated in tachyonic preheating by numerically determining the evolution of the dispersion relation, the equation of state and the kinetic power spectra for the angular degree of freedom of the complex matter field. The importance of the domain structure in the order parameter distribution for the quantitative understanding of the excitation mechanism is emphasized.     Large difference in carbon emission - burial balances between boreal and arctic lakes

    Lakes play an important role in the global carbon (C) cycle by burying C in sediments and emitting CO2 and CH4 to the atmosphere. The strengths and control of these fundamentally different pathways are therefore of interest when assessing the continental C balance and its response to environmental change. In this study, based on new high-resolution estimates in combination with literature data, we show that annual emission:burial ratios are generally ten times higher in boreal compared to subarctic – arctic lakes. These results suggest major differences in lake C cycling between biomes, as lakes in warmer boreal regions emit more and store relatively less C than lakes in colder arctic regions. Such effects are of major importance for understanding climatic feedbacks on the continental C sink – source function at high latitudes. If predictions of global warming and northward expansion of the boreal biome are correct, it is likely that increasing C emissions from high latitude lakes will partly counteract the presumed increasing terrestrial C sink capacity at high latitudes

    Expansion for the solutions of the Bogomolny equations on the torus

    We show that the solutions of the Bogomolny equations for the Abelian Higgs model on a two-dimensional torus, can be expanded in powers of a quantity epsilon measuring the departure of the area from the critical area. This allows a precise determination of the shape of the solutions for all magnetic fluxes and arbitrary position of the Higgs field zeroes. The expansion is carried out to 51 orders for a couple of representative cases, including the unit flux case. We analyse the behaviour of the expansion in the limit of large areas, in which case the solutions approach those on the plane.     Dielectric (p,q) Strings in a Throat

    We calculate the (p,q) string spectrum in a warped deformed conifold using the dielectric brane method. The spectrum is shown to have the same functional form as in the dual picture of a wrapped D3-brane with electric and magnetic fluxes on its world volume. The agreement is exact in the limit where q is large. We also calculate the dielectric spectrum in the S-dual picture. The spectrum in the S-dual picture has the same form as in the original picture but it is not exactly S-dual invariant due to an interchange of Casimirs of the non-Abelian gauge symmetries. We argue that in order to restore S-duality invariance the non-Abelian brane action should be refined, probably by a better prescription for the non-Abelian trace operation

    Dynamics of tachyonic preheating after hybrid inflation

    We study the instability of a scalar field at the end of hybrid inflation, using both analytical techniques and numerical simulations. We improve previous studies by taking the inflaton field fully into account, and show that the range of unstable modes depends sensitively on the velocity of the inflaton field, and thereby on the Hubble rate, at the end of inflation. If topological defects are formed, their number density is determined by the shortest unstable wavelength. Finally, we show that the oscillations of the inflaton field amplify the inhomogeneities in the energy density, leading to local symmetry restoration and faster thermalization. We believe this explains why tachyonic preheating is so effective in transferring energy away from the inflaton zero mode.Comment: 12 pages, 10 figures, REVTeX. Minor changes, some references added.
