    The genus Sternbergia Waldst. & Kit. (Amaryllidaceae) in Italy. Contribution to the cytotaxonomical and morpho-anatomical knowledge

    Abstract Morpho-anatomical and karyologial studies were carried out on plants from S Italy of the three Italian species of the genus Sternbergia: S. colchiciflora, S. lutea and S. sicula. The morpho-anatomical features of the leaves are good taxonomic markers, especially concerning their colour, general shape, width and morphology of the median adaxial keel. Karyological analysis revealed the following chromosome complements: Sternbergia colchiciflora, 2n = 20; Sternbergia lutea, 2n = 22; Sternbergia sicula, 2n = 22. Karyotype variability is discussed in the light of previous literature. According to our results S. colchiciflora is confirmed to be the most isolated species, while S. lutea and S. sicula, easily recognizable on morphological grounds, appear instead very close karyologically

    How much do needlestick injuries cost? a systematic review of the economic evaluations of needlestick and sharps injuries among healthcare personnel

To provide an overview of the economic aspects of needlestick and sharps injury (NSI) management among healthcare personnel (HCP) within a Health Technology Assessment project to evaluate the impact of safety-engineered devices on health care methods. A systematic review of economic analyses related to NSIs was performed in accordance with the PRISMA statement and by searching PubMed and Scopus databases (January 1997–February 2015). Mean costs were stratified by study approach (modeling or data driven) and type of cost (direct or indirect). Costs were evaluated using the CDC operative definition and converted to 2015 International US dollars (Int).results.Atotalof14studieswereretrieved:8data−drivenstudiesand6modelingstudies.Amongthem,11studiesprovideddirectandindirectcostsand3studiesprovidedonlydirectcosts.Themedianofthemeansforaggregate(direct+indirect)costswasInt). results. A total of 14 studies were retrieved: 8 data-driven studies and 6 modeling studies. Among them, 11 studies provided direct and indirect costs and 3 studies provided only direct costs. The median of the means for aggregate (direct + indirect) costs was Int747 (range, Int199–Int199–Int1,691). The medians of the means for disaggregated costs were Int425(range,Int425 (range, Int48–Int1,516)fordirectcosts(9studies)andInt1,516) for direct costs (9 studies) and Int322 (range, Int152–Int152–Int 413) for indirect costs (6 studies). When compared with data-driven studies, modeling studies had higher disaggregated and aggregated costs, but data-driven studies showed greater variability. Indirect costs were consistent between studies, mostly referring to lost productivity, while direct costs varied widely within and between studies according to source infectivity, HCP susceptibility, and post-exposure diagnostic and prophylactic protocols. Costs of treating infections were not included, and intangible costs could equal those associated with NSI medical evaluations. conclusions. NSIs generate significant direct, indirect, potential, and intangible costs, possibly increasing over time. Economic efforts directed at preventing occupational exposures and infections, including provision of safety-engineered devices, may be offset by the savings from a lower incidence of NSIs

    LOCAL ANAESTHETIC ACTIVITY OF (+) AND (-) MENTHOL

    In this work we studied the local anaesthetic activity of (+)- and (-)-menthol, a substance used after topical application to induce a feeling of coolness. We compared its activity to two chemically related compounds thymol and (-)-menthone. Anaesthetic activity was evaluated in vivo in the rabbit conjunctival reflex test and in vitro in a rat phrenic nerve hemidiaphragm preparation. Both enatiomers of menthol (10(-4)-1 mug/ml), but not thymol and (-)-menthone, were able to drastically reduce, in a dose-dependent manner, the electrically evoked contractions of rat phrenic hemidiaphragm. In the rabbit: conjunctival reflex test, treatment with a solution of (+)- and (-)menthol (30-100 mug/ml) allowed a dose-dependent increase in the number of stimuli necessary to provoke the reflex, thus confirming in vivo the local anaesthetic activity observed in vitro. Similar to the in vitro results, thymol and (-)-menthone were ineffective also in the in vivo test. In conclusion, these data evidence the local anaesthetic activity of menthol, which appears to be strictly dependent on its chemical structure

    Neuronal Spike Shapes (NSS): A straightforward approach to investigate heterogeneity in neuronal excitability states

    The mammalian brain exhibits a remarkable diversity of neurons, contributing to its intricate architecture and functional complexity. The analysis of multimodal single-cell datasets enables the investigation of cell types and states heterogeneity. In this study, we introduce the Neuronal Spike Shapes (NSS), a straightforward approach for the exploration of excitability states of neurons based on their Action Potential (AP) waveforms. The NSS method describes the AP waveform based on a triangular representation complemented by a set of derived electrophysiological (EP) features. To support this hypothesis, we validate the proposed approach on two datasets of murine cortical neurons, focusing it on GABAergic neurons. The validation process involves a combination of NSS-based clustering analysis, features exploration, Differential Expression (DE), and Gene Ontology (GO) enrichment analysis. Results show that the NSS-based analysis captures neuronal excitability states that possess biological relevance independently of cell subtype. In particular, Neuronal Spike Shapes (NSS) captures, among others, a well-characterized fast-spiking excitability state, supported by both electrophysiological and transcriptomic validation. Gene Ontology Enrichment Analysis reveals voltage-gated potassium (K+) channels as specific markers of the identified NSS partitions. This finding strongly corroborates the biological relevance of NSS partitions as excitability states, as the expression of voltage-gated K+ channels regulates the hyperpolarization phase of the AP, being directly implicated in the regulation of neuronal excitabilit

    MORPHOLOGICAL ANALYSIS OF ANCIENT GRAPE SEEDS FROM A SINK IN THE MIDDLE-AGE TOWN OF PALERMO

    The archaeological excavations in Piazza della Vittoria, in the Roman-Middle Age town of Palermo (Sicily) put in light a sink 3.20 m deep and 1 square m. large, partially filled by thin organic sediments. Grape seeds (grape-stones), fish scales and few vertebrate bones have been found in specific strata sealed under a stratum chronologically attributed to Islamic Middle-Age period (a post-quem limit). The finding of well preserved grape seeds is peculiar and their study opens the opportunity to improve the actual knowledge about evolution, cultivation, use and trade of Vitis L. in the Mediterranean area. This preliminary work focuses on morphologic and morphometric analysis of the ancient grape seeds with two aims: i) systematically describe the remains collection and, ii) define seeds typology and a consequent morphotaxonomic attribution. Over 200 seeds have been carefully dry cleaned (soft brush), photographed and analyzed for total breadth (B), total length (L) and length of stalk (LS) parameters, the most efficient for typological attribution (1, 2); Stummer index has been also calculated (1). Apical notch length (AN) has been for the first time evaluated. Measurements on digital images have been performed using ImageJ 1.31 platform; morphological parameters have been assembled in a dedicated database. Descriptive analysis and linear correlations have been performed using SYSTAT 10. Analysis of variance (ANOVA) and Tukey’s HSD (5% level of significance, α = 0.05) have been applied. All the parameters approximate a normal distribution. Major variation has been observed in LS (c.v. = 35.6%) and AN (c.v. = 35.6%), while B and L showed a c.v. of 9.5% and 12.6% respectively. All the analyzed parameters behave as independent variables with the exception of a significant correlation between Stummer index and L (R2 = 0.45; y = 8.17-0.047x with y = L and x = Stummer index). This correlation reveals that Stummer index depends more from the L and not from the B parameter. On the base of LS measures three subgroups have been arbitrarily created in relationship with the LS: LS1 0.90 mm (35 seeds). Analyzing together the LS groups toward AN, we have found a proportional and significant correlation (p = 0.05) between the extremes LS1 and LS3. In the entire collection, Stummer index varies from 55.76 to 100.86; in the LS groups, the range is 68.38-97.87 in LS1, 61.02-100.86 in LS2 and 55.76-81.70 in LS3. A small group (17) of seeds has been excluded for the impossibility to measure the stalk. The analyzed ancient grape seeds show a wide range of variability for all the considered parameters, revealing a polymorphic collection. In general, the seeds have a rounded heart-like shape, with a noticeable pointy stalk and a very invaginated apical notch. This typical shape is more marked in LS3 group. On the base of LS measures, LS1 is ascribable to wild grapevines, while LS2 and LS3 seem to be ascribable neither to wild nor to cultivated autochthonous Vitis. Furthermore, these seeds differ from those already described in other archaeological horizons in Italy (3) and in France (2). The Stummer index varies highly, exceeding the known range of wild Vitis vinifera (4), although values close to 100 have been already found in wild grapevines in Spain (5) and values above 80 have been also described in Extra-European Vitis species (6).     Memory dysfunction associated with aging, neurodegenerative and psychiatric disorders represents an increasing medical need. Advances in research exploring the biological mechanisms underlying learning and memory have opened new potential approaches for development of memory-enhancing therapies addressed to selective neuronal targets. In this work, we synthesized some derivatives with a pyrazolo[5,1-c][1,2,4]benzotriazine core to identify ligands on GABA A receptors subtype (benzodiazepine site on GABAA- receptor) endowed with the potential of enhancing cognition activity without the side effects usually associated with non-selective GABAA modulators. In fact, there is much evidence that GABAA-R (γ-aminobutyric acid, type A receptor) subtype ligands have relevance in learning and memory. In vitro and in vivo tests have been performed. Pharmacological data indicate that compounds 7, 13, 14 and 22 act as dual functional modulators of GABA A-Rs (promnemonic and anxiolytic agents) while only compounds 3 and 10 stand out as selectively displaying good antiamnesic and procognitive activity (1 and 3 mg/kg, respectively). © 2013 Elsevier Ltd.
