The Disequilibrium of Nucleosomes Distribution along Chromosomes Plays a Functional and Evolutionarily Role in Regulating Gene Expression
To further understand the relationship between nucleosome-space occupancy (NO) and global transcriptional activity in mammals, we acquired a set of genome-wide nucleosome distribution and transcriptome data from the mouse cerebrum and testis based on ChIP (H3)-seq and RNA-seq, respectively. We identified a nearly consistent NO patterns among three mouse tissuesâcerebrum, testis, and ESCsâand found, through clustering analysis for transcriptional activation, that the NO variations among chromosomes are closely associated with distinct expression levels between house-keeping (HK) genes and tissue-specific (TS) genes. Both TS and HK genes form clusters albeit the obvious majority. This feature implies that NO patterns, i.e. nucleosome binding and clustering, are coupled with gene clustering that may be functionally and evolutionarily conserved in regulating gene expression among different cell types

The sustainability of a process is the objective of modern industries aiming to reduce waste in production, since consumers require high quality and efficiency with fair price. Thus, a good understanding of the process should be its starting point. The manufacture of dental implants is an example in which waste reduction is important for the reduction of prices due to the demand for great quality and accuracy. This study observed the behaviour of sustainable micro-cutting applied to the Ti-6Al-7Nb titanium alloy, considering the ploughing effect on minimum quantity lubrication (MQL) and high-speed machining (HSM) conditions. When compared with dry condition and low-speed cutting in orthogonal micro-cutting, the use of HSM in dry cutting was more efficient than using MQL. Calmodulin mutations and life-threatening cardiac arrhythmias: insights from the International Calmodulinopathy Registry.
AIMS: Calmodulinopathies are rare life-threatening arrhythmia syndromes which affect mostly young individuals and are, caused by mutations in any of the three genes (CALM 1-3) that encode identical calmodulin proteins. We established the International Calmodulinopathy Registry (ICalmR) to understand the natural history, clinical features, and response to therapy of patients with a CALM-mediated arrhythmia syndrome. METHODS AND RESULTS: A dedicated Case Report File was created to collect demographic, clinical, and genetic information. ICalmR has enrolled 74 subjects, with a variant in the CALM1 (nâ=â36), CALM2 (nâ=â23), or CALM3 (nâ=â15) genes. Sixty-four (86.5%) were symptomatic and the 10-year cumulative mortality was 27%. The two prevalent phenotypes are long QT syndrome (LQTS; CALM-LQTS, nâ=â36, 49%) and catecholaminergic polymorphic ventricular tachycardia (CPVT; CALM-CPVT, nâ=â21, 28%). CALM-LQTS patients have extremely prolonged QTc intervals (594â±â73âms), high prevalence (78%) of life-threatening arrhythmias with median age at onset of 1.5âyears [interquartile range (IQR) 0.1-5.5âyears] and poor response to therapies. Most electrocardiograms (ECGs) show late onset peaked T waves. All CALM-CPVT patients were symptomatic with median age of onset of 6.0âyears (IQR 3.0-8.5âyears). Basal ECG frequently shows prominent U waves. Other CALM-related phenotypes are idiopathic ventricular fibrillation (IVF, nâ=â7), sudden unexplained death (SUD, nâ=â4), overlapping features of CPVT/LQTS (nâ=â3), and predominant neurological phenotype (nâ=â1). Cardiac structural abnormalities and neurological features were present in 18 and 13 patients, respectively. CONCLUSION: Calmodulinopathies are largely characterized by adrenergically-induced life-threatening arrhythmias. Available therapies are disquietingly insufficient, especially in CALM-LQTS. Combination therapy with drugs, sympathectomy, and devices should be considered
