    The yields of r-process elements and chemical evolution of the Galaxy

    The supernova yields of r-process elements are obtained as a function of the mass of their progenitor stars from the abundance patterns of extremely metal-poor stars on the left-side [Ba/Mg]-[Mg/H] boundary with a procedure proposed by Tsujimoto and Shigeyama. The ejected masses of r-process elements associated with stars of progenitor mass Mms18MM_{ms}\leq18M_{\odot} are infertile sources and the SNe II with 20MMms40MM_{\odot}\leq M_{ms}\leq 40M_{\odot}are the dominant source of r-process nucleosynthesis in the Galaxy. The ratio of these stars 20MMms40MM_{\odot}\leq M_{ms}\leq40M_{\odot} with compared to the all massive stars is about \sim18%. In this paper, we present a simple model that describes a star's [r/Fe] in terms of the nucleosynthesis yields of r-process elements and the number of SN II explosions.     A simple, sensitive and selective quantum-dot-based western blot method for the simultaneous detection of multiple targets from cell lysates

    Quantum dots (Qdots) are fluorescent nanoparticles that have great potential as detection agents in biological applications. Their optical properties, including photostability and narrow, symmetrical emission bands with large Stokes shifts, and the potential for multiplexing of many different colours, give them significant advantages over traditionally used fluorescent dyes. Here, we report the straightforward generation of stable, covalent quantum dot-protein A/G bioconjugates that will be able to bind to almost any IgG antibody, and therefore can be used in many applications. An additional advantage is that the requirement for a secondary antibody is removed, simplifying experimental design. To demonstrate their use, we show their application in multiplexed western blotting. The sensitivity of Qdot conjugates is found to be superior to fluorescent dyes, and comparable to, or potentially better than, enhanced chemiluminescence. We show a true biological validation using a four-colour multiplexed western blot against a complex cell lysate background, and have significantly improved previously reported non-specific binding of the Qdots to cellular proteins

    The value of shifting cultivation for biodiversity in Northeast India

    Aim Shifting cultivation is a widespread land-use in many tropical countries that also harbours significant levels of biodiversity. Increasing frequency of cultivation cycles and expansion into old-growth forests have intensified the impacts of shifting cultivation on biodiversity and carbon sequestration. We assessed how bird diversity responds to shifting cultivation and the potential for co-benefits for both biodiversity and carbon in such landscapes to inform carbon-based payments for ecosystem service (PES) schemes. Location Nagaland, Northeast India. Methods We surveyed above-ground carbon stocks and bird communities across various stages of a shifting cultivation system and old-growth forest using composite carbon sampling plots and repeated point counts directly overlaying the carbon plots in both summer and winter. We assessed species diversity using species accumulation and rarefaction curves based on Hill numbers. We fitted a linear mixed-effect model to assess the relationship between species richness and fallow age. We also examined possible co-benefits between carbon and biodiversity from fallow regeneration in terms of relative community similarity to old-growth forest across carbons stocks. Results Farmland and secondary forests regenerating on fallowed land had similar bird species richness to old-growth forests in summer and relatively higher species richness in winter. Within regenerating fallows, we did not find any strong evidence that fallow age influenced bird species richness. Bird community resemblance to old-growth forest increased with secondary forest maturity, correlating also with carbon stocks in summer. However, bird community assemblage did not show a strong association with habitat types and carbon stocks during winter. Main conclusions This study underscores the important role of traditional non-intensive shifting cultivation in providing refuges for biodiversity within heterogeneous habitat mosaics. Effectively managing these landscapes is crucial for both biodiversity conservation and carbon sequestration in the subtropics

    Pathway level subtyping identifies a slow-cycling biological phenotype associated with poor clinical outcomes in colorectal cancer

    Molecular stratification using gene-level transcriptional data has identified subtypes with distinctive genotypic and phenotypic traits, as exemplified by the consensus molecular subtypes (CMS) in colorectal cancer (CRC). Here, rather than gene-level data, we make use of gene ontology and biological activation state information for initial molecular class discovery. In doing so, we defined three pathway-derived subtypes (PDS) in CRC: PDS1 tumors, which are canonical/LGR5+ stem-rich, highly proliferative and display good prognosis; PDS2 tumors, which are regenerative/ANXA1+ stem-rich, with elevated stromal and immune tumor microenvironmental lineages; and PDS3 tumors, which represent a previously overlooked slow-cycling subset of tumors within CMS2 with reduced stem populations and increased differentiated lineages, particularly enterocytes and enteroendocrine cells, yet display the worst prognosis in locally advanced disease. These PDS3 phenotypic traits are evident across numerous bulk and single-cell datasets, and demark a series of subtle biological states that are currently under-represented in pre-clinical models and are not identified using existing subtyping classifiers
