This paper presents descriptions of four information-system interface projects in progress at ESRIN, each demonstrating a somewhat different approach to interface design, but ali sharing the commonality of responding to user goals, tasks and characteristics. It is suggested that next-generation scientific information systems will have to be designed for direct access by end users to a large variety of information sources, through a commom interface. Design of such systems, including their interfaces, should be based on a multi-level analysis of user goals, tasks and domain views.

Computational modeling and constructal design theory applied to the geometric optimization of thin steel plates with stiffeners subjected to uniform transverse load

    No full text
Stiffened thin steel plates are structures widely employed in aeronautical, civil, naval, and offshore engineering. Considering a practical application where a transverse uniform load acts on a simply supported stiffened steel plate, an approach associating computational modeling, Constructal Design method, and Exhaustive Search technique was employed aiming to minimize the central deflections of these plates. To do so, a non-stiffened plate was adopted as reference from which all studied stiffened plate\u2019s geometries were originated by the transformation of a certain amount of steel of its thickness into longitudinal and transverse stiffeners. Different values for the stiffeners volume fraction (\u3c6) were analyzed, representing the ratio between the volume of the stiffeners\u2019 material and the total volume of the reference plate. Besides, the number of longitudinal (Nls) and transverse (Nts) stiffeners and the aspect ratio of stiffeners shape (hs/ts, being hs and ts, respectively, the height and thickness of stiffeners) were considered as degrees of freedom. The optimized plates were determined for all studied \u3c6 values and showed a deflection reduction of over 90% in comparison with the reference plate. Lastly, the influence of the \u3c6 parameter regarding the optimized plates was evaluated defining a configuration with the best structural performance among all analyzed cases

Rational design to control the trade-off between receptor affinity and cooperativity

    No full text
Cooperativity enhances the responsiveness of biomolecular receptors to small changes in the concentration of their target ligand, albeit with a concomitant reduction in affinity. The binding midpoint of a two-site receptor with a Hill coefficient of 1.9, for example, must be at least. 19 times higher than the dissociation constant of the higher affinity of its two binding sites. This trade-off can be overcome, however, by the extra binding energy provided by the addition of more binding sites, which can be used to achieve highly cooperative receptors that still retain high affinity. Exploring this experimentally, we have employed an "intrinsic disorder" mechanism to design two cooperative, three-binding-site receptors starting from a single-site-and thus noncooperative-doxorubicin-binding aptamer. The first receptor follows a binding energy landscape that partitions the energy provided by the additional binding event to favor affinity, achieving a Hill coeffident of 1.9 but affinity within a factor of 2 of the parent aptamer. The binding energy landscape of the second receptor, in contrast, partitions more of this energy toward cooperativity, achieving a Hill coefficient of 23, but at the cost of 4-fold poorer affinity than that of the parent aptamer. The switch between these two behaviors is driven primarily by the affinity of the receptors' second binding event, which serves as an allosteric "gatekeeper" defining the extent to which the system is weighted toward higher cooperativity or higher affinity

Exposure to flaxseed during lactation does not alter prostate area or epithelium height but changes lipid profile in rats Exposici贸n a la linaza durante la lactaci贸n no cambia el 谩rea o altura epitelial de la pr贸stata pero cambia el perfil lip铆dico en ratones

Flaxseed intake has increased owing to beneficial effects to health and prevention of diseases. Provided that it's an important source of lignan, a phytoestrogen, the present study aimed at evaluating the possible effect of the intake of this seed during lactation upon prostate, sexual hormones and lipidic profile of the offspring in adult life. Material and methods: 16 female Wistar rats were used. After delivery, they were divided into two different groups to receive one of the following diets during lactation: Control group (CG), with a casein based diet and Flaxseed group (FG), with a flaxseed based diet containing 25% flaxseed. At weaning, male pups received commercial chow until adult life (170 days old), when they were sacrificed. Results: No differences were perceived concerning offspring food intake and body weight at 170 days. There was a reduction in total cholesterol levels (FG = 45.71 ± 8.96 mg/dL; CG = 63.43 ± 15.69 mg/dL, p = 0.02) and triglycerides (FG = 54.29 ± 11.10 mg/dL; CG = 79.86 ± 25.68 mg/dL, p = 0.03). Also, no alterations were observed in prostatic morphology, testosterone or estradiol levelsin the two groups analyzed. Conclusion: Flaxseed intake during lactation did not produce histological alterations in prostatic alveolus or in sexual hormones, but programmed to a reduction in lipid profile in adult life with decreased cardiovascular risk.
