    High Dimensional Classification with combined Adaptive Sparse PLS and Logistic Regression

    Motivation: The high dimensionality of genomic data calls for the development of specific classification methodologies, especially to prevent over-optimistic predictions. This challenge can be tackled by compression and variable selection, which combined constitute a powerful framework for classification, as well as data visualization and interpretation. However, current proposed combinations lead to instable and non convergent methods due to inappropriate computational frameworks. We hereby propose a stable and convergent approach for classification in high dimensional based on sparse Partial Least Squares (sparse PLS). Results: We start by proposing a new solution for the sparse PLS problem that is based on proximal operators for the case of univariate responses. Then we develop an adaptive version of the sparse PLS for classification, which combines iterative optimization of logistic regression and sparse PLS to ensure convergence and stability. Our results are confirmed on synthetic and experimental data. In particular we show how crucial convergence and stability can be when cross-validation is involved for calibration purposes. Using gene expression data we explore the prediction of breast cancer relapse. We also propose a multicategorial version of our method on the prediction of cell-types based on single-cell expression data.     Snow spectral albedo at Summit, Greenland: measurements and numerical simulations based on physical and chemical properties of the snowpack

    The broadband albedo of surface snow is determined both by the near-surface profile of the physical and chemical properties of the snowpack and by the spectral and angular characteristics of the incident solar radiation. Simultaneous measurements of the physical and chemical properties of snow were carried out at Summit Camp, Greenland (72°36® N, 38°25® W, 3210 m a.s.l.) in May and June 2011, along with spectral albedo measurements. One of the main objectives of the field campaign was to test our ability to predict snow spectral albedo by comparing the measured albedo to the albedo calculated with a radiative transfer model, using measured snow physical and chemical properties. To achieve this goal, we made daily measurements of the snow spectral albedo in the range 350–2200 nm and recorded snow stratigraphic information down to roughly 80 cm. The snow specific surface area (SSA) was measured using the DUFISSS instrument (DUal Frequency Integrating Sphere for Snow SSA measurement, Gallet et al., 2009). Samples were also collected for chemical analyses including black carbon (BC) and dust, to evaluate the impact of light absorbing particulate matter in snow. This is one of the most comprehensive albedo-related data sets combining chemical analysis, snow physical properties and spectral albedo measurements obtained in a polar environment. The surface albedo was calculated from density, SSA, BC and dust profiles using the DISORT model (DIScrete Ordinate Radiative Transfer, Stamnes et al., 1988) and compared to the measured values. Results indicate that the energy absorbed by the snowpack through the whole spectrum considered can be inferred within 1.10%. This accuracy is only slightly better than that which can be obtained considering pure snow, meaning that the impact of impurities on the snow albedo is small at Summit. In the near infrared, minor deviations in albedo up to 0.014 can be due to the accuracy of radiation and SSA measurements and to the surface roughness, whereas deviations up to 0.05 can be explained by the spatial heterogeneity of the snowpack at small scales, the assumption of spherical snow grains made for DISORT simulations and the vertical resolution of measurements of surface layer physical properties. At 1430 and around 1800 nm the discrepancies are larger and independent of the snow properties; we propose that they are due to errors in the ice refractive index at these wavelengths. This work contributes to the development of physically based albedo schemes in detailed snowpack models, and to the improvement of retrieval algorithms for estimating snow properties from remote sensing data

    Fgf15 Neurons of the Dorsomedial Hypothalamus Control Glucagon Secretion and Hepatic Gluconeogenesis.

    The counterregulatory response to hypoglycemia is an essential survival function. It is controlled by an integrated network of glucose-responsive neurons, which trigger endogenous glucose production to restore normoglycemia. The complexity of this glucoregulatory network is, however, only partly characterized. In a genetic screen of a panel of recombinant inbred mice we previously identified Fgf15, expressed in neurons of the dorsomedial hypothalamus (DMH), as a negative regulator of glucagon secretion. Here, we report on the generation of Fgf15 <sup>CretdTomato</sup> mice and their use to further characterize these neurons. We show that they were glutamatergic and comprised glucose-inhibited and glucose-excited neurons. When activated by chemogenetics, Fgf15 neurons prevented the increase in vagal nerve firing and the secretion of glucagon normally triggered by insulin-induced hypoglycemia. On the other hand, they increased the activity of the sympathetic nerve in the basal state and prevented its silencing by glucose overload. Higher sympathetic tone increased hepatic Creb1 phosphorylation, Pck1 mRNA expression, and hepatic glucose production leading to glucose intolerance. Thus, Fgf15 neurons of the DMH participate in the counterregulatory response to hypoglycemia by a direct adrenergic stimulation of hepatic glucose production while suppressing vagally induced glucagon secretion. This study provides new insights into the complex neuronal network that prevents the development of hypoglycemia

    Deep Radio Imaging of Globular Clusters and the Cluster Pulsar Population

    We have obtained deep multifrequency radio observations of seven globular clusters using the Very Large Array and the Australia Telescope Compact Array. Five of these, NGC 6440, NGC 6539, NGC 6544, NGC 6624 and Terzan 5 had previously been detected in a shallower survey for steep spectrum radio sources in globular clusters (Fruchter and Goss 1990). The sixth, the rich globular cluster, Liller 1, had heretofore been undetected in the radio, and the seventh, 47 Tucanae, was not included in our original survey. High resolution 6 and 20 cm images of three of the clusters, NGC 6440, NGC 6539, NGC 6624 reveal only point sources coincident with pulsars which have been discovered subsequent to our first imaging survey. 21 and 18 cm images reveal several point sources within a few core-radii of the center of 47 Tuc. Two of these are identified pulsars, and a third, which is both variable and has a steep spectrum, is also most likely a pulsar previously identified by a pulsed survey. However, the 6, 20 and 90 cm images of NGC 6544, Liller 1 and Terzan 5 display strong steep-spectrum emission which cannot be associated with known pulsars. The image of the rich cluster Terzan 5 displays numerous point sources within 30â€Čâ€Č30'', or 4 core radii of the cluster center. The density of these objects rises rapidly toward the core, where an elongated region of emission is found. The brightest individual sources, as well as the extended emission, possess the steep spectra expected of pulsars. Furthermore, the flux distribution of the sources agrees well with the standard pulsar luminosity function. The total luminosity and number of objects observed suggest that Terzan 5 contains more pulsars than any other Galactic globular cluster.Comment: 33 pages, 6 Postscript figures; Accepted for publication in the Astrophysical Journal; abstract abridged.     Functional approach to 2+1 dimensional gravity coupled to particles

    The quantum gravity problem of N point particles interacting with the gravitational field in 2+1 dimensions is approached working out the phase-space functional integral. The maximally slicing gauge is adopted for a non compact open universe with the topology of the plane. The conjugate momenta to the gravitational field are related to a class of meromorphic quadratic differentials. The boundary term for the non compact space is worked out in detail. In the extraction of the physical degrees of freedom functional determinants related to the puncture formulation of string theory occur and cancel out in the final reduction.     A Retinoic Acid Responsive Hoxa3 Transgene Expressed in Embryonic Pharyngeal Endoderm, Cardiac Neural Crest and a Subdomain of the Second Heart Field

    A transgenic mouse line harbouring a ÎČ-galacdosidase reporter gene controlled by the proximal 2 kb promoter of Hoxa3 was previously generated to investigate the regulatory cues governing Hoxa3 expression in the mouse. Examination of transgenic embryos from embryonic day (E) 8.0 to E15.5 revealed regionally restricted reporter activity in the developing heart. Indeed, transgene expression specifically delineated cells from three distinct lineages: a subpopulation of the second heart field contributing to outflow tract myocardium, the cardiac neural crest cells and the pharyngeal endoderm. Manipulation of the Retinoic Acid (RA) signaling pathway showed that RA is required for correct expression of the transgene. Therefore, this transgenic line may serve as a cardiosensor line of particular interest for further analysis of outflow tract development
