    Short communication: NKG2C+ NK cells contribute to increases in CD16+CD56- cells in HIV type 1+ individuals with high plasma viral load.

    Chronic HIV-1 infection results in the expansion of both NKG2C+ and CD16+CD56- human natural killer cells. NKG2C+ cells proliferate in response to human cytomegalovirus (HCMV) and expansion of the dysfunctional CD56-CD16+ natural killer (NK) cells is associated with HIV-1 viremia. Here we report an association between increased proportions of CD56-CD16+ NK cells in viremic HIV-1+ individuals and an increased contribution of NKG2C+ cells to this subset. These data, in addition to anti-HCMV IgG serology, indicate a potential contribution of both HCMV and HIV-1 to NK cell dysfunction in HIV-1-infected individuals

    Micro-focussed XAFS spectroscopy to study Ni-bearing precipitates in the metal of corroded Zircaloy-2

    The present work concerns an investigation of the local atomic environment of Ni-containing secondary phase precipitates (SPP) present in the metal-part of Zircaloy-2 cladding tubes. An unirradiated Zircaloy-2 and two specimens irradiated in a commercial nuclear power plant are characterized using μ-focussed synchrotron radiation, and by x-ray absorption fine structure (XAFS) spectroscopy. The patterns of Ni K-edge XANES and EXAFS of SPP in unirradiated and irradiated cladding are found different. Considering the fact that Ni-bearing SPP in the unirradiated samples are mainly Zintl phase Zr2(Fe, Ni) type, a detailed EXAFS analysis of near-neighbor Ni atoms has been made. The result of a curve fit for the first two shells shows that about 2 Ni(Fe) and 8Zr atoms are coordinated at2.68 and 2.77Å, respectively, around a central Ni atom in the SPP. XANES data analysis provides total electronic density of states at the Fermi level of unirradiated Zr2(Fe, Ni). At the Ni K-edge EXAFS spectra of irradiated SPP, however, only a single scattering peak is observed demonstrating the structural disorder introduced by the neutron irradiation. The coordination number of the Ni neighboring shells is reduced markedly due to the formation of point and extended defects in the damaged SPP lattice. Dissolution of Ni from the SPP is also evident from the data. The results of this study provide a further basis for the description of both crystallographic and electronic structures of intermetallic second-phase precipitates found in Zr-based alloy

    Self-assembled ge nanostructures on polymer-coated silicon: growth and characterization

    Self-assembled Ge nanoparticles have been grown on polymer-coated Si substrates by thermal evaporation under high vacuum utilizing the nonwetting condition given by the surface free-energy relation σ Ge»σpolymer. The nanostructures have been characterized by Raman spectroscopy, atomic-force microscopy (AFM), and optical microscopy. Raman spectrum shows a prominent Ge-Ge vibration peak at 302cm-1. AFM and optical microscopy show the formation of isolated Ge islands (>~100nm base, <~25nm height), nanowires (160 nm base, 25 nm height), and islands in linear chains. The possibility of embedding such nanostructures in waveguide structures are discussed

    The Impact of Waxy Wheat Flour, Inulin and Guar Gum on Post‐Prandial Glycaemic and Satiety Indices, Sensory Attributes and Shelf Life of Tandoori and Pita Breads

    The purpose of this study was to develop two different functional breads (Pita and Tan-doori) supplemented with novel functional ingredients: waxy wheat flour (15%), inulin (8%) and guar gum (2%) independently and in combination. The breads were submitted to sensory characteristics, shelf life, glycaemic and satiety indices assessments. Both guar gum and inulin independently and in combination significantly (p < 0.05) decreased post‐prandial blood glucose and glycemic response of Pita (GI of guar gum bread was 55%, inulin 57%) and Tandoori (GI of guar gum bread was 57% and inulin bread was 60%) compared to the control breads (GI 100%). Moreover, the results of the area under the curve of satiety showed that the addition of functional ingredients increased satiety levels as follows: for Pita, control was 355, a combination of all ingredients was 418, inulin was 451 and guar gum was 452; for Tandoori, control was 329, a combination of all ingredients was 420, inulin was 381 and guar gum was 390. The results showed that all sensory characteristics were improved, and breads were acceptable (all obtained more than five points) when the highest proportions of ingredients were added. Similarly, the shelf life of supplemented Pita and Tandoori breads was improved with the addition of ingredients. Therefore, the functional ingredients such as inulin and guar gum can be used independently and in combination to reduce GI and increase satiety of Pita and Tandoori bread with acceptable quality and shelf life

    Coronaviruses (CoVs) are important human and animal pathogens that induce fatal respiratory, gastrointestinal and neurological disease. The outbreak of the severe acute respiratory syndrome (SARS) in 2002/2003 has demonstrated human vulnerability to (Coronavirus) CoV epidemics. Neither vaccines nor therapeutics are available against human and animal CoVs. Knowledge of host cell proteins that take part in pivotal virus-host interactions could define broad-spectrum antiviral targets. In this study, we used a systems biology approach employing a genome-wide yeast-two hybrid interaction screen to identify immunopilins (PPIA, PPIB, PPIH, PPIG, FKBP1A, FKBP1B) as interaction partners of the CoV non-structural protein 1 (Nsp1). These molecules modulate the Calcineurin/NFAT pathway that plays an important role in immune cell activation. Overexpression of NSP1 and infection with live SARS-CoV strongly increased signalling through the Calcineurin/NFAT pathway and enhanced the induction of interleukin 2, compatible with late-stage immunopathogenicity and long-term cytokine dysregulation as observed in severe SARS cases. Conversely, inhibition of cyclophilins by cyclosporine A (CspA) blocked the replication of CoVs of all genera, including SARS-CoV, human CoV-229E and -NL-63, feline CoV, as well as avian infectious bronchitis virus. Non-immunosuppressive derivatives of CspA might serve as broad-range CoV inhibitors applicable against emerging CoVs as well as ubiquitous pathogens of humans and livestock

    Shape Transition in the Epitaxial Growth of Gold Silicide in Au Thin Films on Si(111)

    Growth of epitaxial gold silicide islands on bromine-passivated Si(111) substrates has been studied by optical and electron microscopy, electron probe micro analysis and helium ion backscattering. The islands grow in the shape of equilateral triangles up to a critical size beyond which the symmetry of the structure is broken, resulting in a shape transition from triangle to trapezoid. The island edges are aligned along Si[110]Si[110] directions. We have observed elongated islands with aspect ratios as large as 8:1. These islands, instead of growing along three equivalent [110] directions on the Si(111) substrate, grow only along one preferential direction.
