Severe Dengue Is Associated with Consumption of von Willebrand Factor and Its Cleaving Enzyme ADAMTS-13
Severe dengue infections are characterized by thrombocytopenia, clinical bleeding and plasma leakage. Activation of the endothelium, the inner lining of blood vessels, leads to the secretion of storage granules called Weibel Palade bodies (WPBs). We demonstrated that severe dengue in Indonesian children is associated with a strong increase in plasma levels of the WPB constituents von Willebrand factor (VWF), VWF propeptide and osteoprotegerin (OPG). An increased amount of the hemostatic protein VWF was in a hyperreactive, platelet binding conformation, and this was most pronounced in the children who died. VWF levels at enrollment were lower than expected from concurrent VWF propeptide and OPG levels and VWF levels did not correlate well with markers of disease severity. Together, this suggests that VWF is being consumed during severe dengue. Circulating levels of the VWF-cleaving enzyme ADAMTS-13 were reduced. VWF is a multimeric protein and a subset of children had a decrease in large and intermediate VWF multimers at discharge. In conclusion, severe dengue is associated with exocytosis of WPBs with consumption of VWF and low ADAMTS-13 activity levels. This may contribute to the thrombocytopenia and complications of dengue
Matrix Metalloproteinase 1: Role in Sarcoma Biology
In carcinomas stromal cells participate in cancer progression by producing proteases such as MMPs. The expression MMP1 is a prognostic factor in human chondrosarcoma, however the role in tumor progression is unknown. Laser capture microdissection and In Situ hybridization were used to determine cellular origin of MMP1 in human sarcomas. A xenogenic model of tumor progression was then used and mice were divided in two groups: each harboring either the control or a stably MMP1 silenced cell line. Animals were sacrificed; the neovascularization, primary tumor volumes, and metastatic burden were assessed. LCM and RNA-ISH analysis revealed MMP1 expression was predominantly localized to the tumor cells in all samples of sarcoma (p = 0.05). The percentage lung metastatic volume at 5 weeks (p = 0.08) and number of spontaneous deaths secondary to systemic tumor burden were lower in MMP1 silenced cell bearing mice. Interestingly, this group also demonstrated a larger primary tumor size (p<0.04) and increased angiogenesis (p<0.01). These findings were found to be consistent when experiment was repeated using a second independent MMP1 silencing sequence. Prior clinical trials employing MMP1 inhibitors failed because of a poor understanding of the role of MMPs in tumor progression. The current findings indicating tumor cell production of MMP1 by sarcoma cells is novel and highlights the fundamental differences in MMP biology between carcinomas and sarcomas. The results also emphasize the complex roles of MMP in tumor progression of sarcomas. Entwicklung eines Wurzelsymbiosesystems zum Testen von Umweltchemikalien auf ihre Wirkung in Boeden
Root symbiosis of legumes with symbiotic bacteria of the genus Rhizobium and with symbiotic fungi (vesicular-arbuscular mycorrhiza) have been established as sensitive bioindicators for soil environmental chemicals. For different plant species also different transfer rates for those substances have been found. Towards heavy metals (cadmium, lead, arsenite) the symbioses react very differentiated. As environmental chemicals chinoline, fluoranthene, phenanthrene und anthracene have been studied. Fuer verschiedene Kulturpflanzen wurden unterschiedliche Transferraten aus dem Boden in die Pflanzenmatrix nachgewiesen. Gegenueber Schwermetallen (Cadmium, Blei, Arsenit) reagieren die Symbiosen sehr unterschiedlich. Als Altstoffe wurden modellhaft Chinolin, Fluoranthen, Phenanthren und Anthrazen verwendet. Not AvailableThe current study focused on the impact of climate change on tropical root and tuber crops of India over some of the major
growing areas using the crop model, WOFOST. As part of this study, the impact of climate change on the yield of rice and
potato is also carried out using the same crop model to understand the advantages of root and tuber crops over the yield variations
of rice and potato. The LARS weather generator is used to derive future climate for 2030, 2050, and 2070 for the two
representative concentration pathways (RCPs)-4.5 and 8.5. The future climate projections in the study locations indicate an
increment in minimum and maximum temperatures up to a value of 3.4 and 3.8 ÂşC respectively. The rainfall also indicates a
drastic fluctuation from -721 to 448 mm in the future. The results reveal that the predicted crop yield varies from one location
to another depending on the future climate. Cassava (-13 to 12%, -17 to 8%), sweet potato (-32 to 14%, -38 to 13%),
greater yam (-11 to 8, -14 to 6), elephant foot yam (-10 to 6, -12 to 4), and taro (-16 to 19, -28 to 18) can be considered as
future crops based on their predicted yield variations and economics compared to that of rice (-26 to 15%, -50 to 18%) and
potato (-37 to 7%, -59 to 8.5%) for both the RCPs. Among other crops, sweet potato has the same crop duration as rice and
potato and can be recommended to increase food access. These crops can be recommended for enhancing the availability of
