    Evidence that high von Willebrand factor and low ADAMTS-13 levels independently increase the risk of a non-fatal heart attack

    Background: A disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13 (ADAMTS-13) may influence von Willebrand factor (VWF) levels and consequently the risk of myocardial infarction (MI). Moreover, ADAMTS-13 influences hemostatic plug formation in mouse models. We therefore studied their associations in the Glasgow MI Study (GLAMIS). Methods and results: We measured ADAMTS-13 and VWF antigen levels by ELISAs in stored plasma from a case–control study of 466 MI cases and 484 age- and sex-matched controls from the same north Glasgow population. There was no correlation between ADAMTS-13 and VWF levels in cases or controls. ADAMTS-13 levels correlated positively with serum cholesterol and triglycerides and body mass index, and negatively with high-density lipoprotein-cholesterol. VWF levels correlated with age, fibrinogen and C-reactive protein. In multivariable analyses including risk factors, VWF correlated positively with risk of MI, and ADAMTS-13 correlated negatively with risk of MI. These associations were independent of each other. The association of ADAMTS-13 with risk of MI was observed only in multivariable analysis. Conclusions: VWF and ADAMTS-13 levels were not associated in this study, and showed associations with MI risk in opposite directions but of similar strength. The association of ADAMTS-13 with MI is influenced by lipid levels, and consequently requires further investigation

    Hormonal replacement therapy, prothrombotic mutations and the risk of venous thrombosis

    Hormone replacement therapy (HRT) increases the risk of venous thrombosis. We investigated whether this risk is affected by carriership of hereditary prothrombotic abnormalities. Therefore, we determined the two most common prothrombotic mutations, factor V Leiden and prothrombin 20210A in women who participated in a case-control study on venous thrombosis. Relative risks were expressed as odds ratios (OR) with 95% confidence intervals (CI95). Among 7 7 women aged 45-64 years with a first venous thrombosis, 51% were receiving HRT at the time of thrombosis, compared with 24% of control women (OR = 3.3, CI95 1.8-5.8). Among the patients, 23% had a prothrombotic defect, versus 7% among the control women (OR = 3.8, CI95 1.7- 8.5). Women who had factor V Leiden and used HRT had a 15-fold increased risk (OR = 15.5, CI95 3.1-77), which exceeded the expected joint odds ratio of 6.1 (under an additive model). We conclude that the thrombotic risk of HRT may particularly affect women with prothrombotic mutations. Efforts to avoid HRT in women with increased risk of thrombosis are advisable

    Reproductive Biology and Phylogeny of Lizards and Tuatara. J. L. Rheubert, D. S. Siegel, and S. E. Trauth (Eds.). 2014. CRC Press. ISBN 9781466579866. 760 p. $143.96 (hardcover).— According to the authors, this book was intended to summarize the current knowledge of phylo- geny and reproduction of the Lepidosauria. I believe the authors have achieved their goal. Many of the chapters in this book are derived from presentations that took place at the Symposium on Reproductive Biology of Lizards at the Joint Meeting of Ichthyologists and Herpetologists held in Chattanooga, Tennessee in 2014. Although the majority of the authors are from the United States, the book includes authors from nine countries representing four continents and New Zealand. In the first chapter, Laurie Vitt states (p. 1), “It is difficult to imagine anything more interesting to biologists... than understanding the origins of reproductive patterns among lizards.” I agree. Vitt continues with the early history of topics such as seasonality of reproduction and fat storage, parthenogenesis, evolution of viviparity, placentation, and several other topics. For each of these topics, Vitt explains the origin of the field and the major researchers responsible for the insights and hypotheses in each field. His chapter ends with a call for natural history studies. In chapter two, John Wiens and Shea Lambert discuss the phylogeny of lizard families. They present a compelling argument for combining molecular and morphological data sets in phylogenetic studies. The authors admit that there are still many unresolved issues and more data are needed to fully understand the phylogeny of the lizards. In chapter three, Jose ́ Mart ́ın and Pilar Lo ́ pez define pheromone, discuss chemosensory abilities, and describe the role of pheromones in lizard reproduction. They present a list of studies on lizards and Tuatara, describe the source of chemicals, and the chemical nature of putative phero- mones. Mart ́ın and Lo ́pez conclude with the evolutionary origin of chemical signaling in lizards and a call for additional work on pheromone communication in lizards. Robert Cox and Atiel Kahrl discuss sexual selection and sexual dimorphism in lizards in chapter four. They include a list of studies and evaluate the data to determine what factors led to sexual dimorphism. They discuss intra- and intersexual selection. Cox and Kahrl discuss the ultimate cause and proximate mechanisms for sexual dimorphism and conclude with a discussion of the consequence of sexual selection on speciation and species recognition

    Managing Dynamic User Communities in a Grid of Autonomous Resources

    One of the fundamental concepts in Grid computing is the creation of Virtual Organizations (VO's): a set of resource consumers and providers that join forces to solve a common problem. Typical examples of Virtual Organizations include collaborations formed around the Large Hadron Collider (LHC) experiments. To date, Grid computing has been applied on a relatively small scale, linking dozens of users to a dozen resources, and management of these VO's was a largely manual operation. With the advance of large collaboration, linking more than 10000 users with a 1000 sites in 150 counties, a comprehensive, automated management system is required. It should be simple enough not to deter users, while at the same time ensuring local site autonomy. The VO Management Service (VOMS), developed by the EU DataGrid and DataTAG projects[1, 2], is a secured system for managing authorization for users and resources in virtual organizations. It extends the existing Grid Security Infrastructure[3] architecture with embedded VO affiliation assertions that can be independently verified by all VO members and resource providers. Within the EU DataGrid project, Grid services for job submission, file- and database access are being equipped with fine- grained authorization systems that take VO membership into account. These also give resource owners the ability to ensure site security and enforce local access policies.     Late Holocene palynology and palaeovegetation of tephra-bearing mires at Papamoa and Waihi Beach, western Bay of Plenty, North Island, New Zealand.

    The vegetation history of two mires associated with Holocene dunes near the western Bay of Plenty coast, North Island, New Zealand, is deduced from pollen analysis of two cores. Correlation of airfall tephra layers in the peats, and radiocarbon dates, indicate that the mires at Papamoa and Waihi Beach are c. 4600 and c. 2900 conventional radiocarbon years old, respectively. Tephras used to constrain the chronology of the pollen record include Rotomahana (1886 AD), Kaharoa (700 yr B.P.), Taupo (Unit Y; 1850 yr B.P.), Whakaipo (Unit V; 2700 yr B.P.), Stent (Unit Q; 4000 yr B.P.), Hinemaiaia (Unit K; 4600 yr B.P.), and reworked Whakatane (c. 4800 yr B.P.) at Papamoa, and Kaharoa and Taupo at Waihi Beach. Peat accumulation rates at Papamoa from 4600 - 1850 yr B.P. range from 0.94 to 2.64 mm/yr (mean 1.37 mm/yr). At Waihi Beach, from 2900 yr B.P. - present day, they range from 0.11 to 0.21 mm/yr (mean 0.20 mm/yr). Peat accumulation at both sites was slowest from 1850 to 700 yr B.P., suggesting a drier overall climate during this interval. At both sites, the earliest organic sediments, which are underlain by marine or estuarine sands, yield pollen spectra indicating salt marsh or estuarine environments. Coastal vegetation communities declined at both sites, as sea level gradually fell or the coast prograded, and were eventually superseded by a low moor bog at Papamoa, and a mesotrophic swamp forest at Waihi Beach. These differences, and the marked variation in peat accumulation rates, probably reflect local hydrology and are unlikely to have been climatically controlled. The main regional vegetation during this period was mixed northern conifer-angiosperm forest. Kauri (Agathis australis) formed a minor component of these forests, but populations of this tree have apparently not expanded during the late Holocene at these sites, which are near its present southern limit. Occasional shortlived forest disturbances are detectable in these records, in particular immediately following the deposition of Taupo Tephra. However, evidence for forest clearance during the human era is blurred by the downward dislocation of modern adventi ve pollen at these sites, preventing the clear differentiation of the Polynesian and European eras

    Learning to represent visual input

    One of the central problems in computational neuroscience is to understand how the object-recognition pathway of the cortex learns a deep hierarchy of nonlinear feature detectors. Recent progress in machine learning shows that it is possible to learn deep hierarchies without requiring any labelled data. The feature detectors are learned one layer at a time and the goal of the learning procedure is to form a good generative model of images, not to predict the class of each image. The learning procedure only requires the pairwise correlations between the activations of neuron-like processing units in adjacent layers. The original version of the learning procedure is derived from a quadratic ‘energy’ function but it can be extended to allow third-order, multiplicative interactions in which neurons gate the pairwise interactions between other neurons. A technique for factoring the third-order interactions leads to a learning module that again has a simple learning rule based on pairwise correlations. This module looks remarkably like modules that have been proposed by both biologists trying to explain the responses of neurons and engineers trying to create systems that can recognize objects

    Probing black holes in non-perturbative gauge theory

    We use a 0-brane to probe a ten-dimensional near-extremal black hole with N units of 0-brane charge. We work directly in the dual strongly-coupled quantum mechanics, using mean-field methods to describe the black hole background non-perturbatively. We obtain the distribution of W boson masses, and find a clear separation between light and heavy degrees of freedom. To localize the probe we introduce a resolving time and integrate out the heavy modes. After a non-trivial change of coordinates, the effective potential for the probe agrees with supergravity expectations. We compute the entropy of the probe, and find that the stretched horizon of the black hole arises dynamically in the quantum mechanics, as thermal restoration of unbroken U(N+1) gauge symmetry. Our analysis of the quantum mechanics predicts a correct relation between the horizon radius and entropy of a black hole.Comment: 30 pages, LaTeX, 8 eps figures. v2: references added. v3: more reference

    Particle dynamics in a class of 2-dimensional gravity theories

    We provide a method to determine the motion of a classical massive particle in a background geometry of 2-dimensional gravity theories, for which the Birkhoff theorem holds. In particular, we get the particle trajectory in a continuous class of 2-dimensional dilaton gravity theories that includes the Callan-Giddings-Harvey-Strominger (CGHS) model, the Jackiw-Teitelboim (JT) model, and the dd-dimensional ss-wave Einstein gravity.
