    Measurement of metabolic processes in lymphocytes in Varicella zoster infection showed highly increased intercellular glycolisys activity with functional cellular overload. Same time, we discovered decreased level of intensivity of first stages of TCC, that rules to lower cycle energetic efficiency and intense metabolitsaminoacides intake for TCC, guiding to high aminoacides transport to lymphocytes. Usage of succinic acid and its salts gives more substrates for TCC, increasing its energetic efficiency and lymphocytes functional activity.Оценка характера и интенсивности метаболических процессов в лимфоцитах больных ветряной оспой по изменениям активности внутриклеточных ферментов показала, что в разгар заболевания отмечена интенсификация реакций гликолиза при значительном повышении функциональной нагрузки на клетки, происходит значительное снижение интенсивности реакций начального этапа ЦТК, что должно уменьшать энергетическую эффективность цикла, а также интенсивное поступление метаболитов на снабжение ЦТК субстратами с аминокислотного обмена, обеспечивая повышенный транспорт аминокислот в лимфоциты. Применение препаратов янтарной кислоты или ее солей дает дополнительное субстратное насыщение ЦТК, повышая энергоэффективность цикла и функциональные возможности лимфоцитов

    Statistical Analysis of Readthrough Levels for Nonsense Mutations in Mammalian Cells Reveals a Major Determinant of Response to Gentamicin

    The efficiency of translation termination depends on the nature of the stop codon and the surrounding nucleotides. Some molecules, such as aminoglycoside antibiotics (gentamicin), decrease termination efficiency and are currently being evaluated for diseases caused by premature termination codons. However, the readthrough response to treatment is highly variable and little is known about the rules governing readthrough level and response to aminoglycosides. In this study, we carried out in-depth statistical analysis on a very large set of nonsense mutations to decipher the elements of nucleotide context responsible for modulating readthrough levels and gentamicin response. We quantified readthrough for 66 sequences containing a stop codon, in the presence and absence of gentamicin, in cultured mammalian cells. We demonstrated that the efficiency of readthrough after treatment is determined by the complex interplay between the stop codon and a larger sequence context. There was a strong positive correlation between basal and induced readthrough levels, and a weak negative correlation between basal readthrough level and gentamicin response (i.e. the factor of increase from basal to induced readthrough levels). The identity of the stop codon did not affect the response to gentamicin treatment. In agreement with a previous report, we confirm that the presence of a cytosine in +4 position promotes higher basal and gentamicin-induced readthrough than other nucleotides. We highlight for the first time that the presence of a uracil residue immediately upstream from the stop codon is a major determinant of the response to gentamicin. Moreover, this effect was mediated by the nucleotide itself, rather than by the amino-acid or tRNA corresponding to the −1 codon. Finally, we point out that a uracil at this position associated with a cytosine at +4 results in an optimal gentamicin-induced readthrough, which is the therapeutically relevant variable

    Aim. In this study we evaluated clinical effectiveness and safety of nebulized prostacyclin in patients with Novel Coronavirus Disease (SARS-CoV-2). Materials and methods: We have included 44 male patients with moderate PCR confirmed SARS-CoV-2 infection in this study. Control group consisted of 23 patients treated with nebulized prostacyclin (PGI2). besides standard therapy. We compared intensiveness and duration of infectious intoxication syndrome, duration of fever, cough as well as SpO2 level, complete blood count and chemokine status values. Results: Statistically significant difference in duration of fever, cough, intensiveness and duration of infectious intoxication syndrome were observed. Lymphocyte and platelet counts were significantly higher in control group We have also noticed significantly lower level of proinflammatory mediators and C4-complement component in control group. Only 1 adverse effect associated with inhaled prostacyclin was reported. Conclusion. Nebulized prostacyclin showed therapeutic efficacy and good safety profile in adults with moderate COVID-19.Цель: оценка клинической эффективности и безопасность ингаляционного простациклина у пациентов с новой коронавирусной инфекцией (SARS-CoV-2). Материалы и методы: в исследование были включены 44 пациента мужского пола с подтвержденной новой коронавирусной инфекцией среднетяжелого течения. Опытную группу составили 23 пациента, которым, помимо стандартной терапии, был назначен ингаляционный простациклин (PGI2). Клиническая эффективность илопроста была оценена по длительности и выраженности общеинфекционных синдромов (интоксикации, лихорадки), длительности кашля, уровню насыщения крови кислородом, значениям параметров общеклинического анализа крови, показателю иммунологического статуса пациентов. Результаты: получено статистически значимое снижение длительности лихорадки, продолжительности кашля, выраженности и длительности синдрома общей инфекционной интоксикации в опытной группе. Также отмечено, что у этих пациентов средние значения количества лимфоцитов, тромбоцитов достоверно увеличивалось, а значение СОЭ снижалось. Средние значения провоспалительных цитокинов, хемокинов, а также С4-компонента комплемента были статистически значимо ниже, чем у больных COVID-19 в группе сравнения. Нежелательные реакции, связанные с инга ляционной терапией простациклином, были отмечены в 1 наблюдении. Заключение: показана терапевтическая эффективность и хороший профиль безопасности ингаляционного простациклина у пациентов с COVID-19 средней степени тяжести

    Supersymmetric QCD corrections to e+etbˉHe^+e^-\to t\bar{b}H^- and the Bernstein-Tkachov method of loop integration

    The discovery of charged Higgs bosons is of particular importance, since their existence is predicted by supersymmetry and they are absent in the Standard Model (SM). If the charged Higgs bosons are too heavy to be produced in pairs at future linear colliders, single production associated with a top and a bottom quark is enhanced in parts of the parameter space. We present the next-to-leading-order calculation in supersymmetric QCD within the minimal supersymmetric SM (MSSM), completing a previous calculation of the SM-QCD corrections. In addition to the usual approach to perform the loop integration analytically, we apply a numerical approach based on the Bernstein-Tkachov theorem. In this framework, we avoid some of the generic problems connected with the analytical method.Comment: 14 pages, 6 figures, accepted for publication in Phys. Rev.

    Abstract. A complex immunologic examination of fifty patients with clinically significant postoperative fibrosis has been performed. Immunogram indexes were determined, and activities of basic oxidation-reduction enzymes were investigated in blood lymphocytes. What concerned immune status of the patients under study, some peculiar features have been revealed, with respect to cellular and humoral compartments, i.e., relative increase in CD3+ population, a shift to CD4+ cells in the ratio of lymphocyte subpopulations, higher indexes of immune regulation and phagocytosis, increased IgG level, and lower content of IgA, as compared to appropriate controls. Metabolic parameters of lymphocytes reflect their functional activation, being expressed as an enhanced ability for intracellular synthetic processes, elevated functional activity of lymphocytes, and increased immune response. The general pattern of changes in immune status and metabolic indexes of lymphocytes allow us to assume participation of autoimmune component in progression of epidural fibrosis. The data obtained may be used in combined diagnostics and correction of this disorder. (Med. Immunol., vol. 10, N 6, pp 589-592)

    The paper describes a clinical case of the rare status epilepticus syndrome FIRES (febrile infection responsive encephalopathy) + NORSE (new-onset refractory status epilepticus). The difficulty of diagnosing this condition is due to the fact that it occurs extremely rarely and the laboratory and instrumental findings are nonspecific. The disease is characterized by acute onset, fever, and subsequent development of status epilepticus, cognitive impairment, and pharmacotherapy-resistant epilepsy. The paper describes a 19-year-old male patient who was observed to have a favorable course of FIRES + NORSE syndrome without pharmacotherapy-resistant epilepsy, motor deficiency, and with the preservation of self-care skills
