    meat quality traits of marchigiana beef cattle

    Variation in quality of meat within the same breed and among different ones is due both to different biological factor (genotype, productivity, sex and age) and environmental and technological factors (composition and nutritional level of diet, rearing, transport and slaughter) acting on animals reared . All these factors interact in a complex way and lead to a high variability of muscular structure that characterizes the qualitative characteristic of meat. For this reason, in a study that expected the control at slaughter of 1000 Marchigiana cattle, on a random, 114 sample's steaks were taken from LD at 5th rib, right side of carcasses grading U or more in SEUROP grid. On meat samples, we tested colour parameters (L*a*b*) using Minolta Chromameter CR 200 and light D65 and Drip Loss as reported by ASPA Commission (1996). Then meat samples were weighted, frozen, lyophilized; their chemical composition (moisture, protein and fat) was determined. Variability of analysis' results was studied according to JMP package (SAS).The colour means (L*, a*, b*) were obtained considering at least 3 readings on every sample. Luminosity value (L* = 40.1 ± 3.6) is similar to the one reported on subjects transported for 1-4 hours, similarly to the Marchigiana cattle object of this study, while is higher than the mean reported by other researchers on Valdostana cattle, English crossbreds and on Charolaise x Maremmana crossbreds. The red parameter (a* = 25.0 ± 3.2) is better than means reported by the same researchers. The yellow parameter (b* = 6.9 ± 2.5) is similar to results obtained on Valdostana cattle, but lower that means obtained by other researchers. The Hue values are lower than those reported for Charolaise x Maremmana (15.1 vs 26.1), while Chroma (26.1 ± 3.7) is similar to the values obtained by the same researchers. The moisture (72.8 ± 1.2) is similar to the mean obtained on Chianina calves, but lower than the one indicated on Marchigiana bulls. The protein (19.8 ± 1.6) is lower than that obtained on Marchigiana cattle. The intramuscular fat (3.9 ± 1.5) is higher than results obtained on Marchigiana calves. The Drip Loss (1.4 ± 0.5) is 1 point % lower than those reported by other researchers. This low Drip Loss can be considered a parameter that indicates a good muscular maturity of slaughtered cattle. All parameters have a low variability except for fat (33.1%) and Drip Loss (35.7%). The Marchigiana cattle confirms the good qualitative characteristics of its meat, and strengthen the opportunity to increase the number of Marchigiana heads to be bred and raisen

    Multislice computed tomography in an asymptomatic high-risk population

    Approximately 50% of all acute coronary syndromes occur in previously asymptomatic patients. This study evaluated the value of multislice computed tomography for early detection of significant coronary artery disease (CAD) in high-risk asymptomatic subjects. One hundred sixty-eight asymptomatic subjects with >or=1 major risk factor (hypertension, diabetes, hypercholesterolemia, family history, or smoking) and an inconclusive or unfeasible noninvasive stress test result (stress electrocardiography, echocardiography, or nuclear scintigraphy) were evaluated in an outpatient setting. After clinical examination and laboratory risk analysis, all patients underwent multislice computed tomographic (MSCT) coronary angiography within 1 week. In all subjects, conventional coronary angiography was also carried out. Multislice computed tomography displayed single-vessel CAD in 16% of patients, 2-vessel CAD in 7%, and 3-vessel CAD in 4%. Selective coronary angiography confirmed the results of multislice computed tomography in 99% of all patients. Sensitivity and specificity of MSCT coronary angiography were 100% and 98%, respectively, with a positive predictive value of 95% and a negative predictive value of 100%. In conclusion, MSCT coronary angiography is an excellent noninvasive technique for early identification of significant CAD in high-risk asymptomatic patients with inconclusive or unfeasible noninvasive stress test results

    Amaretti senza amido e farina come ingredienti in sistemi alimentari ad alta umiditĂ

    Riassunto Due tipologie di amaretti sono state ottenute utilizzando diverse formulazioni, una contenente amido come strutturante (VR) ed una preparata in accordo all’attuale normativa (NR) che non ammette la presenza di amido nella formulazione. Gli amaretti formulati con la nuova ricetta (NR) hanno mostrato valori di densitĂ  e durezza minori di quelli ottenuti con la vecchia ricetta ma una friabilitĂ  non significativamente differente. Le cinetiche di reidratazione e le caratteristiche meccaniche dei prodotti a diversi livelli di idratazione sono state studiate a seguito di immersione degli amaretti in soluzione di saccarosio a 25° C e di contatto su gelato a -15° C. In soluzione di saccarosio il prodotto NR ha mostrato una maggiore capacitĂ  di reidratazione ed una minore durezza e friabilitĂ  a tutti i livelli di idratazione. Nella reidratazione a -15° C su gelato il prodotto NR ha mostrato un acquisto di umiditĂ  non differente rispetto a VR ed una minore riduzione della friabilitĂ , pur presentando una consistenza minore di VR a tutti i livelli di idratazione. La sostituzione dell’amido nella formulazione di biscotti modifica le proprietĂ  fisiche e meccaniche del prodotto ma puĂČ permette migliori performance in termini di friabilitĂ  a bassi livelli di umiditĂ .Two types of ‘amaretti’ biscuits were produced with different formulations: one containing starch as a structuring agent (VR) and the other modified according to the current Italian legislation (NR) which does not permit the use of starch in the ‘amaretti’formulation. Amaretti formulated with the new recipe (NR) showed lower density and hardness than VR amaretti but not a significantly different friability value. Rehydration experiments in sucrose solution (25° C) and on ice-cream (-15° C) were carried out and rehydration kinetics and texture changes upon rehydration were evaluated. During rehydration in sucrose solution, the NR product showed a higher moisture uptake than VR and lower hardness and friability values at different hydration levels. When rehydration was carried out on ice-cream (at -15° C), NR showed a lower friability reduction than VR and a not different moisture uptake but it still showed lower hardness values than the latter at each hydration level. The replacement of starch in biscuits formulation affects the physical and mechanical properties of the products but could permit better performance in terms of friability retention at low moisture content

    Casing contribution to proteolytic changes and biogenic amines content in the production of an artisanal naturally fermented dry sausage

    The effect of two kinds of casings on the production and characteristics of a dry fermented sausage was investigated. In detail, an Italian product, naturally fermented at low temperatures and normally wrapped in beef casing instead of the most diffused hog one, was selected. Two different productions (one traditionally in beef casing (MCB) and another in hog casing (MCH)) were investigated over time to determine the differences particularly regarding proteolytic changes during fermentation and ripening. First of all, the product in hog casing required a longer ripening time, up to 120 days, instead of 45–50 days, because of the lower drying rate, while the microbial dynamics were not significantly modified. Conversely, the proteolysis showed a different evolution, being more pronounced, together with the biogenic amines content up to 341 mg/Kg instead of 265 mg/Kg for the traditional products. The latter products were instead characterized by higher quantities of total free amino acids, 3-methyl butanoic acid, 3-Methyl-1-butanal, and 2-Methylpropanal, enriching the final taste and aroma. The traditional product MCB also showed lower hardness and chewiness than MCH. The results highlight how the choice of casing has a relevant impact on the development of the final characteristics of fermented sausages

    Non-invasive assessment of coronary artery bypass graft patency using 16-slice computed tomography angiography

    <p>Abstract</p> <p>Background</p> <p>Invasive coronary angiography is the gold standard means of imaging bypass vessels and carries a small but potentially serious risk of local vascular complications, including myocardial infarction, stroke and death. We evaluated computed tomography as a non-invasive means of assessing graft patency.</p> <p>Methods</p> <p>Fifty patients with previous coronary artery bypass surgery who were listed for diagnostic coronary angiography underwent contrast enhanced computed tomography angiography using a 16-slice computed tomography scanner. Images were retrospectively gated to the electrocardiogram and two dimensional axial, multiplanar and three dimensional reconstructions acquired. Sensitivity, specificity, positive and negative predictive value, accuracy and level of agreement for detection of graft patency by multidetector computed tomography.</p> <p>Results</p> <p>A total of 116 grafts were suitable for analysis. The specificity of CT for the detection of graft patency was 100%, with a sensitivity of 92.8%, positive predictive value 100%, negative predictive value 85.8% and an accuracy of 94.8%. The kappa value of agreement between the two means of measuring graft patency was 0.9. Mean radiation dose was 9.0 ± 7.2 mSv for coronary angiography and 18.5 ± 4 mSv for computed tomography. Pooled analysis of eight studies, incorporating 932 grafts, confirmed a 97% accuracy for the detection of graft patency by multidetector computed tomography.</p> <p>Conclusion</p> <p>Computed tomography is an accurate, rapid and non-invasive method of assessing coronary artery bypass graft patency. However, this was achieved at the expense of an increase in radiation dose.</p

    HPV8-induced STAT3 activation led keratinocyte stem cell expansion in human actinic keratoses

    Despite epidermal turnover, the skin is host to a complex array of microbes, including viruses, such as HPV, which must infect and manipulate skin keratinocyte stem cells (KSCs) to survive. This crosstalk between the virome and KSC populations remains largely unknown. Here, we investigated the effect of HPV8 on KSCs using various mouse models. We observed that the HPV8 early region gene E6 specifically caused Lrig1+ hair follicle junctional zone KSC proliferation and expansion, which would facilitate viral transmission. Within Lrig1+ KSCs specifically, HPV8 E6 bound intracellular p300 to phosphorylate the STAT3 transcriptional regulatory node. This induced ΔNp63 expression, resulting in KSC expansion into the overlying epidermis. HPV8 was associated with 70% of human actinic keratoses. Together, these results define the “hit-and-run” mechanism for HPV8 in human actinic keratosis as an expansion of KSCs, which lack melanosome protection and are thus susceptible to sun light–induced malignant transformation

    Patient Acceptance of Noninvasive and Invasive Coronary Angiography

    BACKGROUND: Noninvasive angiography using multislice computed tomography (MSCT) is superior to magnetic resonance imaging (MRI) for detection of coronary stenoses. We compared patient acceptance of these two noninvasive diagnostic tests and invasive conventional coronary angiography (Angio). METHODS AND FINDINGS: A total of 111 consecutive patients with suspected coronary artery disease underwent MSCT, MRI, and Angio. Subsequently, patient acceptance of the three tests was evaluated with questionnaires in all patients. The main acceptance variables were preparation and information prior to the test, degree of concern, comfort, degree of helplessness, pain (on visual analog scales), willingness to undergo the test again, and overall satisfaction. Preparation for each test was not rated significantly differently, whereas patients were significantly more concerned about Angio than the two noninvasive tests (p<0.001). No pain during MSCT, MRI, and Angio as assessed on visual analog scales (0 to 100) was reported by 99, 93, and 31 patients, respectively. Among the 82 patients who felt pain during at least one procedure, both CT (0.9±4.5) and MRI (5.2±16.6) were significantly less painful than Angio (24.6±23.4, both p<0.001). MSCT was considered significantly more comfortable (1.49±0.64) than MRI (1.75±0.81, p<0.001). In both the no-revascularization (55 patients) and the revascularization group (56 patients), the majority of the patients (73 and 71%) would prefer MSCT to MRI and Angio for future imaging of the coronary arteries. None of the patients indicated to be unwilling to undergo MSCT again. The major advantages patients attributed to MSCT were its fast, uncomplicated, noninvasive, and painless nature. CONCLUSIONS: Noninvasive coronary angiography with MSCT is considered more comfortable than MRI and both MSCT and MRI are less painful than Angio. Patient preference for MSCT might tip the scales in favor of this test provided that the diagnostic accuracy of MSCT can be shown to be high enough for clinical application

    Is there a gender difference in noninvasive coronary imaging? Multislice computed tomography for noninvasive detection of coronary stenoses

    <p>Abstract</p> <p>Background</p> <p>Multislice computed tomography (MSCT) coronary angiography is the foremost alternative to invasive coronary angiography.</p> <p>Methods</p> <p>We sought to compare the diagnostic accuracy of MSCT in female and male patients with suspected coronary disease. Altogether 50 women and 95 men underwent MSCT with 0.5 mm detector collimation. Coronary artery stenoses of at least 50% on conventional coronary angiography were considered significant.</p> <p>Results</p> <p>The coronary vessel diameters of all four main coronary artery branches were significantly larger in men than in women. The diagnostic accuracy of MSCT in identifying patients with coronary artery disease was significantly lower for women (72%) compared with men (89%, <it>p </it>< 0.05). Also sensitivity (70% vs. 95%), positive predictive value (64% vs. 93%), and the rate of nondiagnostic examinations (14% vs. 4%, all: <it>p </it>< 0.05) were significantly worse for women. The effective radiation dose of MSCT coronary angiography was significantly higher in the examination of women (13.7 ± 1.2 mSv) than of men (11.7 ± 0.9 mSv, <it>p </it>< 0.001), mainly as a result of the fact that the radiosensitive female breast (contributing 24.5% of the dose in women) is in the x-ray path.</p> <p>Conclusion</p> <p>Noninvasive coronary angiography with MSCT might be less accurate and sensitive for women than men. Also, women are exposed to a significantly higher effective radiation dose than men.</p
