Schachbrett fĂŒr Menschen mit einer Sehbehinderung. Fertigung im Technikunterricht und Anwendung
Schach bietet jungen Menschen in der Entwicklung eine Vielfalt an kognitiven Vorteilen, die es ratsam machen, diese Sportart in Schulen anzubieten oder sich zumindest im schulischen Kontext damit auseinanderzusetzen. Im Fach Technik können durch die Fertigung eines Schachbrettes (bspw. wie hier fĂŒr Blinde, aber auch als Reiseschachbrett zum Stecken) zum einen die Kompetenzen des Schachspiels als auch die der Holzverarbeitung widergespiegelt und ĂŒberprĂŒft werden. Mit der HinfĂŒhrung zum Thema, dem ErschlieĂen eines Arbeitsplanes und allen anderen folgenden Schritten, kann man einen Aufwand von etwa einem Schulhalbjahr kalkulieren, der im Voraus schon fertig geplant werden kann. Stabilization of Tryptophan Hydroxylase 2 by L-Phenylalanine Induced Dimerization
Tryptophan hydroxylase 2 (TPH2) catalyses the initial and rateâlimiting step in the biosynthesis of serotonin, which is associated with a variety of disorders such as depression, obsessive compulsive disorder, and schizophrenia. Fullâlength TPH2 is poorly characterized due to low purification quantities caused by its inherent instability. Three truncated variants of human TPH2 (rch TPH2; regulatory and catalytic domain, NÎ47ârch TPH2; truncation of 47 residues in the N terminus of rch TPH2, and ch TPH2; catalytic domain) were expressed, purified, and examined for changes in transition temperature, inactivation rate, and oligomeric state. ch TPH2 displayed 14â and 11âfold higher halfâlives compared to rch TPH2 and NÎ47ârch TPH2, respectively. Differential scanning calorimetry experiments demonstrated that this is caused by premature unfolding of the less stable regulatory domain. By differential scanning fluorimetry, the unfolding transitions of rch TPH2 and NÎ47ârch TPH2 are found to shift from polyphasic to apparent twoâstate by the addition of lâTrp or lâPhe. Analytical gel filtration revealed that rch TPH2 and NÎ47ârch TPH2 reside in a monomerâdimer equilibrium which is significantly shifted toward dimer in the presence of lâPhe. The dimerizing effect induced by lâPhe is accompanied by a stabilizing effect, which resulted in a threefold increase in halfâlives of rch TPH2 and NÎ47ârch TPH2. Addition of lâPhe to the purification buffer significantly increases the purification yields, which will facilitate characterization of hTPH2
The landscape of current trends and procedures in plastic surgery practices across the D-A-CH region:An in-depth analysis of 511 websites
Background: Recent data shows a 139.9% global increase in aesthetic procedures since 2011, with a notable 190.5% rise in Germany. This study analyzes the current landscape of plastic surgery in German-speaking European countries, focusing on the range of procedures offered in plastic surgery practices in Germany, Austria and Switzerland. We aimed to provide an overview of the current trends and procedures of plastic surgery in the practice setting in Germany, Austria, and Switzerland. Methods: The information sampled from the websites of n = 511 plastic surgery practices (314 German, 113 Austrian, 84 Swiss) were analyzed with regards to general information, as well as procedures offered on the websites. A set of different procedures, belonging to each pillar of plastic surgery (i.e., Aesthetic, Reconstructive, Hand, Burn surgery) as well as non-surgical aesthetic procedures was analyzed. Results: Aesthetic surgical procedures were offered most often with 98.8% (506) of all websites analyzed, followed by Hand surgery at 29.7% (152), Reconstructive surgery at 27.8% (142) and Burn surgery at 5.7% (29). Reconstructive and Hand surgery were statistically significantly most commonly offered on Austrian plastic surgery websites, with p = 0.011 and p < 0.001, respectively. Among aesthetic surgical procedures, statistically significant regional differences were found for head and neck, body and breast procedures most commonly offered on Swiss plastic surgery with p = 0.041, p = 0.010 and p = 0.003, respectively, while the procedures of the intimate region were most commonly offered on German websites with p = 0.007.Conclusions: Plastic surgery websites in German-speaking European countries predominantly advertise aesthetic surgical procedures, with other domains of plastic surgery such as reconstructive, hand, and burn surgery being less prominent, suggesting an underrepresentation. Regional differences between the countries investigated are evident, underscoring the need for a more balanced approach in plastic surgery care in the practice setting.</p

Evaluating Tannins and Flavonoids from Traditionally Used Medicinal Plants with Biofilm Inhibitory Effects against MRGN E. coli
In the search for alternative treatment options for infections with multi-resistant germs,
traditionally used medicinal plants are currently being examined more intensively. In this study,
the antimicrobial and anti-biofilm activities of 14 herbal drugs were investigated. Nine of the tested
drugs were traditionally used in Europe for treatment of local infections. For comparison, another
five drugs monographed in the European Pharmacopoeia were used. Additionally, the total tannin
and flavonoid contents of all tested drugs were analyzed. HPLC fingerprints were recorded to ob-
tain further insights into the components of the extracts. The aim of the study was to identify herbal
drugs that might be useable for treatment of infectious diseases, even with multidrug resistant E.
coli, and to correlate the antimicrobial activity with the total content of tannins and flavonoids. The
agar diffusion test and anti-biofilm assay were used to evaluate the antimicrobial potential of dif-
ferent extracts from the plants. Colorimetric methods (from European Pharmacopeia) were used for
determination of total tannins and flavonoids. The direct antimicrobial activity of most of the tested
extracts was low to moderate. The anti-biofilm activity was found to be down to 10 ”g mL â1 for
some extracts. Tannin contents between 2.2% and 10.4% of dry weight and total flavonoid contents
between 0.1% and 1.6% were found. Correlation analysis indicates that the antimicrobial and the
anti-biofilm activity is significantly (p < 0.05) dependent on tannin content, but not on flavonoid
content. The data analysis revealed that tannin-rich herbal drugs inhibit pathogens in different
ways. Thus, some of the tested herbal drugs might be useable for local infections with multi-re-
sistant biofilm-forming pathogens. For some of the tested drugs, this is the first report about anti-
biofilm activity, as well as total tannin and flavonoid content
Wound fluid under occlusive dressings from diabetic patients show an increased angiogenic response and fibroblast migration
INTRODUCTION: Metabolic diseases like diabetes mellitus often show prolonged healing and chronic wounds. Occlusive wound dressings are known to support wound closure by creating a moist environment which supports collagen synthesis, epithelialization and angiogenesis. We aimed to assess the effect of occlusion on diabetic wound fluid on the cellular level regarding fibroblast activity and angiogenetic response. MATERIAL AND METHODS: 22 split skin donor sites from 22 patients (11 patients with diabetes mellitus) were treated with occlusive dressings intraoperatively. On day 3, fluid and blood serum samples were harvested while changing the dressings. The influence of wound fluid on fibroblasts was assessed by measuring metabolic activity (Alamar Blue assay, Casey Counter), cell stress/death (LDH assay) and migration (in vitro wound healing assay) of fibroblasts. Angiogenesis of endothelial cells (HUVEC) was analyzed with the tube formation assay. Furthermore, a Magnetic Luminex Assay for multi-cytokines detection was performed focusing on inflammatory and pro-angiogenetic cytokines. RESULTS: The influence of wound fluid under occlusive dressings from diabetic patients showed a significantly increased angiogenic response and fibroblast migration compared to the non-diabetic patient group. Additionally, cell stress was increased in the diabetic group. Cytokine analysis showed an increase in VEGF-A in the diabetic group. CONCLUSION: Occlusive dressings may stimulate regenerative effects in diabetic wounds. Our in-vitro study shows the influence of wound fluid under occlusive dressings from diabetic patients on angiogenesis, migration and proliferation of fibroblasts, which are essential modulators of wound healing and scar modulation
