Antitumor effect of lysine-isopeptides
Isopeptides (ε-peptides) of lysine, with a given Mw and low polydispersity (10–400 units), were synthesized to study the relationship between their chemical structure and biological effect. The designed compounds were of high purity, low polydispersity and high stereochemical purity. The effect of the compounds was tested on a human erythroleukemia cell line (K-562) and on four transplantable mouse tumors (L1210 lymphoid leukemia, P38 macrophage derived tumor, Ehrlich ascites carcinoma, Lewis lung tumor /LLT/). In case of the L1210 and P388 tumors and the Ehrlich carcinoma, survival of the animals was used as an indicator of the effect. In case of the Lewis lung tumor, the number and size of metastases in the lung and/or liver of treated and untreated mice were used as indicators. The polymers of polymerisation degree 80–120 (Mw 10.2–15.4 KD) showed the strongest antiproliferative effect both on K562 cells and the tumors growing in vivo. This effect was manifest with a significantly higher survival rate as compared to the control (L1210, P38, Ehrlich ascites), furthermore, by a decrease in the number and size of liver and lung metastases (LLT)
VĂrusfertĹ‘zĂ©s hatása a növĂ©nyek fotokĂ©miai rendszereinek kialakulására Ă©s működĂ©sĂ©re = Effect of virus infection on the development and function of plant photosystems
A fotoszintetikus rendszer vĂrusfertĹ‘zĂ©sre kialakult morfolĂłgiai Ă©s molekuláris összetĂ©telben bekövetkezett változásait kĂ©t fĹ‘ gazdanövĂ©ny-kĂłrokozĂł kapcsolatban vizsgáltuk, egyrĂ©szt a teljes sötĂ©tben nevelt, árpa csĂkos mozaik vĂrussal magátvitel formájában fertĹ‘zött árpa csĂranövĂ©nyek leveleiben, másrĂ©szt TobamovĂrusokkal fertĹ‘zött paprika növĂ©nyekben. A fertĹ‘zött árpa növĂ©nyek sejtjeiben már az etioplasztiszok kialakulása is gátlást szenvedett, amit a prolammelláris testek (PLB) Ă©s a protilakoid membránok rendellenessĂ©geivel jellemeztĂĽnk. Bár a fertĹ‘zĂ©s közvetlenĂĽl nem befolyásolta a protoklorofillid oxidoreduktáz (POR) enzim szerepĂ©t, mĂ©gis a megvilágĂtás hatására a fertĹ‘zött szövetekben a zöldĂĽlĂ©si folyamatok jelentĹ‘s gátlást, kĂ©sĂ©st szenvedtek, ami arra utal, hogy ebben a folyamatban a membrán-protein rendszer tĂ©rbeli kialakulásában is zavar támad. A POR enzim immunolokalizálása során a kontroll csĂranövĂ©nyekben a jelölĂ©s nagymĂ©rtĂ©kben a PLB-kra koncentrálĂłdott. A vĂrusfertĹ‘zĂ©s csökkentette a POR enzim jelölĂ©st. A vĂrusfertĹ‘zĂ©s csökkentette a telĂtetlen zsĂrsavak Ă©s a galaktolipidek mennyisĂ©gĂ©t, Ă©s azok arányait is. A tobamovirusok fertĹ‘zĂ©se a kĂĽlönbözĹ‘ paprika fajták illetve vonalak leveleinek kloroplasztiszaiban a tilakoid membránok szerkezetĂ©ben Ă©s összetĂ©telĂ©ben is jellemzĹ‘ változásokat okozott az enyhe serkentĂ©stĹ‘l a sĂşlyos károsĂtásig, Ă©s rámutatott a PSII fotokĂ©miai rendszer mellett a PSI károsodására is. | Effects of virus infections on the morphology and molecular structure of chloroplasts were studied in two host-parasite systems: in etiolated barley seedlings infected with Barley stripe mosaic virus (BSMV); and in pepper varieties infected by different Tobamoviruses. Seed transmission of BSMV altered the membrane structure of dark-grown barley etioplasts and caused the delay of greening processes after illumination, suggesting the early damage of chloroplast development. Immunolabeling intensity of protochlorophyllide oxidoreductase enzyme (POR) in etioplasts of infected barley leaves was weaker and the amount of the enzyme was lower than in non-infected plants. The lower amount of highly unsaturated fatty acids and the reduced abundance of galactolipids correlated with the previously detected reduction in prolamellar body to prothylakoid membrane ratio. The effect pathogenically different Tobamoviruses on the photosynthetic structures and the chlorophyll protein complexes were studied. Virus infection caused characteristic differences in the chlorophyll-protein complexes from the slight increase to the serious damage, including the alterations in PSII as well as in PSI photosystems
Hazai szőlőültetvények virológiai vizsgálata = Study of the virus infection of the Hungarian vineyards
Ăšj csoportosĂtásra kerĂĽltek a Magyarországon eddig leĂrt szĹ‘lĹ‘vĂrus kĂłrokozĂłk. 1) vĂrusos leromlása 2) levĂ©lsodrĂłdás 3) látens foltosság 4) faszöveti barázdáltság 5) szĹ‘lĹ‘ vonalas Ă©s gyűrűs mintázottsága 6) szĹ‘lĹ‘ vonalas mintázottsága 7) vĂrusszerű megbetegedĂ©sek. SzerolĂłgiai felmĂ©rĂ©sek során a 277 mintábĂłl 76 minta volt pozitĂv. A NepovĂrusok közĂĽl a korábbi elĹ‘forduláshoz kĂ©pest alacsonyabb arányban 4 elĹ‘fordulását igazoltuk GFLV, ArMV, TBRV, GCMV. A GLRaV-1, -2 -3 aránya hasonlĂł volt a NepovĂrusokĂ©hoz. JelentĹ‘s GFKV fertĹ‘zöttsĂ©get állapĂtottunk meg. 15 Komplex fertĹ‘zĂ©st találtunk. 4 GLRaV-3 Ă©s 1 GLRaV-1 izolátum HSP70 szakaszát vizsgáltuk molekuláris mĂłdszerekkel. A 3.5., 4.2 Ă©s a 2.2. jelű izolátumok a 2. számĂş GLRaV 3 csoportba sorolhatĂłk. Az 1.4 jelzĂ©sű a dendrogram adatai alapján a 4. számĂş csoportba tartozik. A GLRaV-1 izolátum az „E” csoportba tartozik. A vizsgált izolátumok gĂ©nbaki azonosĂtĂłi (HE794021, HE794022, HE794023, HE794024, HE794025). A szĹ‘lĹ‘ ĂĽltetvĂ©nyben gyűjtött Aristolochia clematitis levĂ©lminták esetĂ©ben az elvĂ©gzett elektronmikroszkĂłpos Ă©s molekuláris vizsgálatok eredmĂ©nyekĂ©ppen a TSWV fertĹ‘zĂ©st azonosĂtottunk, ami ennek a kĂłrokozĂłnak Magyarországon Ăşj gazdanövĂ©nye, Ă©s járványtani szerepe is feltĂ©telezhetĹ‘. | New classification of grapevine viruses, occurring in Hungarian vineyards, has been made according to the symptoms and recent molecular data. 1) virus degeneration, 2) leaf roll, 3) fleck, 4) rugose wood, 5) yellow mottle, 6) line pattern, 7) virus like diseases (enation, vein necrosis and vein mosaic). Among 277 samples, seventy- six gave positive results in the serological tests. Among the Nepovirus group four viruses could be detected, but the occurrence of GFLV, ArMV, TBRV, GCMV were not so frequent, than it was thought earlier. The ratio of GLRaV-1, -2 and -3 were similar, as in case of Nepoviruses. High infection rates of GFKV were detected. One GLRaV-1 and three GLRaV-3 isolates were characterised by molecular methods. The molecular data were run in a phylogenetic analyis in which the HSP70h genes of a large number of isolates from elsewhere in the world were included. The phylogenetic trees showed that the four Hungarian isolates 2.2, 3.5 and the 4.2. isolates could be inserted into the earlier classified five groups of foreign GLRaV-3 isolates into the group Nr. 2., but the isolate 1.4 belonged to the group Nr. 4. The GLRaV-1 CSE.6.4.1 isolate belonged to the earlier created group “E”. These sequence dates get into the Gene bank (HE794021, HE794022, HE794023, HE794024, HE794025). On bases of electronmicroscopic and molecular studies Aristolochia clematitis found in vineyards, which proved to be infected with TSWV

Ă–sszeállĂtásban a legfontosabb ĂĽvegházhatást kiváltĂł tĂ©nyezĹ‘k általános hatásait
vizsgáljuk a növénypatogén mikroorganizmusokra, kártevő rovarokra és a
gyomnövényekre, bemutatják a növény természetes ellenálló képességének útjait az
abiotikus stressz-hatásokkal szemben. A megemelkedett CO2 koncentráció és a globális
hĹ‘mĂ©rsĂ©klet-növekedĂ©s megváltoztatja a károsĂtĂłk viselkedĂ©sĂ©t, egyrĂ©szt, mert a
megváltozott légköri CO2 tartalom miatt megváltozik fejlődési sebességük és a kiváltott
tünetek ill. károk mértéke. Másrészt a megváltozott körülmények miatt megváltoznak
maguk a növények is, amelyek ugyancsak változtatják a kórokozók fertőzési jellemzőit,
a rovarok fejlĹ‘dĂ©sĂ©t is. Az elĹ‘bb emlĂtett kĂ©t fĹ‘ tĂ©nyezĹ‘ károsĂtĂłkra gyakorolt hatása
szerteágazĂł, egyes esetekben stimulálĂł, más esetekben gátlĂł hatásokra kell számĂtanunk.
Emiatt a hatások eredőjét nehéz tudományosan is megalapozott módon megjósolni.
Mindehhez hozzá kell tenni azt is, hogy az éghajlatváltozás a mezőgazdasági régiók
eltolódását is maga után vonhatja, és ez kiváltja a termesztett növények és azok
károsĂtĂłinak változását is. Ennek következmĂ©nye lehet az is, hogy az Ăşj terĂĽleten
megjelenő növénybetegségek és állati kártevők azokat a természetes növényi
társulásokat is megváltoztathatják, amelyek azelőtt nem voltak kitéve a
kultĂşrnövĂ©nyeket károsĂtĂł – számos esetben sokkal agresszĂvebb – fajoknak vagy
változatoknak
A hazai lágyszárĂş dĂsznövĂ©nyeket fertĹ‘zĹ‘ vĂrusok vizsgálata = Examinations on virus infection of herbaceous ornamental plants in Hungary
A kutatás során Magyarország jelentĹ‘s dĂsznövĂ©nytermesztĹ‘ körzeteibĹ‘l (HajdĂşság, DĂ©l-Alföld, Budapest Ă©s környĂ©ke, Nyugat- Ă©s DĂ©l-DunántĂşl) több mint 200 kĂĽlönbözĹ‘ dĂsznövĂ©nyekrĹ‘l származĂł minta esetĂ©ben DAS-ELISA Ă©s TAS ELISA mĂłdszerek valamint biotesztek segitsĂ©gĂ©vel megállapitottuk azok CymRSV, TMV, INSV, CVB, CSNV, AMV, PMMV, TSWV, TCSV, ToMV, PeAMV Ă©s CMV fertĹ‘zöttsĂ©gĂ©t. Vizsgálataink során Ăşj gazda-vĂrus kapcsolatokat tárunk fel Ă©s ezeket a kapcsolatokat tĂĽnettani szempontbĂłl jellemeztĂĽk. Magyarországon elĹ‘ször igazoltunk INSV fertĹ‘zöttsĂ©get a vizsgált Cyclamen, Impatiens, Ocimum Ă©s Eustoma növĂ©nyekben. Magyarország kĂĽlönbözĹ‘ terĂĽleteirĹ‘l, petĂşniárĂłl Petunia x hybrida "Bravo Rose"(KX) Ă©s a vegetatĂv Ăşton szaporĂtott "Surfinia Revolution Purple"(NYB) származĂł PVY izolátumokat vizsgáltunk szimptomatĂłlĂłgiai, szerolĂłgiai, elektronmikroszkĂłpos Ă©s molekuláris genetikai mĂłdszerekkel. A köpenyfehĂ©rje gĂ©n (CP) 380 bp szakaszát határoztuk meg Ă©s hasonlĂtottuk össze az adatbankban szereplĹ‘ adatokkal. A "Surfinia Revolution Purple" fajtárĂłl származĂł izolátum 98%-os hasonlĂłságot mutatott a PVYH: PVYNTN (m95491) izolátummal Ă©s a ?Bravo Rose? fajtárĂłl származĂł izolátum esetĂ©ben 97%-os hasonlĂłságot állapĂtottunk meg a PVYFR: PVYN (d00441) izolátummal. Vizsgálataink során elĹ‘ször igazoltuk a PVYNTN törzsĂ©nek elĹ‘fordulását magyarországi petĂşnián. | More than 200 samples were analysed from species or varieties from different ornamental plants. Samples were taken from Hungarian ornamental plant producing companies, public areas and also private gardens. Virus infection (Potato virus Y, PVY; Potato virus X, PVX; (CymRSV, TMV, INSV, CVB, CSNV, AMV, PMMV, TSWV, TCSV, ToMV, PeAMV and CMV) were screened trough DAS ELISA, TAS ELISA and biotests. During the study new virus-host relations were found, and characterised on the basis of symptoms INSV ifection was first described on ornamental plants Cyclamen, Impatiens, Ocimum and Eustoma. The nucleotide sequence of the coat protein gene of PVY isolate from the ?Surfinia Revolution Purple? (NYB) variety showed 98% homology with the homologous region of the m955491 isolate of PVYH (PVYNTN). Our results were the first data on the occurrence of NTN strain of PVY on Petunia in Hungary. P. hybrida ?Bravo Rose? (KX) isolate showed 97 % homology with a PVYFR : PVYN isolate, which was registered under the number d00441 in the database. In the case of P. hybrida species no information has been found about the occurrence of this pathogen either in international or in Hungarian literature
A burgonya Y vĂrus gumĂł nekrotikus gyűrűsfoltosság törzsĂ©vel szemben extrĂ©m rezisztenciát (immunitást) biztosĂtĂł gĂ©nek vizsgálata vad Solanum fajokban hagyományos Ă©s molekuláris mĂłdszerekkel = Study of extreme resistance genes in wild Solanum species to the tuber necrotic ringspot strain of potato virus Y by traditional and molecular methods
A kutatások munkák során vad Solanum fajok (S. acroscopicum, S. albornosii, S. arnesii, S. ambosinum, S. tarnii) genotĂpusainak PVYNTN fertĹ‘zĂ©ssel szemben mutatott reakciĂłinak virolĂłgiai Ă©s molekuláris jellemzĂ©sĂ©t vĂ©geztĂĽk el a Pannon Egyetem Georgikon Karán. CĂ©lunk az eredmĂ©nyek felhasználása burgonyanemesĂtĂ©si programokban. DAS-ELISA vĂrusfertĹ‘zĂ©si vizsgálatokkal elkĂĽlönĂtettĂĽk a vad fajok fogĂ©kony Ă©s rezisztens klĂłnjait (S. arnesii, S. acroscopicum; illetve S. tarnii esetĂ©ben). A tĂ©rkĂ©pezĂ©si populáciĂł előállĂtása cĂ©ljábĂłl vĂ©gzett keresztezĂ©seink azonban minden alkalommal Ă©s kombináciĂłban sikertelenek voltak. EzĂ©rt vizsgáltuk szomatikus Solanum hibridek (Solanum tarnii + Delikat fajta protoplasztfĂşziĂłjával) Ă©s visszakeresztezett nemzedĂ©kĂĽk tulajdonságait is, de fogĂ©kony egyedet nem sikerĂĽlt azonosĂtanunk. Ismert (5 db) Ă©s saját fejlesztĂ©sű (4 db) Ry marker jelenlĂ©tĂ©t nem sikerĂĽlt kimutatnunk a vizsgált vad fajokban. A Solanum ambosinum (PI 568916) faj fogĂ©kony Ă©s rezisztens klĂłnjait használtuk fel a gĂ©nkifejezĹ‘dĂ©s vizsgálata során, cDNS szubtrakciĂłs kĂsĂ©rletekben. SikerĂĽlt stressz-indukált gĂ©neket kimutatnunk a mintákbĂłl, amelyek PVY rezisztenciával valĂł esetleges kapcsoltságának kimutatására további vizsgálatokat folytatunk. | Conventional and molecular methods of investigations were employed in this research program at the University of Pannonia, Georgikon Faculty, Keszthely, to determine the reactions of different genotypes of wild Solanum species (S. acroscopicum, S. albornosii, S. arnesii, S. ambosinum, S. tarnii) to PVY infection. The aim of the research was to obtain useful information which could be employed in potato breeding programs. Susceptible (S. arnesii, S. acroscopicum) and resistant (S. tarnii) clones were determined by using DAS-ELISA method for showing virus infection. Intra and interspecific crosses for creating mapping populations were unsuccessful in every occasion and combination. Somatic hybrids of Solanum tarnii + Delikat obtained by protoplast fusions and their backcross generations were also investigated, but PVY susceptible plants were not identified. The presence of known and self-developed Ry markers in the Solanum species investigated were not showed. The PVY susceptible and resistant clones of the species of Solanum ambosinum (PI 568916) were used to study gene expression by employing the method of cDNA hybridization. Stress-induced genes were determined, but to verify their possible linkeages to PVY resistance requires further research to be done
General properties of grapevine viruses occurring in Hungary
The past fifty years important advances have been made in the field of grapevine virus research, including characterization of pathogens and control measurements. Still the occurrence of Grapevine fanleaf virus (GFLV), Arabis mosaic virus (ArMV), Tomato black ring virus (TBRV), Grapevine chrome mosaic virus (GCMV), Alfalfa mosaic virus (AMV), Grapevine Bulgarian latent virus (GBLV), Grapevine fleck virus (GFkV), Grapevine leafroll- associated viruses (GLRaV1-4), Grapevine virus A (GVA), Grapevine virus B (GVB) and Grapevine rupestris stem pitting- associated virus (GRSPaV) have been reported in Hungary and characterized by conventional methods as woody indexing, herbaceous indexing and serological methods. Among grapevine viruses the Grapevine line pattern virus (GLPV) seems to be uncial; because it was reported only in Hungary. Causal agents of several grapevine diseases, like enation, vein necrosis and vein mosaic remained undiscovered. These virus-like diseases occurred only sporadically, without economic importance
