    A new speech corpus of super-elderly Japanese for acoustic modeling

    The development of accessible speech recognition technology will allow the elderly to more easily access electronically stored information. However, the necessary level of recognition accuracy for elderly speech has not yet been achieved using conventional speech recognition systems, due to the unique features of the speech of elderly people. To address this problem, we have created a new speech corpus named EARS (Elderly Adults Read Speech), consisting of the recorded read speech of 123 super-elderly Japanese people (average age: 83.1), as a resource for training automated speech recognition models for the elderly. In this study, we investigated the acoustic features of super-elderly Japanese speech using our new speech corpus. In comparison to the speech of less elderly Japanese speakers, we observed a slower speech rate and extended vowel duration for both genders, a slight increase in fundamental frequency for males, and a slight decrease in fundamental frequency for females. To demonstrate the efficacy of our corpus, we also conducted speech recognition experiments using two different acoustic models (DNN-HMM and transformer-based), trained with a combination of data from our corpus and speech data from three conventional Japanese speech corpora. When using the DNN-HMM trained with EARS and speech data from existing corpora, the character error rate (CER) was reduced by 7.8% (to just over 9%), compared to a CER of 16.9% when using only the baseline training corpora. We also investigated the effect of training the models with various amounts of EARS data, using a simple data expansion method. The acoustic models were also trained for various numbers of epochs without any modifications. When using the Transformer-based end-to-end speech recognizer, the character error rate was reduced by 3.0% (to 11.4%) by using a doubled EARS corpus with the baseline data for training, compared to a CER of 13.4% when only data from the baseline training corpora were used

    Comprehensive Analysis of Bacterial Flora in Postoperative Maxillary Cyst Fluid by 16S rRNA Gene and Culture Methods

    Intracystic fluid was aseptically collected from 11 patients with postoperative maxillary cyst (POMC), and DNA was extracted from the POMC fluid. Bacterial species were identified by sequencing after cloning of approximately 580 bp of the 16S rRNA gene. Identification of pathogenic bacteria was also performed by culture methods. The phylogenetic identity was determined by sequencing 517–596 bp in each of the 1139 16S rRNA gene clones. A total of 1114 clones were classified while the remaining 25 clones were unclassified. A total of 103 bacterial species belonging to 42 genera were identified in POMC fluid samples by 16S rRNA gene analysis. Species of Prevotella (91%), Neisseria (73%), Fusobacterium (73%), Porphyromonas (73%), and Propionibacterium (73%) were found to be highly prevalent in all patients. Streptococcus mitis (64%), Fusobacterium nucleatum (55%), Propionibacterium acnes (55%), Staphylococcus capitis (55%), and Streptococcus salivarius (55%) were detected in more than 6 of the 11 patients. The results obtained by the culture method were different from those obtained by 16S rRNA gene analysis, but both approaches may be necessary for the identification of pathogens, especially of bacteria that are difficult to detect by culture methods, and the development of rational treatments for patients with POMC

    Leaf-Movement-Based Growth Prediction Model Using Optical Flow Analysis and Machine Learning in Plant Factory

    Productivity stabilization is a critical issue facing plant factories. As such, researchers have been investigating growth prediction with the overall goal of improving productivity. The projected area of a plant (PA) is usually used for growth prediction, by which the growth of a plant is estimated by observing the overall approximate movement of the plant. To overcome this problem, this study focused on the time-series movement of plant leaves, using optical flow (OF) analysis to acquire this information for a lettuce. OF analysis is an image processing method that extracts the difference between two consecutive frames caused by the movement of the subject. Experiments were carried out at a commercial large-scale plant factory. By using a microcomputer with a camera module placed above the lettuce seedlings, images of 338 seedlings were taken every 20 min over 9 days (from the 6th to the 15th day after sowing). Then, the features of the leaf movement were extracted from the image by calculating the normal-vector in the OF analysis, and these features were applied to machine learning to predict the fresh weight of the lettuce at harvest time (38 days after sowing). The growth prediction model using the features extracted from the OF analysis was found to perform well with a correlation ratio of 0.743. Furthermore, this study also considered a phenotyping system that was capable of automatically analyzing a plant image, which would allow this growth prediction model to be widely used in commercial plant factories

    A Highly Advanced Gastric Cancer Maintaining a Clinical Complete Response after Chemoradiotherapy Comprising S-1 and Cisplatin

    We report a patient with highly advanced gastric carcinoma who was treated successfully with chemoradiotherapy (CRT) comprising S-1 and cisplatin. The patient was a 71-year-old male who was diagnosed with advanced gastric carcinoma by esophagogastroduodenoscopy (EGD) by medical examination. EGD demonstrated type 3 advanced gastric carcinoma in the posterior wall of the upper gastric body. An abdominal computed tomography (CT) scan showed that the gastric wall was thickened due to gastric primary tumor, and large lymph nodes (LNs) including the lesser curvature LN, anterosuperior LN along the common hepatic artery and some para-aortic LNs were detected. The patient was diagnosed with stage IV advanced gastric carcinoma according to the Japanese classification of gastric carcinoma (cT4a, cN3, cM1 [para-aortic LN], cStage IV). Preoperative CRT was carried out in an attempt to downstage the disease. Remarkable reduction of the primary tumor and metastatic LNs was observed after initial CRT, and radiological examination determined that a partial response had been achieved. Adverse effects included grade 2 anorexia and grade 3 ALP elevation (919 U/ml). No grade 4 or more severe adverse event was observed. After CRT, although we recommended curative surgery, the patient refused surgical treatment and opted for conservative treatment. Thus, we continued S-1 oral administration for 1 year. Five months after beginning CRT, upper endoscopy showed that the tumor had maintained regression and scar formation, in which no cancer cells were detected by endoscopic biopsy. The patient is doing well and has maintained a clinical complete response for more than 42 months without curative surgery. CRT could be considered as an option for treatment of patients with locally advanced gastric carcinoma diagnosed as unresectable, or for those who refuse surgical treatment

Light absorption causes retinal chromophore isomerization and subsequent protein conformational changes visualized as optically distinguished intermediates, coupled with channel opening and closing. However, the detailed molecular events underlying channel gating remain unknown. We performed time-resolved serial femtosecond crystallographic analyses of ChR by using an X-ray free electron laser, which revealed conformational changes following photoactivation. The isomerized retinal adopts a twisted conformation and shifts toward the putative internal proton donor residues, consequently inducing an outward shift of TM3, as well as a local deformation in TM7. These early conformational changes in the pore-forming helices should be the triggers that lead to opening of the ion conducting pore

    Chemical characterization of oligosaccharides in the milk of six species of New and Old world monkeys

    Human and great ape milks contain a diverse array of milk oligosaccharides, but little is known about the milk oligosaccharides of other primates, and how they differ among taxa. Neutral and acidic oligosaccharides were isolated from the milk of three species of Old World or catarrhine monkeys (Cercopithecidae: rhesus macaque (Macaca mulatta), toque macaque (Macaca sinica) and Hamadryas baboon (Papio hamadryas)) and three of New World or platyrrhine monkeys (Cebidae: tufted capuchin (Cebus apella) and Bolivian squirrel monkey (Saimiri boliviensis); Atelidae: mantled howler (Alouatta palliata)). The milks of these species contained 6–8% total sugar, most of which was lactose: the estimated ratio of oligosaccharides to lactose in Old World monkeys (1:4 to 1:6) was greater than in New World monkeys (1:12 to 1:23). The chemical structures of the oligosaccharides were determined mainly by 1H-NMR spectroscopy. Oligosaccharides containing the type II unit (Gal(β1-4)GlcNAc) were found in the milk of the rhesus macaque, toque macaque, Hamadryas baboon and tufted capuchin, but oligosaccharides containing the type I unit (Gal(β1-3)GlcNAc), which have been found in human and many great ape milks, were absent from the milk of all species studied. Oligosaccharides containing Lewis x (Gal(β1-4)[Fuc(α1-3)]GlcNAc) and 3-fucosyl lactose (3-FL, Gal(β1-4)[Fuc(α1-3)]Glc) were found in the milk of the three cercopithecid monkey species, while 2-fucosyl lactose (5'-FL, Fuc(α1-2)Gal(β1-4)Glc) was absent from all species studied. All of these milks contained acidic oligosaccharides that had N-acetylneuraminic acid as part of their structures, but did not contain oligosaccharides that had N-glycolylneuraminic acid, in contrast to the milk or colostrum of great apes which contain both types of acidic oligosaccharides. Two GalNAc-containing oligosaccharides, lactose 3′-O-sulfate and lacto-N-novopentaose I (Gal(β1-3)[Gal(β1-4)GlcNAc(β1-6)]Gal(β1-4)Glc) were found only in the milk of rhesus macaque, hamadryas baboon and tufted capuchin, respectively. Further research is needed to determine the extent to which the milk oligosaccharide patterns observed among these taxa represent wider phylogenetic trends among primates and how much variation occurs among individuals or species
