Some considerations of occasional income
In this paper, I consider the meaning of occasional income(individual income tax)in terms of legal interpretation.
Occasional income should be levied because Japanese income tax system is set up based on comprehensive income concept. But I explain some cases that it is not appropriate to impose income tax on some kind of occasional income.
How should we consider?
My conclusion is that we should use conventional wisdom when we consider legal interpretation concerning tax law
How tax notices should be
This paper examines the state of tax notices in statutory interpretation.
While the legal system obliges administrative offi cials to follow tax notices, the notices are not a source of law and taxpayers and courts are not required to follow them.
However, tax notices play an important role in terms of tax jurisprudence for both administrative and taxpayers.
Therefore, the tax authorities should develop tax notices as necessary, and taxpayers should understand the tax notices and submit their opinions
Adenovirus-mediated transfection of caspase-8 sensitizes hepatocellular carcinoma to TRAIL- and chemotherapeutic agent-induced cell death
AbstractCaspase-8 belongs to the cysteine protease family and is known to be activated at the initial step in the cascade of TRAIL-induced apoptosis. The activation of procaspase-8 can be blocked by a relatively large amount of c-FLIP, which renders resistance to death receptor-mediated apoptosis in many types of cancer cells. To ask if extrinsic over-expression of caspase-8 contributes to the induction of apoptosis, we introduced the caspase-8 gene into HCC cells using an adenoviral (Adv) vector (Adv-Casp8). We demonstrated that Adv-Casp8 increased expression of active forms of caspase-8 in MOI-dependent manner. A large amount of Adv-Casp8 (MOI of 50) induced apoptosis significantly in HCC cells and resulted in downregulation of c-FLIP (in SK-Hep1, HLE, and HepG2 cells), XIAP, survivin, and Bcl-xL (in HLE cells) and dynamic release of cytochrome c and Smac from the mitochondria into the cytosol. On the other hand, a small amount of Adv-Casp8 (MOI of 10) causes a slight but detectable increase in the level of apoptosis with only a small effect on anti-apoptotic proteins and mitochondrial activation. However, small amounts of Adv-Casp8 augmented TRAIL- or chemotherapeutic agent-induced cell death (with an MOI of 10 or 20, respectively). These results suggest both that exogenous over-expression of caspase-8 by Adv-Casp8 may be essential for induction of HCC cell death and that the combination of Adv-Casp8 and TRAIL or chemotherapeutic agents could provide a useful strategy for treatment of HCC
Thermal Control System to Easily Cool the GAPS Balloon-borne Instrument on the Ground
This study developed a novel thermal control system to cool detectors of the
General AntiParticle Spectrometer (GAPS) before its flights. GAPS is a
balloon-borne cosmic-ray observation experiment. In its payload, GAPS contains
over 1000 silicon detectors that must be cooled below -40^{\circ}\mbox{C}.
All detectors are thermally coupled to a unique heat-pipe system (HPS) that
transfers heat from the detectors to a radiator. The radiator is designed to be
cooled below -50^{\circ}\mbox{C} during the flight by exposure to space. The
pre-flight state of the detectors is checked on the ground at 1 atm and ambient
room temperature, but the radiator cannot be similarly cooled. The authors have
developed a ground cooling system (GCS) to chill the detectors for ground
testing. The GCS consists of a cold plate, a chiller, and insulating foam. The
cold plate is designed to be attached to the radiator and cooled by a coolant
pumped by the chiller. The payload configuration, including the HPS, can be the
same as that of the flight. The GCS design was validated by thermal tests using
a scale model. The GCS design is simple and provides a practical guideline,
including a simple estimation of appropriate thermal insulation thickness,

Regulation of Gut Microbiota and Metabolic Endotoxemia with Dietary Factors
金沢大学医学系細胞分子機能学Metabolic endotoxemia is a condition in which blood lipopolysaccharide (LPS) levels are elevated, regardless of the presence of obvious infection. It has been suggested to lead to chronic inflammation-related diseases such as obesity, type 2 diabetes mellitus, non-alcoholic fatty liver disease (NAFLD), pancreatitis, amyotrophic lateral sclerosis, and Alzheimer\u27s disease. In addition, it has attracted attention as a target for the prevention and treatment of these chronic diseases. As metabolic endotoxemia was first reported in mice that were fed a high-fat diet, research regarding its relationship with diets has been actively conducted in humans and animals. In this review, we summarize the relationship between fat intake and induction of metabolic endotoxemia, focusing on gut dysbiosis and the influx, kinetics, and metabolism of LPS. We also summarize the recent findings about dietary factors that attenuate metabolic endotoxemia, focusing on the regulation of gut microbiota. We hope that in the future, control of metabolic endotoxemia using dietary factors will help maintain human health
Glucoraphanin Ameliorates Obesity and Insulin Resistance Through Adipose Tissue Browning and Reduction of Metabolic Endotoxemia in Mice
Low-grade sustained inflammation links obesity to insulin resistance and nonalcoholic fatty liver disease (NAFLD). However, therapeutic approaches to improve systemic energy balance and chronic inflammation in obesity are limited. Pharmacological activation of nuclear factor (erythroid-derived 2)–like 2 (Nrf2) alleviates obesity and insulin resistance in mice; however, Nrf2 inducers are not clinically available owing to safety concerns. Thus, we examined whether dietary glucoraphanin, a stable precursor of the Nrf2 inducer sulforaphane, ameliorates systemic energy balance, chronic inflammation, insulin resistance, and NAFLD in high-fat diet (HFD)–fed mice. Glucoraphanin supplementation attenuated weight gain, decreased hepatic steatosis, and improved glucose tolerance and insulin sensitivity in HFD-fed wild-type mice but not in HFD-fed Nrf2 knockout mice. Compared with vehicle-treated controls, glucoraphanin-treated HFD-fed mice had lower plasma lipopolysaccharide levels and decreased relative abundance of the gram-negative bacteria family Desulfovibrionaceae in their gut microbiomes. In HFD-fed mice, glucoraphanin increased energy expenditure and the protein expression of uncoupling protein 1 (Ucp1) in inguinal and epididymal adipose depots. Additionally, in this group, glucoraphanin attenuated hepatic lipogenic gene expression, lipid peroxidation, classically activated M1-like macrophage accumulation, and inflammatory signaling pathways. By promoting fat browning, limiting metabolic endotoxemia-related chronic inflammation, and modulating redox stress, glucoraphanin may mitigate obesity, insulin resistance, and NAFLD
Measurement of low-energy antiproton detection efficiency in BESS below 1 GeV
An accelerator experiment was performed using a low-energy antiproton beam to
measure antiproton detection efficiency of BESS, a balloon-borne spectrometer
with a superconducting solenoid. Measured efficiencies showed good agreement
with calculated ones derived from the BESS Monte Carlo simulation based on
GEANT/GHEISHA. With detailed verification of the BESS simulation, the relative
systematic error of detection efficiency derived from the BESS simulation has
been determined to be 5%, compared with the previous estimation of
15% which was the dominant uncertainty for measurements of cosmic-ray
antiproton flux.Comment: 13 pages, 7 figure
Role of poly (A) tail as an identity element for mRNA nuclear export
Different RNA species are rigorously discriminated and exported by distinct export factors, but this discrimination mechanism remains largely unknown. We previously showed, by RNA microinjection experiments, that intronless mRNAs are discriminated from U snRNAs based on their difference in RNA length. However, it was unclear how they are discriminated in the natural situation in which their nascent transcripts emerge progressively during transcription. We hypothesized that transcription from the corresponding promoters is important for this discrimination. Here we show that contrary to our hypothesis, the discrimination process was not significantly influenced by whether transcription occurred from an mRNA- versus a U snRNA-type promoter. Rather, the features of transcribed RNAs determined the RNA identity, consistent with our previous results of RNA microinjection. Moreover, we found that the poly (A) tail can function as an identity element for mRNA export. The presence of a poly (A) tail of an appropriate length committed otherwise short Pol II transcripts to the mRNA export pathway in a dominant manner, indicating that the poly (A) tail either contributes to increasing the RNA length or functions as a platform to recruit mRNA export factors.
