A novel mutation in isoform 3 of the plasma membrane Ca2+ pump impairs cellular Ca2+ homeostasis in a patient with cerebellar ataxia and laminin subunit 1\u3b1 mutations.
The particular importance of Ca2+ signaling to neurons demands its precise regulation within their cytoplasm. Isoform 3 of the plasma membrane Ca2+ ATPase (the PMCA3 pump), which is highly expressed in brain and cerebellum, plays an important role in the regulation of neuronal Ca2+. A genetic defect of the function of the PMCA3 pump has been described in one family with X-linked congenital cerebellar ataxia. Here we describe a novel mutation of the PMCA3 pump (ATP2B3) in a patient with global developmental delay, generalized hypotonia and cerebellar ataxia. The mutation (a R482H replacement) impairs the Ca2+ ejection function of the pump. It reduces the ability of the pump expressed in model cells to control Ca2+ transients generated by cell stimulation and impairs its Ca2+ extrusion function under conditions of low resting cytosolic Ca2+ as well. In silico analysis of the structural effect of the mutation suggests a reduced stabilization of the portion of the pump surrounding the mutated residue in the Ca2+-bound state. The patient also carries two missense mutations in LAMA1, encoding for laminin subunit 1\u3b1. On the basis of the family pedigree of the patient, the presence of both PMCA3 and LAMA1 mutations appears to be necessary for the development of the disease. Considering the observed defect in cellular Ca2+ homeostasis and the previous finding that PMCAs act as digenic modulators in Ca2+-linked pathologies, the PMCA3 dysfunction along with LAMA1 mutations could act synergistically to cause the neurological phenotype

O artigo propõe uma investigação acerca da definição de erro como privação na Ética II de Espinosa, com vistas a compreender como o conceito de privação se inscreve no discurso espinosano sobre o erro e qual sentido tal conceito assume. O problema que move a investigação é a aparente contradição ao constatarmos que Espinosa introduz o conceito privação numa dimensão em que não se faz mais necessário garantir a liberdade da vontade como seu principal requisito. Assim, examinando as diversas figuras da privação nos textos de Espinosa (Cartas a Blijenbergh e Pensamentos Metafísicos), trata-se de esclarecer o uso espinosano do termo e de mensurá-lo em relação a um aparente retorno ao cartesianismo

Damage Detection Based On Damping Analysis Of Ambient Vibration Data
Enabling an automated, remote and rapid detection of structural damage, sensor-based structural health monitoring is becoming a powerful tool for maintenance of civil engineering structures. In this study, a baseline-free, time-domain damage detection method was developed for concrete structures, which is based on analysis of nonlinear damping from measured structural vibration responses. The efficacy of the proposed method was demonstrated through a large-scale concrete bridge model subjected to different levels of seismic damage caused by shaking table tests. By applying the random decrement signature technique, the proposed method successfully identified, from its ambient vibration responses, nonlinear damping of the bridge associated with the seismic damage. The amount of the nonlinear damping increases as the seismic damage becomes more severe. This paper also compares the damage detection results with those obtained by stiffness-based methods, demonstrating a strong correlation between the increase in nonlinear damping and the decrease in structural stiffness associated with the increase in damage severity
Usefulness of mid-infrared spectroscopy as a tool to estimate body condition score change from milk samples in intensively-fed dairy cows.
Directly measuring individual cow energy balance is not trivial. Other traits, like body condition score (BCS) and BCS change (ΔBCS) can, however, be used as an indicator of cow energy status. Body condition score is a metric used world-wide to estimate cow body reserves and the estimation of ΔBCS was, until now, conditional on the availability of multiple BCS assessments. The aim of the present study was to estimate ΔBCS from milk mid-infrared (MIR) spectra and days in milk (DIM) in intensively-fed dairy cows using statistical prediction methods. Daily BCS was interpolated from cubic splines fitted through the BCS records and daily ΔBCS was calculated from these splines. Body condition score change records were merged with milk MIR spectra recorded on the same week. The data set comprised 37,077 ΔBCS phenotypes across 9,403 lactations from 6,988 cows in 151 herds based in Quebec (Canada). Partial least squares regression (PLSR) and a neural network (NN) were then used to estimate ΔBCS from 1) MIR spectra only, 2) DIM only, or 3) MIR spectra and DIM together. ΔBCS data in both the first 120 DIM and 305 DIM of lactation were used to develop the estimates. Daily ΔBCS had a standard deviation of 4.40*10-3 BCS units in the 120-d data set and of 3.63*10-3 BCS units in the 305-d data set. 4-fold cross-validation was used to calibrate and test the prediction equations. External validation was also conducted using more recent years of data. Irrespective of whether based on the first 120 or 305 DIM, or when MIR spectra only, DIM only or MIR spectra and DIM were jointly used as prediction variables, NN produced the lowest root mean square error (RMSE) of cross-validation (1.81*10-3 BCS units and 1.51*10-3 BCS units, respectively, using the 120-d and 305-d data set). Relative to predictions for the entire 305 DIM, the RMSE of cross-validation was 15.4% and 1.5% lower in the first 120 DIM when using PLSR and NN, respectively. Predictions from DIM only were more accurate than those using just MIR spectra data but, irrespective of the data set and of the prediction model used, the combining DIM information with MIR spectral data as prediction variables reduced the RMSE compared with inclusion of DIM alone, albeit the benefit was small (the RMSE from cross-validation was reduced up to 5.5% when DIM and spectral data were jointly used as model features instead of DIM only). However, when predicting extreme ΔBCS records, the MIR spectral data was more informative than DIM. Model performance when predicting ΔBCS records in future years was similar to that from cross-validation demonstrating the ability of MIR spectra of milk and DIM combined to estimate ΔBCS, particularly in early lactation. This can be used to routinely generate estimates of ΔBCS to aid in day-to-day individual cow management
Selecting Milk Spectra to Develop Equations to Predict Milk Technological Traits
peer-reviewedIncluding all available data when developing equations to relate midinfrared spectra to a phenotype may be suboptimal for poorly represented spectra. Here, an alternative local changepoint approach was developed to predict six milk technological traits from midinfrared spectra. Neighbours were objectively identified for each predictand as those most similar to the predictand using the Mahalanobis distances between the spectral principal components, and subsequently used in partial least square regression (PLSR) analyses. The performance of the local changepoint approach was compared to that of PLSR using all spectra (global PLSR) and another LOCAL approach, whereby a fixed number of neighbours was used in the prediction according to the correlation between the predictand and the available spectra. Global PLSR had the lowest RMSEV for five traits. The local changepoint approach had the lowest RMSEV for one trait; however, it outperformed the LOCAL approach for four traits. When the 5% of the spectra with the greatest Mahalanobis distance from the centre of the global principal component space were analysed, the local changepoint approach outperformed the global PLSR and the LOCAL approach in two and five traits, respectively. The objective selection of neighbours improved the prediction performance compared to utilising a fixed number of neighbours; however, it generally did not outperform the global PLSR

Interview: Roberto Bolzani FilhoRoberto Bolzani Filho é bacharel e licenciado em Filosofia pela Universidade de São Paulo (USP), onde defendeu a Dissertação de Mestrado O ceticismo pirrônico na obra de Sexto Empírico (1992) e a Tese de Doutorado O ceticismo acadêmico e a ideia de subjetividade (2003), ambos sob a orientação de Oswaldo Porchat Pereira. Desde 1988, leciona História da Filosofia Antiga na mesma instituição, na qual também obteve o título de Livre-Docente, com a tese Ensaios sobre Platão (2013). Dentre a publicação de inúmeros artigos em revistas especializadas, bem como do livro Acadêmicos versus pirrônicos (São Paulo: Alameda, 2013), destaca-se a sua participação na redação de Filosofia: temas e percursos (1ª ed. 2013; 2ª ed. 2016), o qual integrou a escolha do Programa Nacional do Livro Didático (PNLD, 2018) para a área de filosofia, tornando-se uma importante referência para a seleção de livros didáticos a serem adotados nas escolas e ao trabalho desenvolvido pelos professores, em seu cotidiano.
