    Raw poultry meatballs with soya flour: Shelf life and nutritional value

    Poultry meat is a valuable source of protein for human consumption. It plays an important role in countries with poor ungulate meat production, including the Republic of Kazakhstan. The intake of fibre by the Kazakh population also remains low, while the intake of saturated fatty acids is excessive. Therefore, it is recommended to combine meat with plant products, e.g. soya flour. In the present research, we developed and evaluated a new meatball product containing different amounts of soya flour. The meatballs proved to be a semi-finished high-protein product. They also demonstrated a good fatty acid and mineral profile. The product with 30% of soya flour showed the best results: 27% of protein, low content of saturated fatty acid, and shelf life of 48 h. To extend the shelf life of the meatballs under refrigerator conditions, new disinfection methods should be developed.S

    A new and simple method was developed for the extraction and derivatization of fatty acid (FA) in milk. Lipid extraction of milk was carried out in H2SO4/methanol. Methylation was performed for 2 h at 60°C, and FA methyl esters were recovered for chromatographic analysis by the addition of hexane. The method parameters were optimized and the simple method was compared to the official reference procedure for the extraction and methylation of FAs in milk samples. For most of the 24 FAs determined, similar or significantly higher recoveries were obtained by the simple method than by the conventional method. The simple method allows processing a high number of samples, at the same time, minimizing the sample manipulation and, consequently, the sample loss and contamination. In conclusion, the proposed method is simple, rapid, low cost, and achieves good resultsUn nuevo método abreviado se desarrolló para la extracción y derivatización de ácidos grasos en leche. La extracción de los lípidos de la leche se llevó a cabo en H2SO4/metanol. La metilación se realizó a 60°C durante 2 h y los ésteres metílicos de los ácidos grasos fueron recuperados para el análisis cromatográfico mediante la adición de hexano. Los parámetros del método fueron optimizados y el método abreviado fue comparado con el método de referencia para la determinación de ácidos grasos en leche. En la mayor parte de los 24 ácidos grasos analizados, el método abreviado mostró recuperaciones similares o mejores que el método convencional.     An Evaluation of Alternatives to Nitrites and Sulfites to Inhibit the Growth of Salmonella enterica and Listeria monocytogenes in Meat Products

    In recent years, the use of nitrites and sulfites as food preservatives has been a cause for concern due to the health problems that these additives can cause in humans. Natural products have been studied as an alternative, but most of them have hardly been applied in the food industry for technological and economic reasons. In this sense, organic salts such as sodium acetate are a good alternative due to their affordability. Thus, this study evaluated the capacity of sodium nitrite, sodium sulfite, a sodium acetate product (TQI C-6000), and chitosan to inhibit two important foodborne pathogens, Salmonella enterica and Listeria monocytogenes. The MIC of each chemical was in vitro evaluated and their antibacterial action was subsequently checked in situ using minced meat as a food model. MIC values of sodium nitrite (10,000 mg/L) and sodium sulfite (50,000 mg/L) for Salmonella enterica were higher than the values allowed by legislation (450 mg/L for sulfites and 150 mg/L for nitrites). Additionally, the sodium acetate product caused the inhibition of Salmonella enterica and Listeria at a relative low quantity. The two foodborne pathogens were inhibited in the food model with 1% of the sodium acetate product. Additionally, there were no significant differences between sodium nitrite, sodium sulfite, and sodium acetate products in the inhibition of Salmonella enterica and Listeria monocytogenes in the food model.     Short chain fatty acids commonly produced by gut microbiota influence salmonella enterica motility, biofilm formation, and gene expression

    Short chain fatty acids (SCFAs) are commonly produced by healthy gut microbiota and they have a protective role against enteric pathogens. SCFAs also have direct antimicrobial activity against bacterial pathogens by di usion across the bacterial membrane and reduction of intracellular pH. Due to this antimicrobial activity, SCFAs have promising applications in human health and food safety. In this study, the minimum inhibitory concentrations (MICs) of four SCFAs (acetic acid, butyric acid, propionic acid, and valeric acid) in Salmonella strains isolated from poultry were determined. The e ect of subinhibitory concentrations of SCFAs in Salmonella biofilm formation, motility, and gene expression was also evaluated. Butyric acid, propionic acid, and valeric acid showed a MIC of 3750 g/mL in all strains tested, while the MIC of acetic acid was between 1875 and 3750 g/mL. Subinhibitory concentrations of SCFAs significantly (p < 0.05) reduced the motility of all Salmonella strains, especially in the presence of acetic acid. Biofilm formation was also significantly (p < 0.05) lower in the presence of SCFAs in some of the Salmonella strains. Salmonella strain. Salmonella Typhimurium T7 showed significant (p < 0.05) upregulation of important virulence genes, such as invA and hilA, especially in the presence of butyric acid. Therefore, SCFAs are promising substances for the inhibition of the growth of foodborne pathogens. However, it is important to avoid the use of subinhibitory concentrations that could increase the virulence of foodborne pathogen Salmonella.S

    Tracing (r)bST in cattle: Liquid-based options for extraction and separation

    Growth hormone (GH) or somatotropin (ST) is a species-specific polypeptide hormone produced in the pituitary gland of vertebrates. When administered exogenously to dairy cattle, it has galactopoietic effects and is capable of increasing the milk yield. Recombinant bST enabled large-scale applications in farms, enhancing significantly milk production. While it is banned in the European Union (EU), several countries permit the trade and use of recombinant somatotropins in animal husbandry. In this context, effective analytical methods are needed for residue control to avoid an illegal use of rbST but also to prevent fraudulent labeling in some cases. The present review includes studies published in the last 5 years (from 2012 to 2017) to monitor rbST in bovine animals, using liquid-based applications.     Detección cuantitativa de microorganismos resistentes a la tetraciclina en carne convencional y ecológica de ternera, cerdo y pollo

    The use of antimicrobials has increased the number of resistant bacteria to these drugs; however, the organic production has restricted the use of these compounds. The objectives of this work were to assess counts of tetracycline-resistant bacteria using conventional microbiology, to compare these results with those obtained for tet(A) and tet(B) genes by qPCR and to investigate both genes in conventional and organic meat. Counts of mesophilic aerobic bacteria were higher in organic beef, while chicken meat obtained higher counts for Enterobacteriaceae. Only tet(B) was higher in conventional pork and chicken meat than in their organic counterparts. The tet(A) gene was found in almost 100% of samples and tet(B) gene changed according to the type of meat. The presence of tet genes suggests that they are widely distributed, especially tet(A), in food of animal origin, even in organic meat samples obtained from animals in which the use of antimicrobials is restrictedEl uso de los antimicrobianos ha incrementado sustancialmente el número de bacterias resistentes a estos fármacos sin embargo, la producción ecológica, ha limitado el uso de estos medicamentos. Los objetivos del trabajo fueron evaluar los recuentos obtenidos de bacterias resistentes a tetraciclina mediante microbiología convencional, obtener recuentos de bacterias con los genes tet(A) y tet(B)mediante qPCR e investigar la distribución de ambos genes en carne convencional y ecológica. Los recuentos de bacterias aerobias mesófilas fueron significativamente mayores en carne ecológica de ternera, mientras que los recuentos de Enterobacteriaceae fueron superiores en carne convencional de pollo. Sólo el gen tet(B) fue significativamente mayor en carne convencional de cerdo y de pollo que en sus homólogas ecológicas. El gen tet(A) se encontró en casi todas las muestras mientras que el tet(B) varió según la especie.     Tracing recombinant bovine somatotropin ab(use) through transcriptomics: the potential of bovine somatic cells in a multi-dose longitudinal study

    In the European Union, the use of recombinant bovine somatotropin (rbST) in dairy cattle is forbidden. Monitoring rbST (ab)use by its direct detection in animal matrices still remains a challenging task. New monitoring methods based on indirect detection of the substance are necessary. A new transcriptomic system based on the use of high-throughput real-time PCR in combination with somatic cells was developed to control rbST administration in dairy animals. A total of nine cows, separated into control and rbST-treated groups, were included in the study. A subcutaneous injection containing 500 mg of rbST was administered to the treated group every 14 days, up to a total of 12 doses. Milk somatic cells (MSCs) were sampled from each animal at different time points throughout 8 months of study. It was possible to obtain the transcriptomic profile of 18 genes in MSCs of rbST-treated and control groups, and using univariate and multivariate statistical analysis control and treated animals were discriminated. The transcription of CCND1, IGF-1R, TNF and IL-1β genes resulted strongly influenced by rbST treatment.     Determination of the Presence of Three Antimicrobials in Surface Water Collected from Urban and Rural Areas

    Due to the continuous release of antimicrobials into the environment, the aim of this study was to compare the frequency of detection of sulfamethazine, sulfamethoxypyridazine and trimethoprim in surface water collected from urban and rural areas in Northwestern Spain. A monitoring study was conducted with 314 river water samples analyzed by high-performance liquid chromatography coupled to tandem mass spectrometry. The results indicated that 37% of the samples contained residues of at least one of the investigated antimicrobials, and every sampling site yielded positive samples.     Prevalence, molecular characterization and antimicrobial resistance of Salmonella serovars isolated from northwestern Spanish broiler flocks (2011–2015)

    The present study investigated the prevalence, antimicrobial resistance to twenty antibiotics, and class 1 integron and virulence genes of Salmonella isolated from poultry houses of broilers in northwestern Spain between 2011 and 2015. Strains were classified to the serotype level using the Kauffman-White typing scheme and subtyping with enterobacterial repetitive intergenic consensus PCR. The prevalence of Salmonella spp. was 1.02%. Sixteen different serotypes were found, with S. typhimurium and S. arizonae 48:z4, z23:- being the most prevalent. A total of 59.70% of strains were resistant to at least one, and 19.70% were resistant to multiple drugs. All Salmonella spp. were susceptible to cefotaxime, ciprofloxacin, gentamicin, kanamycin, levofloxacin, neomycin, and trimethoprim. The highest level of resistance was to sulfamethoxazole (40.29%), doxycycline (17.91%), and nalidixic acid (17.91%). None of the isolates carried class 1 integron and only isolates of S. enterica subspecies enterica were positive for all virulence factors tested, whereas S. arizonae lacked genes related to replication and invasion in nonphagocytic cells. This study demonstrates that the prevalence and antimicrobial resistance of Salmonella spp. in poultry houses of broilers of northwestern Spain is low compared with those found in other studies and in other steps of the food chainS

    Evolution of Resistance in Poultry Intestinal Escherichia coli During Three Commonly Used Antimicrobial Therapeutic Treatments in Poultry

    The resistance rates of intestinal Escherichia coli populations from poultry were determined during treatment and withdrawal period with 3 antimicrobial agents commonly used as therapeutics in poultry medicine. A total of 108 chickens were considered: 18 were treated orally with enrofloxacin, 18 with doxycycline, and 18 with sulfonamides, whereas another 18 chickens were maintained as controls for each antimicrobial group. Fecal samples were taken during the treatment and after the withdrawal period, and E. coli were isolated through Fluorocult media plating. A total of 648 E. coli strains (216 per antimicrobial tested) were isolated and identified though biochemical methods. Minimal inhibitory concentrations to the antimicrobials used were also determined using a broth microdilution method. The resistance rates of intestinal E. coli to all of the antimicrobials tested significantly increased during the course of the therapeutic treatment. In addition, significant differences (P = 0.0136) in resistance rates persisted between the intestinal E. coli of the enrofloxacin-treated and control batches until the end of the withdrawal period, but this difference was not observed for the cases of doxycycline or sulfonamides treatments. Antimicrobial use in poultry medicine seems to select for antimicrobial-resistant strains of pathogenic bacterial species such as E. coli.
