Produção de celulases através da fermentação em estado sólido utilizando como substrato sorgo lignocelulosico
The solid state cultivation of the filamentous fungus isolated from the nest of leaf-cutting ants of
the species Acromyrmex balzani was carried out, using lignocellulosic sorghum as a substrate
aiming at the maximum production of cellulases, through the optimization of the cultivation
process, evaluating the influence of the variables: initial concentration inoculum, humidity, time,
pH and temperature. The effects of the variables were first evaluated in terms of the activity of
the FPase enzyme, where humidity and time were statistically significant at 95%. In conditions of
greater time and humidity, the enzymatic activity decreases showing a negative effect. The other
variables did not show a statistically significant effect at 95% confidence. The greatest enzymatic
activities were provided by the combination of the variables of test 2, which presents humidity
60%, pH 4, incubation time of 4 days, initial inoculum concentration of 108 and temperature of
40oC. The significant variables were subsequently optimized. In the second part of the
experiment, the activity of the FPase enzyme showed the best response with the moisture
content of 60 and 75%, and the incubation time 4 to 6 days, with a maximum productivity of 4.1
IU / g. The CMCase activity showed maximum activity between 55 and 65% humidity and with
an incubation time of 5 to 8 days, reaching an enzymatic activity of 4.1 IU / g. As for Avicelase
activity, the highest production was with the moisture content around 65 and 80% and with the
incubation time of 2 to 5 days, reaching an enzymatic activity of 3.8 U / g. The activities
presented demonstrate an expressive cellulolytic capacity of the studied fungus, compared to
lignocellulosic sorghum.Realizou-se o cultivo em estado sólido do fungo filamentoso isolado do ninho de formigas
cortadeiras da espécie Acromyrmex balzani, utilizando como substrato sorgo lignocelulosico
visando à máxima produção de celulases, através da otimização do processo de cultivo,
avaliando a influência das variáveis: concentração inicial de inóculo, umidade, tempo, pH e
temperatura. Os efeitos das variáveis foram primeiramente avaliados em termos da atividade da
enzima FPase, onde a umidade e o tempo mostraram-se estatisticamente significativos a 95%.
Em condições de maior tempo e umidade a atividade enzimática diminuí mostrando um efeito
negativo. As demais variáveis não apresentaram efeito estatisticamente significativo a 95% de
confiança. As maiores atividades enzimáticas foram proporcionadas pela combinação das
variáveis do ensaio 2 que apresenta umidade 60%, pH 4, tempo de incubação de 4 dias,
concentração inicial de inoculo de 108 e temperatura de 40oC. As variáveis significativas foram
subsequentemente otimizadas. Na segunda parte do experimento a atividade da enzima FPase
apresentou melhor resposta com o teor de umidade de 60 e 75%, e o tempo de incubação 4 a 6
dias, com produtividade máxima de 4,1 UI/g. A atividade CMCase apresentou máxima atividade
entre 55 e 65% de umidade e com o tempo de incubação de 5 a 8 dias, alcançando uma
atividade enzimática de 4,1 UI/g. Já para atividade Avicelase a maior produção foi com o teor de
umidade em torno de 65 e 80% e com o tempo de incubação de 2 a 5 dias, alcançando uma
atividade enzimática de 3,8 U/g. As atividades apresentadas demonstram expressiva
capacidade celulolítica do fungo estudado, frente ao sorgo lignocelulósico

The purpose of this study was to define the factors that influence the production of cellulases by Penicillium oxalicum, a strain obtained from a leaf-cutting ant colony and identified based on the ITS gene. The experimental design included solid state fermentation using sugarcane bagasse and lignocellulosic sorghum as the lignocellulosic substrate. The variables were analyzed using a 25-1 fractional factorial design, with three replicates on the central point. All the variables analyzed influenced the production of at least one of the three cellulose types analyzed. The highest values observed were: FPase 4.2 U g-1, CMCase 9.2 U g-1 and Avicelase 8.4 U g-1 using lignocellulosic sorghum as the substrate. The best conditions for enzyme production were: incubation temperature at 40ºC, initial moisture of 60%, pH of 4.0 and a growth period of four days using lignocellulosic sorghum as the substrate.
