Crowdsourcing eXtension: Communities of Practice Provide Rapid Response
This article provides an example of how you can use eXtension\u27s Communities of Practice for crowdsourcing information rapidly and thoroughly. It contends that unlike traditional Google or Yahoo searches, asking colleagues within eXtension provides a depth of discovery with multiple layers of vetting already built in. In addition to that, it\u27s free. The example herein stems from a recent inquiry to the Community, Local and Regional Food Systems (CLRFS) Community of Practice and provides excerpts from the responses

Depressive symptoms are associated with visceral adiposity in a community-based sample of middle-aged women and men
To examine the relation between measures of adiposity and depressive symptoms in a large well characterized community-based sample, we examined the relations of visceral adipose tissue (VAT) and subcutaneous adipose tissue (SAT) to depressive symptoms in 1581 women (mean age 52.2 years) and 1718 men (mean age 49.8 years) in the Framingham Heart Study. Depressive symptoms were measured using the Center for Epidemiologic Studies Depression (CES-D) scale. Regression models were created to examine the association between each fat depot (exposure) and depressive symptoms (outcome). Sex specific models were adjusted for age, body mass index, smoking, alcohol consumption, diabetes, hypertension, total and HDL cholesterol, lipid lowering treatment, CVD, menopause, C-reactive protein, and physical activity. Mean CES-D scores were 6.8 and 5.6 in women and men. High levels of depressive symptoms were present in 22.5% of women and 12.3% of men. In women, one standard deviation increase in VAT was associated with a 1.3 point higher CES-D score after adjusting for age and BMI (p<0.01) and remained significant in the fully adjusted model (p=0.03). The odds ratio of depressive symptoms per 1 standard deviation increase in VAT in women was 1.33 (p=0.015); results were attenuated in fully adjusted models (OR 1.29, p=0.055). In men, the association between VAT and CES-D score and depressive symptoms was not significant. SAT was not associated with CES-D score or depressive symptoms. This study supports an association between VAT and depressive symptoms in women. Further work is needed to uncover the complex biologic mechanisms mediating the association
Spectroscopic Discovery of the Broad-Lined Type Ic Supernova 2010bh Associated with the Low-Redshift GRB 100316D
We present the spectroscopic discovery of a broad-lined Type Ic supernova (SN
2010bh) associated with the nearby long-duration gamma-ray burst (GRB) 100316D.
At z = 0.0593, this is the third-nearest GRB-SN. Nightly optical spectra
obtained with the Magellan telescopes during the first week after explosion
reveal the gradual emergence of very broad spectral features superposed on a
blue continuum. The supernova features are typical of broad-lined SNe Ic and
are generally consistent with previous supernovae associated with low-redshift
GRBs. However, the inferred velocities of SN 2010bh at 21 days after explosion
are a factor of ~2 times larger than those of the prototypical SN 1998bw at
similar epochs, with v ~ 26,000 km/s, indicating a larger explosion energy or a
different ejecta structure. A near-infrared spectrum taken 13.8 days after
explosion shows no strong evidence for He I at 1.083 microns, implying that the
progenitor was largely stripped of its helium envelope. The host galaxy is of
low luminosity (M_R ~ -18.5 mag) and low metallicity (Z < 0.4 Z_solar), similar

Cell-free translation and purification of Arabidopsis thaliana regulator of G signaling 1 protein
Arabidopsis thaliana Regulator of G protein Signalling 1 (AtRGS1) is a protein with a predicted N-terminal 7-transmembrane (7TM) domain and a C-terminal cytosolic RGS1 box domain. The RGS1 box domain exerts GTPase activation (GAP) activity on GĪ± (AtGPA1), a component of heterotrimeric G protein signaling in plants. AtRGS1 may perceive an exogenous agonist to regulate the steady-state levels of the active form of AtGPA1. It is uncertain if the full length RGS1 protein exerts any atypical effects on GĪ±, nor has it been established exactly how AtRGS1 contributes to perception of an extracellular signal and transmits this response to a G-protein dependent signaling cascade. Such an understanding can be deduced from in vitro studies using the purified and soluble RGS1-box domain. Further studies on full-length AtRGS1 have been inhibited due to the extreme low abundance of the endogenous AtRGS1 protein in plants and lack of a suitable heterologous system to express AtRGS1. Here, we describe methods to produce full-length AtRGS1 by cell free synthesis into unilamellar liposomes and nanodiscs. The cell-free synthesized AtRGS1 exhibits GTPase activating activity on GĪ± and can be purified to a level suitable for biochemical analyses
Forced Abstinence from Cocaine Self-Administration is Associated with DNA Methylation Changes in Myelin Genes in the Corpus Callosum: a Preliminary Study
Background: Human cocaine abuse is associated with alterations in white matter integrity revealed upon brain imaging, an observation that is recapitulated in an animal model of continuous cocaine exposure. The mechanism through which cocaine may affect white matter is unknown and the present study tested the hypothesis that cocaine self-administration results in changes in DNA methylation that could result in altered expression of several myelin genes that could contribute to the effects of cocaine on white matter integrity. Methods: In the present study, we examined the impact of forced abstinence from cocaine self-administration on chromatin associated changes in white matter. To this end, rats were trained to self-administer cocaine (0.75āmg/kg/0.1āmL infusion) for 14ādays followed by forced abstinence for 1āday (nā=ā6) or 30ādays (nā=ā6) before sacrifice. Drug-free, sham surgery controls (nā=ā7) were paired with the experimental groups. Global DNA methylation and DNA methylation at specific CpG sites in the promoter regions ofmyelin basic protein (Mbp), proteolipid protein-1 (Plp1), and SRY-related HMG-box-10 (Sox10) genes were analyzed in DNA extracted from corpus callosum. Results: Significant differences in the overall methylation patterns of the Sox10 promoter region were observed in the corpus callosum of rats at 30ādays of forced abstinence from cocaine self-administration relative to sham controls; the ā189, ā142, ā93, and ā62 CpG sites were significantly hypomethylated point-wise at this time point. After correction for multiple comparisons, no differences in global methylation or the methylation patterns of Mbp or Plp1 were found. Conclusion: Forced abstinence from cocaine self-administration was associated with differences in DNA methylation at specific CpG sites in the promoter region of the Sox10 gene in corpus callosum. These changes may be related to reductions in normal age related changes in DNA methylation and could be a factor in white matter alterations seen after withdrawal from repeated cocaine self-administration. Further research is warranted examining the effects of cocaine on DNA methylation in white matter
A comprehensive study of GRB 070125, a most energetic gamma ray burst
We present a comprehensive multiwavelength analysis of the bright, long
duration gamma-ray burst GRB 070125, comprised of observations in -ray,
X-ray, optical, millimeter and centimeter wavebands. Simultaneous fits to the
optical and X-ray light curves favor a break on day 3.78, which we interpret as
the jet break from a collimated outflow. Independent fits to optical and X-ray
bands give similar results in the optical bands but shift the jet break to
around day 10 in the X-ray light curve. We show that for the physical
parameters derived for GRB 070125, inverse Compton scattering effects are
important throughout the afterglow evolution. While inverse Compton scattering
does not affect radio and optical bands, it may be a promising candidate to
delay the jet break in the X-ray band. Radio light curves show rapid flux
variations, which are interpreted as due to interstellar scintillation, and are
used to derive an upper limit of cm on the radius of the
fireball in the lateral expansion phase of the jet. Radio light curves and
spectra suggest a high synchrotron self absorption frequency indicative of the
afterglow shock wave moving in a dense medium. Our broadband modeling favors a
constant density profile for the circumburst medium over a wind-like profile
(). However, keeping in mind the uncertainty of the parameters, it is
difficult to unambiguously distinguish between the two density profiles. Our
broadband fits suggest that \event is a burst with high radiative efficiency

Properties of a Gamma-Ray Burst Host Galaxy at z ~ 5
We describe the properties of the host galaxy of the gamma-ray burst GRB 060510B based on a spectrum of the burst afterglow obtained with the Gemini North 8 m telescope. The galaxy lies at a redshift of z = 4.941, making it the fourth highest spectroscopically identified burst host. However, it is the second highest redshift galaxy for which the quality of the spectrum permits a detailed metallicity analysis. The neutral hydrogen column density has a logarithmic value of 21.0-21.2 cm-2, and the weak metal lines of Ni, S, and Fe show that the metallicity is in excess of a tenth of solar, which is far above the metallicities in damped Lya absorbers at high redshift. The tightest constraint is from the Fe lines, which place [Fe/H] in excess of -0.8. We argue that the results suggest that metallicity bias could be a serious obstacle to inferring star formation from the GRB population, and we consider how future higher quality measurements could be used to resolve this issue
