    The contribution of muscle hypertrophy to strength changes following resistance training

    Purpose Whilst skeletal muscle hypertrophy is considered an important adaptation to resistance training (RT), it has not previously been found to explain the inter-individual changes in strength after RT. This study investigated the contribution of hypertrophy to individual gains in isometric, isoinertial and explosive strength after 12 weeks of elbow flexor RT. Methods Thirty-three previously untrained, healthy men (18–30 years) completed an initial 3-week period of elbow flexor RT (to facilitate neurological responses) followed by 6-week no training, and then 12-week elbow flexor RT. Unilateral elbow flexor muscle strength [isometric maximum voluntary force (iMVF), single repetition maximum (1-RM) and explosive force], muscle volume (V m), muscle fascicle pennation angle (Ξ p) and normalized agonist, antagonist and stabilizer sEMG were assessed pre and post 12-week RT. Results Percentage gains in V m correlated with percentage changes in iMVF (r = 0.527; P = 0.002) and 1-RM (r = 0.482; P = 0.005) but not in explosive force (r ≀ 0.243; P ≄ 0.175). Percentage changes in iMVF, 1-RM, and explosive force did not correlate with percentage changes in agonist, antagonist or stabilizer sEMG (all P > 0.05). Percentage gains in Ξ p inversely correlated with percentage changes in normalized explosive force at 150 ms after force onset (r = 0.362; P = 0.038). Conclusions We have shown for the first time that muscle hypertrophy explains a significant proportion of the inter-individual variability in isometric and isoinertial strength gains following 12-week elbow flexor RT in healthy young men

    Whey protein does not enhance the adaptations to elbow flexor resistance training

    Purpose: It is unclear whether protein supplementation augments the gains in muscle strength and size observed following resistance training (RT), as limitations to previous studies include small cohorts, imprecise measures of muscle size and strength, and no control of prior exercise or habitual protein intake (HPI). We aimed to determine whether whey protein supplementation affected RT-induced changes in elbow flexor muscle strength and size. Methods: We pair-matched 33 previously untrained, healthy young men for their HPI and strength response to 3-wk RT without nutritional supplementation (followed by 6-wk no training), and then randomly assigned them to protein (PRO; n = 17) or placebo (PLA; n = 16) groups. Participants subsequently performed elbow flexor RT 3 d/wk for 12-wk and consumed PRO or PLA immediately before and after each training session. We assessed elbow flexor muscle strength [unilateral 1-RM and isometric maximum voluntary force (MVF)] and size [total volume and maximum anatomical cross-sectional area (ACSAmax) determined with MRI] before and after the 12-wk RT. Results: PRO and PLA demonstrated similar increases in muscle volume (PRO, 17.0 ± 7.1% vs. PLA, 14.9 ± 4.6%; P = 0.32), ACSAmax (PRO, 16.2 ± 7.1% vs. PLA, 15.6 ± 4.4%; P = 0.80), 1-RM (PRO, 41.8 ± 21.2% vs. PLA, 41.4 ± 19.9%; P = 0.97) and MVF (PRO, 12.0 ± 9.9% vs. PLA, 14.5 ± 8.3%; P = 0.43). Conclusion: In the context of this study, protein supplementation did not augment elbow flexor muscle strength and size changes that occurred after 12-wk RT.     Monocyte Subset Recruitment Marker Profile Is Inversely Associated With Blood ApoA1 Levels.

    Dyslipidemia promotes development of the atherosclerotic plaques that characterise cardiovascular disease. Plaque progression requires the influx of monocytes into the vessel wall, but whether dyslipidemia is associated with an increased potential of monocytes to extravasate is largely unknown. Here (using flow cytometry) we examined recruitment marker expression on monocytes from generally healthy individuals who differed in lipid profile. Comparisons were made between monocyte subsets, participants and relative to participants' lipid levels. Monocyte subsets differed significantly in their expression of recruitment markers, with highest expression being on either the classical or non-classical subsets. However, these inter-subset differences were largely overshadowed by considerable inter-participant differences with some participants having higher levels of recruitment markers on all three monocyte subsets. Furthermore, when the expression of one recruitment marker was high, so too was that of most of the other markers, with substantial correlations evident between the markers. The inter-participant differences were explained by lipid levels. Most notably, there was a significant inverse correlation for most markers with ApoA1 levels. Our results indicate that dyslipidemia, in particular low levels of ApoA1, is associated with an increased potential of all monocyte subsets to extravasate, and to do so using a wider repertoire of recruitment markers than currently appreciated

    The removal of thermally aged films of triacylglycerides by surfactant solutions

    Thermal ageing of triacylglycerides (TAG) at high temperatures produces films which resist removal using aqueous surfactant solutions. We used a mass loss method to investigate the removal of thermally aged TAG films from hard surfaces using aqueous solutions of surfactants of different charge types. It was found that cationic surfactants are most effective at high pH, whereas anionics are most effective at low pH and a non-ionic surfactant is most effective at intermediate pH. We showed that the TAG film removal process occurs in several stages. In the first ‘‘lag phase’’ no TAG removal occurs; the surfactant first partitions into the thermally aged film. In the second stage, the TAG film containing surfactant was removed by solubilisation into micelles in the aqueous solution. The effects of pH and surfactant charge on the TAG removal process correlate with the effects of these variables on the extent of surfactant partitioning to the TAG film and on the maximum extent of TAG solubilisation within the micelles. Additionally, we showed how the TAG removal is enhanced by the addition of amphiphilic additives such as alcohols which act as co-surfactants. The study demonstrates that aqueous surfactant solutions provide a viable and more benign alternative to current methods for the removal of thermally aged TAG films

    Using a knowledge exchange event to assess study participants' attitudes to research in a rapidly evolving research context [version 1; peer review: 3 approved]

    BACKGROUND: The UK hosts some of the world’s longest-running longitudinal cohort studies, who make repeated observations of their participants and use these data to explore health outcomes. An alternative method for data collection is record linkage; the linking together of electronic health and administrative records. Applied nationally, this could provide unrivalled opportunities to follow a large number of people in perpetuity. However, public attitudes to the use of data in research are currently unclear. Here we report on an event where we collected attitudes towards recent opportunities and controversies within health data science. METHODS: The event was attended by ~250 individuals (cohort members and their guests), who had been invited through the offices of their participating cohort studies. There were a series of presentations describing key research results and the audience participated in 15 multiple-choice questions using interactive voting pads. RESULTS: Our participants showed a high level of trust in researchers (87% scoring them 4/5 or 5/5) and doctors (81%); but less trust in commercial companies (35%). They supported the idea of researchers using information from both neonatal blood spots (Guthrie spots) (97% yes) and from electronic health records (95% yes). Our respondents were willing to wear devices like a ’Fit-bit’ (78% agreed) or take a brain scan that might predict later mental illness (73%). However, they were less willing to take a new drug for research purposes (45%). They were keen to encourage others to take part in research; whether that be offering the opportunity to pregnant mothers (97% agreed) or extending invitations to their own children and grandchildren (98%). CONCLUSIONS: Our participants were broadly supportive of research access to data, albeit less supportive when commercial interests were involved. Public engagement events that facilitate two-way interactions can influence and support future research and public engagement efforts

    LO-Phonon Emission Rate of Hot Electrons from an On-Demand Single-Electron Source in a GaAs/AlGaAs Heterostructure.

    Using a recent time-of-flight measurement technique with 1 ps time resolution and electron-energy spectroscopy, we develop a method to measure the longitudinal-optical-phonon emission rate of hot electrons traveling along a depleted edge of a quantum Hall bar. Comparison to a single-particle model implies the scattering mechanism involves a two-step process via an intra-Landau-level transition. We show that this can be suppressed by control of the edge potential profile, and a scattering length >1  mm can be achieved, allowing the use of this system for scalable single-electron device applications
